{"title":"[Seroepidemiologic study of human leptospirosis in Lombardy].","authors":"L Gelosa, A Perone","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>In 1987 a seroepidemiological research was performed to verify the prevalence of leptospira agglutinins on the urban and rural population of Milan, Bergamo and Mantova Provinces in Lombardia Region. Were controlled 150 subjects (75 males and 15 females) in Milan town, 58 (49 males and 9 females) in municipalities of Bergamo Province, and 68 (55 males and 13 females) of Mantova Province. La prevalence of leptospira agglutinins in exposed subjects was 14.3% and in unexposed subjects 8.6%. In the females was noted a significative higher prevalence of agglutinins (14.4%) than in males (9.5%), especially in the females exposed (36.3%) than in females unexposed (9.6%). The leptospira serovars most interested were L. sejroe (15.9%), L. poi (13.6%), L. bratislava, L. canicola and L. Zanoni (9.1%). Thus, in such cases, a completed series of serological and virological assay of HIV antibodies should be performed in follow-up studies. Twenty children living in Modena (Italy) were studied since 1985. Seven of these children were of ages ranging from 3 to 10 years. Only one of them was found to be seropositive for HIV. The remaining 13 children were studied from birth. Six had lost maternal antibodies to HIV at a median age of 15.1 months. Four showed decreasing levels of the anti-body by Western blotting. HIV antigen was detected in serum samples from 2 children. The investigation required the use of the Staphylo-Zyme P.B.I. kit and RPLA Oxoid set. {"title":"Cellular and humoral modifications during response to HBsAg vaccine in healthy subjects: comparison between French and American vaccines.","authors":"R D'Amelio, P M Matricardi, A Di Addario, S Le Moli, E Paganelli, R Nisini, M C Sirianni, A Fattorossi, M Cherchi, C Gaetano","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Two groups of 18 healthy adults, chosen from a risk population, were vaccinated against hepatitis B virus with French (Hevac B, Pasteur) or American (HB-VAX, MSD) vaccine; immunological and biochemical parameters (HBsAb titers, Ig, IgM-rheumatoid factor (IgM-RF) levels, autologous mixed lymphocyte reaction (AMLR), T4/T8 ratio, lymphocyte migration inhibition test (LMIT) with specific antigen and transaminase levels) were compared at preselected time intervals. The vaccines were found to be clinically safe, as documented by the absence of demonstrable side effects and changes in serum transaminase levels. The positivity of the LMIT test demonstrated an early immunization on day 45 in all subjects tested, along with a marked reduction in AMLR, independent of the vaccine used. A slightly higher, though not statistically significant, percentage of seroconversion was observed for the Pasteur vaccine. This same preparation induced a significant increase in the main Ig classes and in Ig;-RF levels on day 45. This data indicates that aspecific modification of the immune system occur to a greater extent with Hevac B, presumably for its lower degree of purification. Fortyfour out of 48 strains (91.7%) resulted tolerant. The high percentage of tolerant was dependent on survivors above the 0.1% allowed in several tube dilutions. Prolonged incubation consistently reduced this number. Only 37.5% of our strains remained tolerant after 48h of incubation. Bactericidal activity of teicoplanin was also significantly influenced by other technical factors. Human serum enhanced the killing of teicoplanin. For 9 strains of Staphylococcus aureus geometric mean MBC decreased from 49.9 muug/ml to 3.9 micrograms/ml (p less than 0.05); also actively growing bacteria demonstrated a higher killing rate, resulting in lower MBCs (mean 3.8 micrograms/ml). Micromethod technique was greatly influenced by the carryover phenomenon. The spot subculture technique is not accurate for bactericidal studies. The experimental and therapeutic employment of many antibiotics has largely been tested \"in vitro\" and \"in vivo\". In the following research we tried to evaluate, by experimental \"in vitro\" method, the sensitivity difference of three serovar strains of Leptospira interrogans to two macrolides, Erythromycin and Josamycin, compared with Penicillin. From standard cultures, previously treated with serial dilution of these antibiotics, the MIC and MSC, as quantitative parameters, have been stated. For the qualitative evaluation of the damages induced at ultrastructural level by the drug activity. Electron Microscopy investigations were performed on specimens from cultures treated for 6 hr with twice and tenfold the MSC. {"title":"Modulation of macrophage functions by corynebacterium parvum preparations.","authors":"L Bonina, D Delfino, G B Costa, A Arena","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Corynebacterium parvum has been used as an anti-tumor agent in both animal experiments and clinical trials; nevertheless it is not well established the mechanism of action, since both T-dependent and T-independent ways of action have been proposed. On the other hand, mononuclear phagocytic system activation cannot be ruled out in the immunomodulation carried out by C. parvum. In the attempt to better understand the effects of C. parvum on macrophage functions, experiments were carried out on tumor-bearing rats with a decline of peritoneal macrophage activities as a result of the tumor development. For this purpose three different preparations of C. parvum were investigated: a) the first preparation was obtained by heat-inactivation of bacteria at 60 degrees C for 1h; b) the second by formalin-treatment of bacteria; c) the third obtained from Burrough Wellcome as a formaline-killed suspension in 0.01% w/v thiomersal saline. Each preparation was used at 7 mg/ml dry weight and administered subcutaneously, three days after tumor grafting. The results showed that all three preparations were capable to hinder the tumor mass growth. Macrophage functions as phagocytosis, intracellular killing, intrinsic and extrinsic anti-Herpes Simplex Virus activities, were restored to normal values by each of the preparations used.
