{"title":"Cytochemical investigation on lipids in human major salivary glands. A light and electron microscope study.","authors":"P Sirigu, G Diaz, M S Lantini, M Del Fiacco","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The localization of lipids in the secretion of human parotid and submandibular glands has been here investigated. Ultrastructural observations following lipid extraction and histochemical data showed that lipid components are not present in the secretory granules. In fact, while histochemical reactions are completely negative, secretory granules from lipid-depleted glands retain all their morphological characters. These results led us to emphasize that, at least in human seromucous cells, lipids are not directly involved in the salivary secretion. This conclusion, which is supported by the biochemical data concerning the lipid content in human saliva, disagrees with the findings obtained in rat and guinea pig salivary glands. {"title":"Development and metamorphosis of the larvae of Ascidia malaca in different experimental conditions.","authors":"G Materazzi, L Vitaioli, A M Bondi","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>In larvae of Ascidia malaca that have developed in sea-water without sulphate ions, the cells of the caphalo-ventral region are devoid of sulphated mucopolysaccharides, and the chordal and muscular cells of the tail turn out to be altered; after metamorphosis no adhesive ampullae appear. The addition of cysteine to water lacking sulphate ions leads to a normal synthesis of sulphated mucopolysaccharides in the cells of the cephalo-ventral region, and to a normal secretion of matrix by the chordal cells; but the larvae do not regain the ability to swim. {"title":"[Quantitative limits of the Feulgen reaction: analysis of interference caused by dehistonization and denaturation and renaturation treatments].","authors":"C A Redi","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The Feulgen reaction intensity (measured with a microdensitometer Vickers M86 on the nucleus of erytrocytes of Xenopus laevis Daud.) is increased after dehistonization according to Brody (1974) only if the dehistonization is made before the fixation in acetic acid. The denaturation and renaturation treatments which should act specifically on the screws of the DNA and therefore should not affect the Feulgen reaction, act in a specific manner, probably going away another histonic components. On the dehistonized material the action of the hydrolysis of the Feulgen reaction would add up to that implicit in the dehistonization treatment and would cause a rapid fall of the values for loss of material as consequence of depolymerization facts, according to Andersson and Kjellstrand (1975).
