{"title":"[Enzymatic determination of glandular kallikrein in human pancreatic secretions].","authors":"H Weber, W Dummler, S Liebe","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>A method for the enzymatic determination of tissue kallikrein (EC 3.4. 21.8) in human pancreatic juice using D-Val-cyclohexyl-Ala-Arg-4-nitroaniline as substrate as well as the reaction conditions optimized for 37 degrees C are described. The reaction is characterized by the following kinetic parameters, determined according to Hanes and Eisenthal, Cornish-Bowden: KM = 2.10(-5) mol/l; Vmax = 0.800 mumol/(l.s). The coefficient of intra-assay variation (N = 10) was determined to 2.54%. Salt intake is high (151 mmol/d), but lower then expected. There are no important differences between people without (140 mmol/d) and with (161 mmol/d) a essential hypertension. Salt intake of male is higher than female persons. {"title":"[Prostacyclin and thromboxane synthesis in liver tissue in chronic liver diseases].","authors":"B Zipprich, C Gerhardt, C Giessler, C Taube, F W Rath, H J Mest, R Nilius","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>This paper reports on investigations of the formation of PGI2 and TXA2 using their stabile products 6-keto-PGF1 alpha and TXB2 (RIA) in liver biopsy specimens of 46 patients suffering from fatty liver (n = 19), chronic hepatitis B (n = 11), liver cirrhosis (n = 13), and miscellaneous diseases (n = 3). The measured formation rates in chronic liver disease were evaluated in comparison to a reference group (n = 19) consisting of minimal liver lesions. The 6-keto-PGF1 alpha formation correlating to the degree of the portal inflammation in the liver (morphometric evaluation). The same trend existed in relation to the intralobular inflammation. {"title":"[The determination of apolipoproteins Apo A-I and Apo B. A comparative study of methods].","authors":"H Keller, W F Riesen","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>We compared the analytical performance of a new in house developed ELISA and the commercial TurbiTime system for the determination of Apo AI and Apo B. Six dilutions of a poolserum, whose concentration had been determined by two other independent methods, were used as test material. A serum from which the apolipoproteins had been removed by immunadsorption was used as a diluent. Six specimens with known true values of Apo AI and Apo B were thus prepared. We established the analytical precision, the regression function and the correlation with respect to the true values and we constructed precision profiles and bias profiles. We found that the precision of turbidimetry is superior for the determination of Apo AI with concentrations less than 1 g/l. With Apo B both systems are nearly equivalent, the results of the turbidimetric methods were a little too low, the results of the ELISA a little too high. {"title":"[Diagnostic importance of HDL cholesterol determination].","authors":"J Reissner, W Herrmann","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The present paper describes the sensitivity to quantification of changes of HDL-cholesterol in serum by two different precipitation and analytical techniques during the treatment of patients. After the precipitation of VLDL and LDL by phosphotungstic acid/magnesium chloride the chemical determination of HDL-cholesterol in serum with the Liebermann-Burchard reaction yields different results in comparison to enzymatic HDL-cholesterol determined in serum supernatant after the precipitation by polyethylene glycol 20.000.
