{"title":"Production of potent antivenin against cobra venom with conjugated-cobrotoxin.","authors":"H C Huang,&nbsp;M Y Liau,&nbsp;Y C Lee","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Cobra venom (Naja naja atra) and its fractions obtained by ammonium sulfate precipitation were subjected to chromatography on CM-Cellulose colum. A highly purified cobrotoxin obtained by the repeated chromatography on preparative CM-Cellulose column was 6.7 times more toxic than the original cobra venom. The toxin was detoxified by a bifunctional reagent, glutaraldehyde, to about 99.8% and utilized for immunization in animals. Mice received 4 weekly immunization with detoxified cobrotoxin and challenged one week after the last injection showed 60% protection in rabbits by immunization with detoxified cobrotoxin reached 360 LD50 neutralizing level against the cobra venom within 30 days. Therefore, the toxin could be purified with trypsin digest and then absorbed on kaolin and Kieselgel, followed by eluting the different pH buffer solutions. We failed to find true mates in this trial, even when the cells cured by acridine orange were used to mate with non-cured ones. There were some positive growth when two densed cell suspensions were dropped on the minimum medium, but after subcultured on the minimum medium no growth was found. It seem likely that the bacteria used here, at least, require an enforced cell adhesion, unless true mating is not feasible. The impairment was found to be contributed by certain unknown factors in their plasma and subnormal reactivity of their T lymphocytes. The plasma derived from active lepromatous leprosy patients depressed the reaction of normal cells and normal plasma enhanced the reaction of active lepromatous lymphocytes. The cellular factor was studied by using a one-way reaction in which one of the two lymphocyte preparations was inactivated with mitomycin C. Ten percent were found to be HBsAg positive. All of the 289 cord blood samples were HBsAg negative. Babies born to the HBsAg positive mothers were followed 2-8 months (average 3.5 months) after delivery and 37.5% were positive for HBsAg.
