Study design: A calvarial bone defect was created in 20 male Wistar rats and filled with bioactive glass alone (n=10) or combined with simvastatin (n=10). After 4 weeks, the defects were histomorphometrically evaluated for volume fraction (Vv) of woven bone, vessel density, bioglass quantity, and inflammation. Results: Compared to the bioglass-only group, rats treated with simvastatin had greater Vv of blood vessels (3.3%+0.7 vs 1.6%+0.1, P=0.0002) and new bone (2.3%+0.2 vs 1.8%+2.5, P=0.003). The Vv of the bioglass remnants in the bioglass-only group was higher than in the group treated with simvastatin (2.4% +0.08 vs 1.7%+0.3, P<0.0004). Chronic inflammation was noted in one rat from each group. This newly developed implant achieves osseous integration by utilizing a hydroxyapatite (HA) coating plus a specialized grafting technique that produces much improved success rates relative to its fibro-integrated subperiosteal predecessor. This case reported herewith represents a maxillary CEI implant that was placed and in functional service for nine years before later being retrieved and processed for histologic examination subsequent to the patient's demise. In addition, due to infection that occurred shortly after placement, an early provisional procedure with fluoridated HA was also performed. Histologic analysis of the post-mortem specimen revealed fully integrated new bone formation intimately surrounding the previously dehisced implant strut. The latter had previously been decontaminated and grafted with a thin layer of fluorapatite (FA) material. Here we have described the mechanism how PDGF can be used in surgeries where bone grafting is required. The importance of describing the molecular mechanism is that it will provide future application for faster bone regeneration and also the inhibitory mechanism in condition for bone regeneration as in osteosarcoma. The important specific activities of PDGF include mitogenesis (increase in the cell populations of healing cells), angiogenesis (endothelial mitoses into functioning capillaries) and macrophage activation (debridement of the wound site and a second phase source of growth factors for continued repair and bone regeneration). The aim of this study was to evaluate whether sinus membrane elevation and the simultaneous insertion of titanium implants without additional grafting material constitute a valid technique for bone augmentation of the maxillary sinus floor.Material and methods: A 35-year-old female patient evaluated with lost dentition in the left posterior maxilla. Preoperative cone beam computerized tomography (CBCT) were taken to guide the surgery. The sinus lift was performed and two implants were placed simultaneously in the residual subantral bone. The collected peripheral venous blood was applied to support the sinus membrane over the implant apex. Computed tomography (CT) was performed after 6 months healing period.Results: Comparisons of pre- and postoperative CT radiography clearly demonstrated new bone formation and new sinus floors were found within the compartment created by the sinus membrane elevation procedure. All implants remained stable during the healing period in clinical evaluations therefore definitive fixed prosthesis inserted. The described method allows the accurate transfer of the implant position and the registration of the interocclusal relationship using transfer key and electroformed gold copings. Only articles on human subjects were considered. At least 12 month of mean follow-up was required for inclusion. No restriction was placed regarding study design.Results: Ten studies were included in this review. Survival rates ranged from 92% to 100%. A total of 497 implants were placed in sites with endodontic infection. Therefore, the aim of this study was to assess the correlation between insertion torque (IT) and resonance frequency analysis (RFA) of implants placed in the mandible and maxilla of different bone densities. Eighty dental implants were placed in the maxilla and mandible and IT and the implant stability quotient (ISQ) were measured at the time of implant insertion. Bone density was assessed subjectively by the Lekholm and Zarb index (1985). The Type I and II densities were grouped together (Group A) and Type III and IV densities in another group (Group B). The IT in Group A was higher (student-t Test, p=0.0013) than IT in Group B (46.27 + 18.51 N/cm, 33.62 + 14.74 N/cm, respectively). The implants placed in Group A showed higher ISQ (student-t Test, p=0.0004) than those placed in Group B (70.09 + 7.50, 63.66 + 8.00, respectively). A significant correlation between IT and the ISQ value was observed for Group A (Pearson correlation test; r= 0.35; p= 0.0213) and for Group B (r= 0.37; p= 0.0224). Endotoxin or bacterial challenges to inflammatory cells are directly relevant to dental implants pathologies involving bone resorption, such as osteointegration failure and periimplantitis. While the endotoxin amount on implant devices is regulated by standards, it is not known whether commercially available dental implants elicit different levels of adherent-endotoxin stimulated cytokines. The objective of this work is to develop a model system and to evaluate endotoxin-induced expression of pro-inflammatory cytokines genes relevant to osteoclasts activation on commercially available dental implants. Murine J774-A1 macrophages were cultured on Ti disks with different level of Lipopolysaccharide (LPS) contamination, to define the time-course of the inflammatory response to endotoxin, as evaluated by RT-PCR analysis. The developed protocol was then used to measure adherent endotoxin on commercially available dental implants, packaged, sterile, that is in the \"as-implanted\" condition. Results show that tested dental implants induce variable expression of endotoxin-stimulated genes, sometime over the level expected to promote bone resorption in vivo. Results are not affected by the specific surface treatment, rather they likely reflect cares in cleaning and packaging protocols. In conclusion, expression of genes that enhance osteoclasts activity through endotoxins stimulation of inflammatory cells is widely different on commercially available dental implants.
