Flanzy","doi":"10.1051/RND:19790214","DOIUrl":"https://doi.org/10.1051/RND:19790214","url":null,"abstract":"In the rat, prolonged intake of a large quantity of lauric acid (C12:0) before and during pregnancy had no effect on the amount of lipids present in adipose stores at the time of parturition. However, this medium chain fatty acid was intensely esterified in the triglycerides of the maternal adipose tissues. As compared to a diet rich in long chain fatty acids, the intake of lauric acid during lactation considerably modified mammary gland fatty acid secretion. Lauric acid was esterified in very high proportions (about 35 p. 100 of the total fatty acids) in milk triglycerides, thus considerably increasing its saturated fatty acid content (about 80 p. 100 of the total fatty acids). In these nutritional conditions, the mammary gland could not efficiently regulate the qualitative aspects of fatty acid secretion. However, these large modifications of the milk triglyceride fraction did not change the weight of the pups at weaning and had no effect on their body composition. This would indicate that the growth of the lipid compartment was unchanged. The weight of the liver, its total lipid content and the distributions between neutral lipids and phospholipids were identical to those of the controls. A large amount of the lauric acid ingested by the mother was transferred to the pup in the milk before weaning. A large quantity was found in the body triglycerides (30 p.100 of the total fatty acids) and in neutral liver lipids. They were killed when adult during the sexual season in autumn and compared with 4 normal adult rams of the same age, slaughtered at the same time. In the cryptorchid testes, there was a drastic decrease in the total number of Leydig cells per testis, in the length and diameter of seminiferous tubules, and in the numbers of stem spermatogonia. After 56 days of treatment the animals were killed and pituitary and plasma levels of luteinizing hormone (LH), follicle stimulating hormone (FSH) and prolactin (PRL) were measured by radioimmunoassay. Ovariectomy caused marked rises in both the content and concentration of LH and FSH within the pituitary gland while pituitary PRL levels fell. Plasma levels of LH and FSH also rose while PRL titers remained unchanged. The daily afternoon injection of melatonin (25 ¡Lg subcutaneously) further increased the accumulation of LH and FSH within the pituitary. Juillard","doi":"10.1051/RND:19790310","DOIUrl":"https://doi.org/10.1051/RND:19790310","url":null,"abstract":"Electron dense-cored vesicles (DCV), originating in the Golgi apparatus and migrating into the perivascular processes, constitute a characteristic feature of the pinealocytes in the pineal gland of mice and hamsters. This report presents the results of ultracytochemical studies carried out to clarify the nature and physiological significance of these vesicles. Using proteases and the PA-TCH-silver technique on ultrathin sections, it was concluded that the dense core of DCV was proteinaceous in nature. These data, correlated with previous pharmacological and cytophysiological studies, showed the important role of DCV in the storage and intracellular migration of a proteinaceous compound of unknown significance. However, in agreement with authors who hypothesized the elaboration of active peptidergic compounds in the mammalian pineal gland to explain some effects that could not be ascribed to indoleamines, it was proposed that DCV might store a carrier-neurohormone complex.
