In contrast to numerous investigations on hypothalamic hormone release, the regulation of the hypophysial-portal blood flow (HPBF) has been scarcely studied. Hypophysial-portal vessels were exposed according to the Worthington's method [1966]. The 10-min blood samples were collected before and during unilateral or alternative bilateral electrical stimulation of the preganglionic fibers of the superior cervical ganglia (SCG). During blood samples collection the stable systemic arterial blood pressure was maintained by a barostat. The HPBF was estimated according to the determination of the hemoglobin in samples of washed and collected blood from the cut pituitary portal vessels. The mean HPBF was 3.5 microliters/min. Electrical stimulation of SCG. did not change HPBF. Non-sonicated liposomes (LMV) and sonicated liposomes (SUV) contained about 20% and 5% of insulin, respectively. Free insulin was removed from liposomes-associated insulin by ultracentrifugation, or ultrafiltration on Sepharose 6B column. Insulin preparations were administered parenterally and non-parenterally into male, Wistar rats with alloxan diabetes to produce the hypoglycaemia. In case of i.v. and s.c. routes of administration all preparations acted in the similar manner giving the clear hypoglycaemia after 2 h. When administered intragastrically only liposome insulin caused hypoglycaemia. Peak inspiratory and expiratory pressures at both sites were recorded in control conditions and then compared to values obtained in the course of progressive denervation of the airways. The two methods of denervation consisted of (1) bilateral section of superior and recurrent laryngeal nerves and of the midcervical vagotomy (horizontal method); (2) right-sided sections of the three nerves followed by left-sided sections (vertical method). Motor denervation of the larynx due to RLNs neurotomy (horizontal method) produced significant increases in intratracheal pressures in both phases of the respiratory cycle. Less prominent increments in pressures were achieved on RLNs neurotomy in the vertical method. SLNs section and vagotomy had little additional effect on airway pressures. The thrombolytic action of kallikrein was mediated by an unstable substance. The generation of this substance was abolished by either acetylsalicylic acid (ASA) or aprotonin and enhanced by captopril. The hypotensive action of kallikrein was only partially inhibited by ASA. It is proposed that both these pharmacological effects of kallikrein are mediated by bradykinin which in turn releases prostacyclin from the endothelium. During handgrip maintained for 4 min at 30% maximal voluntary contraction HR and BP increased significantly while duration of the pre-ejection period (PEP) and isovolumic contraction time (ICT) were shortened with a significant lowering in the ratio of PEP to the left ventricle ejection time (LVET). Occlusion of the circulation to the forearm muscles for 2 min after cessation of exercise did not prevent a rapid decline of HR or increment in PEP, ICT and PEP-to-LVET ratio while BP remained elevated for as long as blood flow to muscles was restricted. Intraduodenal infusion of trypsin (150 mg) during 1 h caused the reduction of alpha-amylase and lipase output by ca 30%. The infusion of aprotinin at the dose of 0.5.10(6) KIU during 30 min caused return of the alpha-amylase and lipase output to the pretryptic values. The infusion of trypsin in higher dose (300 mg) caused more pronounced decrease of amylase and lipase output (ca 45%). Our data indicate that active trypsin in duodenum is responsible for the inhibition of L-phenylalanine stimulated pancreatic enzyme secretion in man. {"title":"Adipogenic activity in the plasma of genetically obese Zucker rats.","authors":"G Sypniewska","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The potential of plasma to stimulate differentiation and lipid filling of adipose precursors in primary culture was investigated in the groups of genetically obese Zucker rats (fafa) and their lean littermates (FaFa). The effect of age, feeding status and possible role of growth hormone in the process of adipogenesis was also studied. Differences in lipid-filling activity of the tested plasma samples were much more dependent on age than the genotype of plasma donors were. The plasma taken from the oldest (20-week-old) rats stimulated the accumulation of triglycerides in the cells to significantly higher levels than the plasma from other rats. The influence of the feeding status on the lipid-filling activity of plasma was not significant. The differentiation potential of plasma in terms of the stimulation of glycerophosphate dehydrogenase activity measured in adipocyte precursors was 30-50% higher when the culture medium contained plasma from obese rats. Furthermore, glycerophosphate dehydrogenase activity in the growing cells declined with age and tended to be higher in the presence of plasma from fed rats. It was the growth hormone that was in a considerable degree responsible for the differentiation potential of Zucker rat plasma. This effect of growth hormone seemed to be less dependent on fafa genotype. Selenium content in the younger group, aged 11-15 years (n = 41), was 13.7 +/- 6.5 micrograms/g-1 creatinine. In the older group, aged 17-97 years (n = 62), slightly but statistically significant lower selenium concentration in urine (11.4 +/- 4.9 micrograms/g Ct, p less than 0.05) was found. We have also shown a significant difference in the excretion of the element between the group of boys and men (p less than 0.05). Norethisterone increased concentration of zymogen granules in the serous acini and also of mucoid material in the mucous-containing acini in the submaxillary gland. In the parotid gland, the acini were hypertrophied, accompanied by cytoplasmic degranulation. In addition, the mitotic cells were seen in both the glands after the treatment.
