Ahmed","doi":"10.21608/aunj.2022.139022.1013","DOIUrl":"https://doi.org/10.21608/aunj.2022.139022.1013","url":null,"abstract":"DNA-protein cross-links and thymidine dimer are special types of DNA damages that formed when DNA exposed to many inducers as UV irradiations. Anthocyanins are members of phytochemicals found in many plants including Hibiscus sabdariffa. It was reported that anthocyanins protect DNA from UV-induced damages. Here we investigate the role of autophagy induced by anthocyanins in protection of UVB-induced DNA-protein cross-links and thymidine dimer in HepG2 cells . Cells were treated either with anthocyanins, Rapamycin, or combination of anthocyanins and Bafilomycin A1 or anthocyanins and 3-methyladenine for 24 h and then subjected to 0.36 J/cm 2 UVB irradiation. The results revealed that anthocyanins induce autophagy formation as detected by acridine orange pH dependent green to red (G/R) color shifting. To investigate the role of anthocyanins-induced autophagy on DNA-protein cross-links and thymidine dimer formation after UVB exposure, electrophoresis of protein covalently linked to DNA or thymidine dimer dot plot analysis of DNA were carried out. It was found that anthocyanins treated cells have reduced DNA-protein cross-links and thymidine dimer compared with the anthocyanins untreated cells. methyladenine together with anthocyanins significantly increased the DNA-protein cross-links and thymidine dimer formation compared with anthocyanins treated cells. The major reason for human arsenic toxicity is tainting of drinking water. Arsenic trioxide is inorganic arsenic that is classified by the US Environmental Protection Agency as a known human carcinogen. Aim: Illustration of morphological and histological effects of arsenic trioxide on mice offspring. Methods: Adult female albino mice were divided into five groups: Control, negative control treated with Hcl (arsenic trioxide solvent), and three groups daily treated orally with different doses of arsenic trioxide (0.3, 0.7 and 1 mg/kg). Offspring of 21 day old mice were morphologically examined, weighed and morphometric measurements were carried out. Liver and kidney were histopathologically examined. Furthermore, the treated larvae were allowed to recover from the particle-effect for additional 3 and 6 days to address whether the particle impact is sustainable or temporary. To this end, a sub lethal concentration (LC30) of ZnONPs was used to address their impact on the tissues of the alimentary canal and Malpighian tubules of Culex larvae. The treated larvae suffered negative signs on foregut, midgut , hindgut and Malpighian tubule epithelia which appeared in the form of columnar cell vaculation and damage of the microvilli. In the meantime, the larvae which allowed recovering for 3 and 6 days didn’t show signs of improvement in the alimentary canal and Malpighian tubules suggesting sustainable impact of the particles on the larval tissues. Future work will be directed toward ultra-structural studies in the gut cells.
