DAAO genes have not yet been found in archaean genomes. D-amino acid oxidase is increasingly used in various fields, which requires the development of new variants of the enzyme with specific properties. However, even within one related group (bacteria, yeasts and fungi, mammals), DAAOs show very low homology between amino acid sequences. In particular, this fact is clearly observed in the case of DAAO from bacteria. The high variability in the primary structures of DAAO severely limits the search for new enzymes in known genomes. As a result, many (if not most) DAAO genes remain either unannotated or incorrectly annotated. We propose an approach that uses bioinformatic methods in combination with general 3D structure and active center structure analysis to confirm that the gene found encodes D-amino acid oxidase and to predict the possible type of its substrate specificity. Using a homology search, we obtained a set of candidate sequences, modelled the tertiary structure of the selected enzymes, and compared them with experimental and model structures of known DAAOs. The effectiveness of the proposed approach for discrimination of DAAOs and glycine oxidases is shown. Using this approach, new DAAO genes were found in the genomes of six strains of extremophilic bacteria, and for the first time in the world, one gene was identified in the genome of halophilic archaea. Disruption of their anti-inflammatory functions may lead to the acceleration of immunopathological processes, and to autoimmune diseases, in particular. Unfortunately, the exact mechanism underlying the functioning and development of regulatory B cells (Breg) has not yet been fully elucidated. Almost nothing is known about their specificity and the structure of their B-cell receptors (BCRs). In this research, we analyzed the BCR repertoire of the transitional Breg (tBreg) subpopulation with the CD19<sup>+</sup>CD24<sup>high</sup>CD38<sup>high</sup> phenotype in patients with multiple sclerosis (MS), using next-generation sequencing (NGS). We show, for the first time, that the immunoglobulin germline distribution in the tBreg subpopulation is different between MS patients and healthy donors. The registered variation was more significant in patients with a more severe form of the disease, highly active MS (HAMS), compared to those with benign MS (BMS). CD14 is a common receptor for LPS and LTA that transfers the ligands to TLR4 and TLR2, respectively. In this work, we have demonstrated that the non-toxic LPS from Rhodobacter capsulatus PG blocks the synthesis of pro-inflammatory cytokines during the activation of blood cells by Streptococcus pyogenes LTA through binding to the CD14 receptor, resulting in the signal transduction to TLR2/TLR6 being blocked. Cette pratique fournit énormément de données mais leur interprétation nécessite un certain recul, des outils d’analyse standard permettant de quantifier l’activité acoustique et des données de référence pour évaluer cette activité. Sur l’île de la Réunion aucune référence n’existait jusqu’à présent pour l’interprétation de ces données. Nous proposons ici une méthode de standardisation des données acoustiques ainsi qu’un référentiel quantifiant objectivement l’activité acoustique des trois espèces de microchiroptères actuellement présentes sur l’île : Mormopterus francoismoutoui Goodman, Jansen van Vuuren, Ratrimomanarico, Probst & Bowie, 2008, Taphozous mauritianus É. Geoffroy Saint-Hilaire, 1818 et Chiroptera sp1 (présence observée par l’acoustique mais pas encore confirmée par la capture et la génétique). La finalité est d’objectiver la prise en compte et l’évaluation des enjeux de conservation des chauves-souris dans le cadre d’évaluations environnementales à la Réunion (Mascareignes / Océan Indien). Pour définir ce référentiel d’activité, nous nous sommes appuyés sur des travaux équivalents menés en métropole, en les adaptant au contexte local. For most species, during 11 consecutive years, phenology was rather constant from year to year, which is rather unusual in Mediterranean temporary ponds. The occurrence of summer breeding was not rare for three species: Pelobates cultripes (Cuvier, 1829), Pelodytes punctatus (Daudin, 1803) and Hyla meridionalis Böttger, 1874. Breeding occurrence is linked to rewetting events caused by active karst resurgences, fuelled by periods of intense rainfall episodes called “cévenol” episodes. We document for the first time a triannual breeding occurrence (spring, summer, and autumn), and an in situ record of a short tadpole development period, concerning particularly P. cultripes. The ecological factors which can explain this species richness and these unusual observations are outlined. Geoffroy Saint Hilaire, 1811) en réponse aux variations hydrauliques nous ont amenés à mettre en œuvre trois opérations de radiopistage sur deux tronçons court-circuités de