E. Smith] (FAW) is a serious pest in field corn and sweet corn [Zea mays L.] in many parts of the world. Sweet corn germplasm with effective levels of resistance to damage by the fall armyworm is needed to transfer resistance to commercial hybrids. The objectives of this study were to estimate heritability and to estimate the correlation among some selected morphological traits and their relationship with FAW damage. Seven shrunken-2 (sh2) inbred lines and four commercial sh2 hybrids of sweet corn were crossed to Mp708, a FAW-resistant field corn line. The F2 populations were subdivided with one half being selected for the sh2 trait and the other half was left unselected. Parent, F2, and F3 populations were artificially infested with FAW and evaluated for leaf damage caused by FAW. Heritability estimates for FAW resistance ranged from 0.22 to 0.61 depending on method of estimation used. The highest correlations occurred between silk color and anther color (0.70) and silk color and glume color (0.49). There were no consistent correlations of most morphological traits with FAW damage. {"title":"[The glands of the dorso-distal mucosa of the toad Bufo marinus (Amphibia: Anura) tongue: histology and ultrastructure ].","authors":"Miren González-Elorriaga","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Frequently, the studies on the lingual epithelium ultrastructure in anurans do not show differences between filiform papilae and glandular epithelia in the non-sensory dorsal epithelium. To accomplish the study of glandular epithelium, samples of dorso-distalis mucosae of Bufo marinus tongue were processed for transmission electron microscopy and also semithin sections of the same material were used for light microscopy study. The results showed that the glandular epithelium is constituted by three different types of cells: (1) serous granular secretory cells; (2) mitochondria rich cells (CRM); and (3) ciliated cells. Detailed structures of each type of cell under light and electron microscopy were given. Notably, in the lingual portion studied, secretory cells showed no signs of exocytosis of their granules, thus suggesting that they do not have a basal or continuous rate of secretory activity. In this paper, 15 species of diatoms belonging to the subclass Bacillariophyceae D.G. Mann are described. Water samples from Unare and Píritu lagoons (Venezuela) were collected from November 1988 to July 1989. Samples were fixed with neutral formaldehyde (10%) and were examined under phase contrast microscopy. Species under study were Dictyoneis marginata (Lewis) Cleve, Cymbella affinis (Kützing) Cleve, Cymbella sp., Achnanthes brevipes var. intermedia (Kützing) Cleve, Lyrella irroratoides (Hustedt) Mann, Navicula carinifera (Grunow) Peragallo, Navicula liber (W. The wort was prepared by diluting honey in distilled water to 21% total solids and by adding ammonium sulfate and ammonium phosphate. Saccharomyces cerevisiae was inoculated to the wort (4 g/L). Ethanol production was carried out at room temperature during 84 hours. In this study, 1 Kg of honey yielded about 5 L of wine, containing 8% alcohol (v/v), from a wort with 17.11% total sugars (w/v). The efficiency of the alcoholic fermentation was 81.34%. The acetic fermentation with an inoculum of mixed acetic microorganisms was performed by quick process in a 15 L vertical fermenter. This resulted in a vinegar containing up to 9% of acetic acid (w/v) and about 1% of alcohol (v/v). The acetic fermentation yielded between 91.24 and 97.21%. Approximately 5 L of honey vinegar with 9% acetic acid (w/v) were obtained from 1 Kg of bee honey. The intrinsic resistance of tumoral cells is one of major causes of treatment failure. The overexpression of a membrane associated glycoprotein, P-glycoprotein, in tumoral cell lines, resistant to a wide range of drugs, permitted the description of a multidrug resistance (MDR) phenotype. This P-glycoprotein, which appears to play a role in drug efflux is encoded by the mdr1 gene in humans. The frequent mdr1 gene overexpression in clinically resistant tumours suggest that this gene may be the cause of treatment failure in human cancer. {"title":"Disruption of the intestinal barrier and bacterial translocation in an experimental model of intestinal obstruction.","authors":"R Antequera, A Bretaña, A Cirac, A Brito, M A Romera, R Zapata","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Clinical evidence and the use of experimental models in laboratory animals indicate that the intestine is a reservoir of microorganisms that can cause systemic infection in the human. The purpose of this work was to study the possible effect of intestinal obstruction (IO) on the mechanical and chemical barriers that bring protection against microorganisms crossing from the intestinal lumen towards the systemic tissues. We demonstrated that 24 hours after IO, histological