In metastatic patients, CA549 and CA M26 showed a sensitivity only slightly lower (70% and 67%, respectively) than that of TPA which was the highest (78%), while CA M29 sensitivity (58%) was much lower than that of TPA and similar to CEA (57%). In non-relapsed patients, CA M26 and CA M29 showed a specificity (95% and 92% respectively) similar to CEA (86%) and CA15-3 (92%) and much higher than TPA (73%). Among the possible associations of all the studied tumor markers, the CA M26-CA M29-CEA one seems the most promising because of its high sensitivity and specificity. Benign liver diseases and/or diabetes are probably the reasons most often responsible for the aspecific increase in serum CA549 values. On the other hand, they seem to affect the CA M29 much less and the CA M26 almost not at all.</p>","PeriodicalId":76654,"journal":{"name":"The Journal of nuclear medicine and allied sciences","volume":"34 4","pages":"309-13"},"PeriodicalIF":0.0,"publicationDate":"1990-10-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"13246258","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

We used P-Tyr antibodies to investigate the existence of abnormal levels of phosphoproteins in human breast cancer. Three human breast cancer cell lines (SK-BR-3, MCF-7 and CG-5) and 37 human breast cancer specimens were examined by Western blot analysis and \"in vitro\" kinase assay. In the SK-BR-3 cell line three major phosphoproteins of the approximate Mr of 185,000 (p185), 135,000 (p135) and 110,000 (p110) were detected. The former was identified as the HER-2 gene product by specific antibodies against HER-2 encoded protein. In the other cell lines, a product of the approximate Mr of 170,000 (p170), together with a p135 and a p110, were phosphorylated on tyrosine. P185 and p170 were shown to have an associated tyrosine kinase activity. Two proteins, comigrating with p135 and p110, were found to be highly phosphorylated on tyrosine in 50% of the breast cancer samples, but not in samples harvested from 12 human tumors of the gastro-intestinal tract. These data show that 50% of human breast cancer samples display an abnormal level of tyrosine phosphorylated proteins.</p>","PeriodicalId":76654,"journal":{"name":"The Journal of nuclear medicine and allied sciences","volume":"34 4 Suppl","pages":"205-10"},"PeriodicalIF":0.0,"publicationDate":"1990-10-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"12873319","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
