{"title":"Interaction of estrogen and prolactin in spontaneous mammary tumorigenesis of the mouse.","authors":"C W Welsch","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Daily treatment for 12 months of young nulliparous C3H mice with 0.1 mg 2-bromo-alpha-ergocryptine (CB-154), an effective suppressor of prolactin secretion, significantly reduced and in some instances virtually eliminated the incidence of spontaneous mammary tumors. The CB-154-treated mice had normal body weight gains and normal estrous cycles. Ovariectomy of young nulliparous C3H mice also reduced mammary tumor incicence, but appeared to be slightly less effective than treatment with the ergot. Chronic administration of 17beta-estradiol for 12-19 months (via drinking water) to young nulliparous intact or ovariectomized-hysterectomized C3H mice significantly increased mammary tumor incidence. Concurrent administration of 0.1 mg CB-154 to the estrogen-treated mice significantly reduced the incidence of mammary tumors when compared with mice treated with the steroid alone. Chronic sc administration (0.1 mg/mouse, twice weekly for 20 months) of Enovid to young, nulliparous, intact C3H mice significantly increased mammary tumor incidence, whereas concurrent administration of 0.1 mg CB-154 to the Enovid-treated mice significantly reduced the incidence of these tumors. Thus significant inhibition of spontaneous or estrogen- or Enovid-induced mammary tumorigenesis by concurrent drug-induced prolactin suppression has been demonstrated in these studies.</p>","PeriodicalId":76061,"journal":{"name":"Journal of toxicology and environmental health. Basal levels of circulating PRL follow no consistent pattern in relation to the incidence of mammary tumors. Circulating PRL after a provocative stimulus, however, presents a profile characteristic of the mammary tumor incidence: strains with high incidence of mammary tumors generally have have lower levels, whereas strains with low incidences exhibit higher levels. These differences in circulating PRL levels are apparently not caused by differences in the pituitary stores or pituitary release of PRL. The PRL release patterns appear not to be entirely genetic, since they can be modified by foster nursing, a procedure that may introduce MTV in a virus-free strain. The urine concentrations of PRL and GH and the tissue uptake data suggest that the C3H/St strain may metabolize PRL and GH differently from the C5BL/St strain. It remains to be determined whether the observed differences are significant factors in the development of spontaneous mammary tumors in mice.</p>","PeriodicalId":76061,"journal":{"name":"Journal of toxicology and environmental health. Addition of 5-adenosyl-L-methionine increased the quantity of nonpolar metabolites. Incubation with rat liver microsomes yielded similar results. When polar metabolites from incubation with rat liver microsomes were incubated with catechol O-methyltransferase and S-adenosyl[methyl-3 H]-L-methionine there was conversion to 3H-labeled nonpolar products. Specific binding sites for each of these classes of steroid hormones have been characterized in vivo and in cell-free preparations of lactating mammary gland of the rat. These properties were compared with those of steroid-binding proteins in the R3230AC and dimethylbenz[a]anthracene-induced mammary tumors of the rat and of human breast carcinomas. The binding components that were protein in nature were more concentrated in tissue from lactating than from virgin or pregnant rats. The steroid-binding capacities of mammary tumors were highly variable. Using a dextran-coated charcoal procedure, the rate constants of 3H-ligand association with specific binding sites were temperature dependent. Ligand specificity indicated that [3H]estradoil-17beta(3H-E) binding was inhibited only by estrogens, whereas [3H]triamcinolone acetonide (3 H-TA) was blocked by glucocorticoid/s and progesterone. Likewise, [3H]progesterone (3H-P) binding was inhibited by progestins and certain glucocorticoids. Incubation of minced mammary gland with 3H-steroid in the presence and absence of unlabeled competitor demonstrated specific binding sites in cytoplasm that were translocated to nuclei in a tempreature-dependent fashion. Using a steroid-exchange procedure both DEX and TA inhibited 3H-P binding to exchangeable sites that were loaded previously with unlabeled progesterone. These data suggest that certain glucocorticoid and progesterone binding sites were translocated simultaneously to nuclei. Administration in vivo of either 3H-E or 3H-TA indicated binding in cytoplasm and subsequent translocation to nuclei.</p>","PeriodicalId":76061,"journal":{"name":"Journal of toxicology and environmental health. The native receptor is a 200,000 MW dimer of two dissimilar 4S subunits with different intranuclear function. The receptors directly regulate RNA chain initiation sites in oviduct chromatin by interactions involving target tissue nuclear acceptor sites. There is a 1:1 correspondence between receptor \"acceptor\" sites and RNA sites. Only the dimer form of the receptor is active in vitro on chromatin templates.