and ultrastructural alterations do occur, seriously compromising the structure of the intestinal barrier in 100% of the studied animals. Likewise, it was observed that during the same period, microorganisms translocation from intestine to the peritoneal cavity and liver (100 and 80% respectively) occurred. The lungs were spared. Changes observed in the intestinal epithelium are related to a process similar to that produced by intestinal ischemia: mitochondrial destruction, with subsequent decrease of its capacity to supply energy and to preserve the equilibrium and structure of the intestinal epithelium. {"title":"[Occupational exposure and health effects of metallic mercury among dentists and dental assistants: a preliminary study. The total population was 66 people, with a sample of 37 (56%), 22 dentists (59.5%, 19 male, 3 female) and 15 dental assistants (40.5%, all female). This was accomplished by an interview, Hg in urine (Hg-U) and N-acetyl-B-D-glucosaminidase activity in urine (NAG-U). Average values of Hg-U for dentists were 22.4 +/- 6.4 micrograms/g creatinine and 22.2 +/- 6.1 micrograms/g creatinine for dental assistants NAG-U average values were 2.9 +/- 3 U/L and 5.2 +/- 8.1 U/L respectively. There were no statistically significant differences between these averages (p > 0.05). There was no correlation between the quantity of amalgam prepared and working hours with Hg-U and NAG-U. Most frequent symptoms referred by dentists were: irritability (54.5%), cephalalgia (45.4%), arthralgias (40.9%), and the ones more referred by assistants were arthralgias (53.3%), irritability (46.7%) and cephalalgia (46.7%). It was not found a significative risk of having them among these groups. There is a need for further investigations including environmental monitoring of Hg, clinical evaluation and neurobehavioural tests to detect early effects. Wood stakes from various wood species particularly Caribbean pine, with 500 mm x 50 mm x 25 mm in dimensions, were treated with several chemicals and concentrations and were installed in natural soil together non treated wood samples. Results concerning natural and induced durability are discussed; observations in Caribbean pine wood showed that the termite attack was responsible for failure of all non treated stakes and some samples treated with triazoles. This cassette would allow the cloning and analysis of a wide range of replicons. The aac(6')-lc amikacin gene was isolated and ligated to the multiple cloning site of the pUC18 vector. This construction was HindIII digested and cloned in the HindIII site of the vector. The resulting pHJ13 clone conferred to the recipient cells the ability to grow in presence of amikacin (cassette marker) and ampicillin (vector gene). By restriction analysis, the cassette orientation was established. Cassette versatility is provided by the presence of the unaltered multiple cloning site segment, and also because it allows sequencing of any replication origin inserted. Cassette functionality was demonstrated by ligation to a replicative region of H plasmid pHH1457. Presence of the ori region from pHH1457 and the aac(6')-lc gene was confirmed in E. coli transformed clones. The incompatibility properties of the pHH1457 and its capability to replicate in a Poll defective strain were preserved in the pHJII14 construct. {"title":"[Measurement of serum amiodarone by protein precipitation followed by high performance liquid chromatography with ultraviolet detection].","authors":"M Valencia, V Zapata, A Bider, M Pérez-González, V Lamanna","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Our aim was to develop a quantitative method for serum amiodarone measurement using high performance liquid chromatography with ultraviolet photometric detection. We studied previous reports in the literature in order to obtain a simpler method to be used routinely in our TDM unit. Sample preparation was based on protein precipitation adding 2 parts of acetonitrile to 1 part of serum, followed by vortex-agitation for 45 s, incubation at 24 degrees C for 5 min, and centrifugation at 6000 x g for 2 min. Twenty microliters of the supernatant was directly injected into the chromatographic system. A microBondapak CN RP column (3.9 x 150 mm) at 45 degrees C, with a mobile phase consisting of KH2PO4 10 mM/methanol/acetonitrile (40:37:23 v/v/v), pH 3.5, were used. Eluting with a flow rate of 0.6 mL/mm the retention time of amiodarone was approximately 4.9 min. Detection was performed at 242 nm and the quantification was made by peak height comparison with external standards. The mass/response ratio is linear (r2 > 0.99) within a mass range of 2.96 to 18,930 ng of injected amiodarone, which exceeds the requirements for the monitoring of serum levels (0.3 to 6.0 micrograms/mL). Sample storage should be done with acetonitrile-extracted sera at -16 degrees C to avoid degradation.
