One component, ABP, has been shown to be produced by Sertoli cell cultures for at least 9 days in the absence of exogenously added hormones. FSH (10-100 microgram/ml) markedly enhances the secretion of ABP. MIX has a potentiating effect after long treatment intervals (7 days). In order to study the second component, intracellular androgen receptor, a nuclear exchange assay was developed. Competition for exchange activity using 3H-dihydrotestosterone was significant for a 500 fold excess of testosterone, dihydrotestosteron, progesterone, and cyproterone acetate. The exchange activity was increased 2-10 fold by prior treatment in vitro or in vivo with testosterone. Significant exchange activity was found in long-term hypophysectomized adult and immature animals and in tubule and germ cell fractions. In isolated germ cell fractions, the highest concentration of exchange activity was associated with the most mature elements. After an initial lag phase of about 2 hours, follicle-stimulating hormone (FSH) stimulates synthesis of estradiol up to 50-fold, synthesis being approximately linear for 24 hours when medium is changed every 2-6 hours. Luteinizing hormone (LH) causes only a marginal stimulation, at concentrations consistent with contamination with FSH. Dibutyryl cyclic 3',5'-adenosine monophosphate (dbcAMP) (10(-4)M) mimicks the effect of FSH. No significant synthesis occurs in the absence of steroid substrate or in the presence of pregnenolone (5 X 10(-7) M). When cells are pre-incubated for 24 hours with FSH in the absence of testosterone, addition of testosterone results in an immediated increase in estradiol synthesis without a lag phase, suggesting that FSH or dbcAMP are capable of inducing the conditions necessary for stimulation in the absence of testosterone. Specific testicular oestradiol receptors were present in cytoplasmic and nuclear fractions of interstitial tissue, but not in seminiferous tubules of rat testis. After in vivo or in vitro administration of estradiol the steroid was bound by the nuclear fraction of interstitial tissue only in the presence of the cytoplasmic receptor.
