In 1986, ESACP was founded in Munich in the context of the Concerted Action for Automated Cytology (CAAC) of the European Community with the goal to promote the advancement of clinically oriented cytometry as it relates to medically oriented cell and tissue imaging in Europe. The founding members decided to launch a new journal addressing all aspects of analytical and quantitative cytology and pathology. As a founding member, I still remember the heated discussions about an attractive name for the journal of the new society. By February 1989 these ideas had materialised with G. Brugal, as editor-in-chief and Elsevier as publisher until 1996 when IOS Press took over. The new journal, Analytical Cellular Pathology, became with respect to impact factor one of the fastest growing journals in the histoand cytopathology field starting at 1.14 in 1993, followed by 1.82 in 1996 and 0.97 in 1999 when I was elected editor-in-chief. In 2002 ACP’s impact factor was 1.08.","PeriodicalId":76996,"journal":{"name":"Analytical cellular pathology : the journal of the European Society for Analytical Cellular Pathology","volume":"76 1","pages":"207 - 208"},"PeriodicalIF":0.0,"publicationDate":"2003-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"83866921","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Our study did not reveal any DNA copy number changes or allelic loss on 3p. The results suggest that DNA copy number changes are not associated with the histological grade or clinical behaviour of PT and the chromosomal changes on 3p appear to be rare. The most distinct feature of these methods is that separate correlation (covariance) matrices are produced for every level in the hierarchy. In karyometric research, the cell-level (i.e., within-tumor) correlations seem to be of major interest. Beside their biological importance, these correlation coefficients (CC) are compulsory when dimensionality reduction is required. Using MLwiN, a dedicated program for multilevel modeling, we show how to use multivariate multilevel models (MMM) to obtain and interpret CC in each of the levels. A comparison with two usual, \"single-level\" statistics shows that MMM represent the only way to obtain correct cell-level correlation coefficients. The summary statistics method (take average values across each patient) produces patient-level CC only, and the \"pooling\" method (merge all cells together and ignore patients as units of analysis) yields incorrect CC at all. We conclude that multilevel modeling is an indispensable tool for studying correlations between morphometric variables.</p>","PeriodicalId":76996,"journal":{"name":"Analytical cellular pathology : the journal of the European Society for Analytical Cellular Pathology","volume":"25 4","pages":"187-91"},"PeriodicalIF":0.0,"publicationDate":"2003-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.1155/2003/562508","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"40894310","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The optimal area is defined as the well spread part of the smear. This zone starts when the erythrocytes stop overlapping (on the body film side) and finishes when the erythrocytes start losing their clear central zone (on the feather edge side). The approach yields a quick detection of this area in images scanned under low magnifying power (immersion objective x 25 or x 16). The algorithm consists of two stages. First, an image analysis procedure using mathematical morphology is applied for extracting the erythrocytes, the centers of erythrocytes and the erythrocytes with center. Second, the number of connected components from the three kinds of particles is counted and the coefficient of spreading rho(s) and the coefficient of overlapping rho(o) are calculated. The data from fourteen smears illustrate how the technique is used and its performance. Numerous studies have indicated that these assessments may provide clinically relevant information related to the degree of progression and malignant potential of breast neoplasia. Nuclear features are derived from computerized analysis of digitized microscope images, and a quantitative Feulgen stain for DNA was used. Features analyzed included: (1) DNA content; (2) nuclear size and shape; and (3) texture features, describing spatial features of chromatin distribution. In this study replicated measurements are described on a series of 54 breast carcinoma specimens of differing pathologic grades. Duplicate measurements were performed using two serial sections, which were processed and analyzed separately. The value of a single feature measurement, the nuclear area profile, was shown to be the strongest indicator of progression. A quantitative nuclear grade was derived and shown to be strongly correlated with not only the pathologic nuclear grade, but also with tubule formation, mitotic grade, and with the overall histopathologic grade. Analysis of replication precision showed that the standard methods of the histopathology laboratory, if practiced in a uniform manner, are sufficient to ensure reproducibility of these assessments. We argue that nuclear morphometry provides a standardized and reproducible framework for quantitative pathologic assessments.</p>","PeriodicalId":76996,"journal":{"name":"Analytical cellular pathology : the journal of the European Society for Analytical Cellular Pathology","volume":"25 3","pages":"129-38"},"PeriodicalIF":0.0,"publicationDate":"2003-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.1155/2003/262918","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"22409636","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Non-invasive early diagnosis of oral cavity malignancies. 口腔恶性肿瘤的无创早期诊断。

