From the viewpoint of forensic dental medicine, it is a theme no less valuable. This experimental study was attempted for this reason. Teeth from an individual body of human, monkey, dog, rabbit and rat were heated in turn on the muffle furnace. The heating temperatures were from 200 degrees C to 1,000 degrees C in time spans of 5, 30 and 60 minutes. After heating, each tooth and its control were observed by a scanning electron microscope (magnifying power: 3,500 x or 3,600 x). At heating temperature of 500 degrees C or 600 degrees C, enamel starts to come off in lumps and cracks appear in the enamel rods. The arcaded form in human and monkey, hexagon in dog, elongated chain in rabbit, rows of short, diagonal parallel lines equally directed at every other row in rat--these basic morphological features of the enamel rods--are retained till the heat reaches 700 degrees C. The enamel rods in monkey yield to heat more easily than those in human. At 600 degrees C many cracks appear and deformation of the arcaded form starts. With heating of 5 minutes at 800 degrees C the outline of the pattern is obscured. Human and animal teeth get varied forms of cracks in the enamel rods with heating more than 5 minutes at 800 degrees C. The structure of the enamel rods is broken and morphological characteristics are lost. {"title":"Evidence of transformed androgen receptor in rat submandibular gland by three-step column chromatography.","authors":"H Ejiri","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The transformation of cytosolic androgen receptors was studied in the male rat submandibular gland (SMG) at 3 and 7 weeks of age, using 3H-R1881 as the labeled ligand. Sephacryl S-200 column chromatography revealed two R1881 binding proteins having a mol wt of 150-200 kDa and 14-20 kDa in cytosol. The affinity column of DE52 cellulose gave the main peak of cytosolic receptor at the KCl concentration of 1.5-2.5 M, demonstrating that the association of the 150-200 kDa with 14-20 kDa forms would depend on the ionic environment. The cytosolic 150-200 kDa protein, which had a high specificity to androgen, became lower in molecular weight to 14-20 kDa following DE52 cellulose column chromatography. The nuclear KCl extracted receptor, after dialyzing by Sephadex G-75 column, showed two peaks at 150-200 kDa and 14-20 kDa by Sephacryl S-200 column. The nuclear 150-200 kDa receptor was also associated with the 14-20 kDa protein by DE52-cellulose column at 1.5-2.5 M KCl. The binding eluate at 1.5-2.5 M KCl on DE52 was estimated to be a 14-20 kDa protein and was not dissociated into two forms by Sephacryl S-200. The androgen specificity to the nuclear 14-20 kDa binding protein was emphasized by high ionic strength. The sialoadenectomy markedly reduced the enzyme activities at 5 and 7 weeks of age, when measured 2 and 4 weeks after the operation. The weights of the ovary and uterus increased by the ectomy. However, such increases in the uterus and ovary weights were not shown in the rats sialoadenectomized at 7 weeks of age when measured 4 weeks after the operation, revealing rather a decrease in the weight. The formation of estrogens from delta 4-androstenedione was decreased by the ectomy at 11 weeks of age. When 3H-testosterone and ovary microsome were incubated, the aromatase activities rather increased in the sialoadenectomized rats. We, thereby, attempted to estimate the age by examining tooth samples from two unidentified bodies. In both cases, we were able to estimate almost accurately the age from the total amino acids and also from the insoluble collagen as well as the soluble peptides (+/- 0 to +4 years). Age estimation from the average of the D/L ratios of 3 fractions was as accurate as +/- 3 years. Therefore, a differential examination of amino acid fractions seemed to increase information dramatically for age estimation by racemization of amino acids. {"title":"Isolation and characterization of 15K noncollagenous protein from bovine periodontal ligament.","authors":"Y Sasaki","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>In pursuit of the nature of noncollagenous proteins (NCP) and their biosynthesis in periodontal ligament (PDL), isolation, characterization and a short incubation study were made on the PDL of the bovine mandibular incisors. A sodium chloride extract from the PDL contained several noncollagenous protein components. Eliminating serum derived proteins by a cross-reaction with antibody against bovine serum proteins, a protein estimated to be 15K on 5-15% gradient NaDodSO4-gel was the major constituent in the extract. This protein was isolated by sequential chromatography using Sepharose CL-6B and Sephacryl S-200 under dissociative conditions (4M guanidium hydrochloride). A chemical analysis of the protein showed that it was rich in aspartic acid, glutamic acid, glycine, alanine, leucine and lysine, and contained 5% sugars. Antibody directed against the purified PDL 15K protein reacted with the 15K protein, but no cross-reaction was observed with other components from the bovine PDL. The cell-mediated immunity, assessed by enumeration of total rosette forming cells [TRFC] and high affinity rosette forming cells [HARFC], was found to be depressed in LJP patients compared to controls. Estimation of serum immunoglobulins G,A,M,D and E levels were done using single radial immunodiffusion. All the immunoglobulins except IgD were found to be elevated significantly in LJP patients. The defective immune response found in LJP patients may be the cause or effect of the disease process. The procedure consisted of extraction with a 10 mM Tris/Mg buffer containing 0.1% Nonidet P40, chromatography on a hyaluronate-conjugated Affi-Gel 15, and fast protein liquid chromatography. The hyaluronate-binding alkaline phosphatase has an apparent molecular weight of 110,000, contained sialic acid and was more stable to heat treatment than were two other species (Mr = 120,000 and 130,000) extracted from PDL. The raised and constant concentration of 3 alpha-diol was revealed in female SMG.
