{"title":"Malignant pleural mesothelioma: factors influencing the prognosis.","authors":"Tommaso Claudio Mineo, Vincenzo Ambrogi","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Malignant pleural mesothelioma (MPM) is a highly severe primary tumor of the pleura mainly related to exposure to asbestos fibers. The median survival after symptom onset is less than 12 months. Conventional medical and surgical therapies--either as single lines or combined--are not wholly effective. No universally accepted guidelines have yet been established for patient selection and the use of therapeutic strategies. In addition, retrospective staging systems have proved inadequate at improving therapeutic outcomes. Therapy is currently guided by gross tumor characteristics and patient features; however, these seem less accurate than the biological fingerprint of the tumor. A number of clinical prognostic factors have been considered in large multicenter series and independently validated. A series of novel biomarkers can predict the evolution of the disease. {"title":"Prostate cancer: what did we learn from the 2012 Annual Scientific Meeting of ASCO?","authors":"Earle F Burgess, Derek Raghavan","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>In recent years, management of prostate cancer has benefited from an expanding array of new pharmacological options that are the result, in part, of an increased understanding of, and ability to target, androgen signaling. Further characterization of additional molecular pathways relevant to prostate cancer tumorigenesis has identified new molecular targets in prostate cancer that may be amenable to therapeutic intervention with novel agents. In addition, novel radioisotope therapies and cytotoxic agents have shown significant anticancer efficacy with manageable patterns of toxicity, and will eventually have to be integrated into our algorithms of therapy. {"title":"Adult medulloblastoma, from spongioblastoma cerebelli to the present day: a review of treatment and the integration of molecular markers.","authors":"Nicole Shonka, Alba Brandes, John F De Groot","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Although they represent the most common malignant brain tumor in the pediatric population, medulloblastomas are rare in adults, with an incidence of 0.5 per million. With only one exception, all of the prospective clinical trials in this disease have been done in the pediatric population, and therefore therapy for adult medulloblastoma has been either extrapolated from the pediatric literature or based on retrospective reviews. A growing body of literature underscores the genetic similarities between adult and pediatric disease, which may allow tailored therapy directed towards specific molecular pathways and may have an impact on clinical outcomes.
