{"title":"[Comparison of 7 anti-hepatitis C virus enzyme immunoassays of the 2nd generation in blood donation services].","authors":"C Gabriel, B Blauhut","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Seven enzyme-linked immunosorbent assays for the detection of antibodies to hepatitis C virus were compared: three assays with synthetic peptides in the coating showed overproportional higher rates of initial reactive and also the highest rate of so-called false-positive results in comparison with the RIBA assay. One assay with synthetic peptides had also the most false-negative results in comparison with RIBA-reactive results. This approach is especially useful for human monoclonal antibodies due to instability of human antibody-producing cell lines. {"title":"[Critical limits of hemodilution: theoretical principles].","authors":"R Zander","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>For daily clinical practice an isovolemic hemodilution down to an arterial O2 content of 10 ml/dl, corresponding to a hemoglobin content of 7.5 g/dl or a hematocrit value of 22.5%, is described as a tolerable value, as long as normovolemia and normoxia (no disturbances of lung function) are guaranteed and local restrictions in perfusion (coronary or cerebral sclerosis) are excluded. This value is not derived from the mixed venous O2 status but from the situation of the myocardium as the main limiting organ for anemic hypoxemia. Compensation of anemia is regulated hemodynamically: First, by an increase in stroke volume; secondary, by an increase in heart frequency and, tertiary, by an increase in venous utilization. {"title":"[Recombinant plasma proteins for therapeutic use--status and developmental trends].","authors":"H Storch","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The enormous progress made in biotechnology and purification of plasma proteins (pp) and the demands to avoid risks of transmitting HIV, hepatitis and other virus infections by these have resulted in the development of numerous recombinant human (rh) pp, which are now about to be used as replacement therapy in transfusion medicine. Human rh albumin has been used in clinical trials last year, a competition to serum albumin can be expected in the next time. During the last decade, the genes or cDNA have been cloned and characterized for all relevant pp involved in blood coagulation. Beside the rh factor VIII (rh FVIII) which has been introduced clinically in 1991, the rh FVIIa is under investigation in patients with hemophilia A and inhibitors. After establishing of rhFIX in triple transgenic mice, the industrial potential will be evaluated in terms of scale up culturing and production.