{"title":"Non-invasive early diagnosis of oral cavity malignancies.","authors":"Jon Sudbø","doi":"10.1155/2003/527484","DOIUrl":"https://doi.org/10.1155/2003/527484","url":null,"abstract":"Oral cancer is a multistep, multipathway and multifocal process extending over a period of 10–20 years, where most manifest carcinomas are assumed to be preceeded by readily detectable visible changes of the oral mucosa [3,4]. It is not known to which extent carcinomas may arise from clinically normal oral mucosa, but reliable molecular markers indicate that cancer may arise from lesions that by traditional histological assessment are assumed to be without any malignant potential [6,7]. However, oral carcinogenesis most likely represents a field process, with multiple clones developing independently, in a metachronous manner [1,2,5]. Conceivably, aggressive cells clones that have not yet given rise to visible lesions could subsequently give rise to subsequent carcinomas. Currently, there are no reliable non-invasive methods for screening for oral cancer in patients with a high-risk behaviour for oral cancer, but with clinically normal appearing oral mucosa. However, risk of oral cancer can be assessed by measuring DNA content in cells of the mucosal lining in patients with visible lesions. In this issue of the journal, Remmerbach and colleagues present a case study of 4 patients, from which they obtained cytobrushings to look for genetic changes relevant for development of oral squamous cell carcinomas. Notably, the initially performed biopsies did not indicate malignant transition. Brush biopsies were obtained and smears of these were investigated cytologically after Papanicolaou staining. After cytological assessment, cells were restained with Feulgen and measurement of DNA content was performed by image cytometry. Although the results will need to be reproduced by other independent groups, this limited case study demonstrates the proof of principle that significant genetic changes in epithelial linings can be detected by non-invasive methods. The incidence rate of oral cancer is increasing. Today, a substantial portion of new cases is diagnosed at an advanced stage of disease, which contributes to the poor prognosis of this group of patients. Early detection of subclinical lesions makes possible an early intervention, e.g., by chemoprevention in this patients group [8]. Demonstrating that person at high risk may be identified by non-invasive methods is an important contribution that opens for opportunistic screening of high-risk persons. Molecular risk markers to identify persons at high risk of cancer (>50 percent cancer risk within a time span of 3–4 years) can only be feasible in a screening setting if the procedures related to the work-up are not associated with undue strain on persons at risk. In this respect, the study by Remmerbach marks an interesting advance.","PeriodicalId":76996,"journal":{"name":"Analytical cellular pathology : the journal of the European Society for Analytical Cellular Pathology","volume":"25 4","pages":"157-8"},"PeriodicalIF":0.0,"publicationDate":"2003-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.1155/2003/527484","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"40816001","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In this study monoclonal plasma cells/mm in light chain immunoglobulin immunostained archival bone marrow sections from 22 patients in whom a diagnosis of multiple myeloma (MM) had been excluded but who had monoclonal proteins were counted by two observers at light microscopic level. There was good correlation between the counts of the two observers. The levels of monoclonal plasma cells/mm in biopsies were not related to the % counts in the aspirates taken at the same time as the biopsies. Three of seven patients with biopsy levels in excess of the polyclonal levels in patients without plasma cell dyscrasia developed progressive MM within the observation time. A new combination of image analysis algorithms for segmentation of cells imaged by fluorescence microscopy is presented. The algorithm consists of an image pre-processing step, a general segmentation and merging step followed by a segmentation quality measurement. The quality measurement consists of a statistical analysis of a number of shape descriptive features. Objects that have features that differ to that of correctly segmented single cells can be further processed by a splitting step. By statistical analysis we therefore get a feedback system for separation of clustered cells. After the segmentation is completed, the quality of the final segmentation is evaluated. By training the algorithm on a representative set of training images, the algorithm is made fully automatic for subsequent images created under similar conditions. Automatic cytoplasm segmentation was tested on CHO-cells stained with calcein. The fully automatic method showed between 89% and 97% correct segmentation as compared to manual segmentation.</p>","PeriodicalId":76996,"journal":{"name":"Analytical cellular pathology : the journal of the European Society for Analytical Cellular Pathology","volume":"24 2-3","pages":"101-11"},"PeriodicalIF":0.0,"publicationDate":"2002-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.1155/2002/821782","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"22124890","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