cours d’eau ariégeois. Les opérations ont eu lieu en octobre 2014 et mai 2015 sur l’Aston, puis en septembre 2018 sur le Vicdessos, dans des contextes d’hydrologie variable, avec de fortes variations de débit liées à l’exploitation des ouvrages ou à des travaux de réfection. Six Desmans ont été suivis, dont un au cours des deux opérations sur l’Aston. Les données recueillies montrent que l’espèce présente une activité nocturne et diurne, avec cependant des déplacements plus importants la nuit. Elles ont également mis en exergue le caractère social des individus. En effet, certains partagent tout ou partie de leurs domaines vitaux et certains de leurs gîtes, auxquels ils sont d’ailleurs très fidèles. Nous avons pu observer des réactions aux variations de débit chez les Desmans suivis, parmi lesquelles des changements de gîtes, des translations de domaines vitaux ou bien encore une modification des cycles d’activité. It is characterized by an impaired cognitive function that is due to a progressive loss of neurons in the brain. Extracellular β-amyloid (Aβ) plaques are the main pathological features of the disease. In addition to abnormal protein aggregation, increased mitochondrial fragmentation, altered expression of the genes involved in mitochondrial biogenesis, disruptions in the ER-mitochondria interaction, and mitophagy are observed. Reactive oxygen species are known to affect Aβ expression and aggregation. In turn, oligomeric and aggregated Aβ cause mitochondrial disorders. In the last decade, non-invasive imaging modalities, including optical imaging using non-radioactive substrates, deep-tissue imaging with radioactive substrates, and magnetic resonance imaging have been developed. Reporter genes play the most important role among visualization tools; their expression in tumors and metastases makes it possible to track changes in the tumor growth and gauge therapy effectiveness. Oncolytic viruses are often chosen as a vector for delivering reporter genes into tumor cells, since oncolytic viruses are tumor-specific, meaning that they infect and lyse tumor cells without damaging normal cells. The choice of reporter transgenes for genetic modification of oncolytic viruses depends on the study objectives and imaging methods used. Optical imaging techniques are suitable for in vitro studies and small animal models, while deep-tissue imaging techniques are used to evaluate virotherapy in large animals and humans. For optical imaging, transgenes of fluorescent proteins, luciferases, and tyrosinases are used; for deep-tissue imaging, the most promising transgene is the sodium/iodide symporter (NIS), which ensures an accumulation of radioactive isotopes in virus-infected tumor cells. It is primarily diagnosed clinically. A correct diagnosis of PD in its early stages is important for the development of a pathogenic treatment, which necessitates a search for potential biomarkers of the disease. We evaluated the diagnostic value of several microRNAs and their relationship with the clinical characteristics of PD. The study included 70 PD patients and 40 healthy volunteers. We analyzed the expression of 15 microRNAs in blood leukocytes, which were selected based on literature data and modern concepts of molecular PD pathogenesis. All patients were evaluated using the Hoehn and Yahr scale, UPDRS, NMSQ, and PDQ-39. The data analysis revealed a statistically significant increase in the expression of miR-7-5p, miR-29c-3p, and miR-185-5p and a statistically significant decrease in the expression of miR-29a-3p and miR-30c-1-5p in leukocytes in PD. However, the altered microRNA profile was shown to have a moderate diagnostic value for PD diagnosis. MicroRNA expression changes were associated with the motor and non-motor phenotypic features of PD and administration of anti-Parkinson's drugs. This study showed for the first time that extracellular vesicles secreted by the metastatic melanoma cell lines mel H, mel Kor, and mel P contain, both at the mRNA and protein levels, the α7-type nicotinic acetylcholine receptor (α7-nAChR), which is involved in the regulation of the oncogenic signaling pathways in epithelial cells. Incubation with the vesicles secreted by mel H cells and containing the highest amount of mRNA coding α7-nAChR increased the surface expression of α7-nAChR in normal Het-1A keratinocytes and stimulated their growth. Meanwhile, both of these effects disappeared in the presence of α-bungarotoxin, an α7-nAChR inhibitor. A bioinformatic analysis revealed a correlation between the increased expression of the CHRNA7 gene coding α7-nAChR in patients with metastatic melanoma and a poor survival prognosis. Therefore, extracellular vesicles derived from metastatic melanoma cells can transfer mRNA coding α7-nAChR, thus enhancing the surface expression of this receptor and stimulating the growth of normal keratinocytes.
