Materials and Methods:A total of 108 patients with suspected CC admitted to our hospital between March 2020 and May 2022 were retrospectively analysed; 37 patients were diagnosed with CC, and the remaining 71 were determined to have benign colonic lesions.To compare the diagnostic value of MSCT, miR-let-7, and MSCT combined with miR-let-7 detection for CC, we detected the expression of miR-let-7 and high mobility group AT hook protein 2 (HMGA2) in normal human colonic epithelial FHC cells and human colonic mucosal epithelial HCT116 cells and observed the effect of miR-let-7 on the biological behaviour of CC cells.Results: Compared with the results of pathological biopsy, the diagnostic sensitivity of MSCT combined with miR-let-7 for CC was 97.30%, the specificity was 98.59%, and the accuracy was 98.15%, with a misdiagnosis rate of only 1.85%.After miR-let-7 expression was silenced, the viability of colon cancer cells increased, and the inflammatory response and oxidative stress increased.In addition, there was a targeted regulatory relationship between HMGA2 and miR-let-7, which could reverse the effect of miR-let-7 on colon cancer cells.Conclusion: The combination of MSCT and the miR-let-7 assay is accurate for the diagnosis of CC, and miR-let-7 regulates the biological behaviour of CC cells through HMGA2.","PeriodicalId":48815,"journal":{"name":"International Journal of Radiation Research","volume":"23 3","pages":"569-576"},"PeriodicalIF":0.0,"publicationDate":"2025-07-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"http://ijrr.com/files/site1/user_files_fad21f/admin-A-10-1-1369-e1b68db.pdf","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147906937","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Fei, Caicun Zhou, Li Xie, Li Zhou, Wei Wu","doi":"10.61186/ijrr.23.1.201","DOIUrl":"https://doi.org/10.61186/ijrr.23.1.201","url":null,"abstract":"Background: High-intensity focused ultrasound (HIFU) represents a therapeutic medical procedure that operates by inducing ablation and mechanical disruption.Despite its established efficacy, its potential impact on tumor chemotherapy remains uncertain.Long noncoding RNA (lncRNA) MAFA-AS1 facilitated cancer cell proliferation and fostering drug resistance, but the precise significance and functional implications of MAFA-AS1 in the context of ovarian cancer (OC) remain largely unexplored.The objective of this experiment is to further investigate the potential of HIFU in inhibiting the chemotherapy resistance mechanism of OC. Materials and Methods: Five types of human ovarian cancer cells were employed in this study.There were four different groups, namely MAFA-AS1 siRNA, si-NC, pcDNA3.1-HIF-1(hypoxiainducible factor-1), and pcDNA3.1-control.Quantitative real-time PCR, cell proliferation assay, apoptosis assay, western blot assay, subcellular fractionation, aerobic glycolysis analysis, RNA immunoprecipitation (RIP), and luciferase reporter assay were conducted for experimentation and validation.Results: Significant upregulation of MAFA-AS1 in OC cells was observed.Through loss-of-function experiments based on HIFU, we unraveled its oncogenic functions in OC.MAFA-AS1 was discovered to bind to HIF-1 mRNA, thereby enhancing its expressed stability.Further investigations revealed an interaction between MAFA-AS1 and fat mass and obesity-associated protein (FTO), positively modulating HIF-1 mRNA stability in an FTO -dependent manner.Importantly, MAFA-AS1 exerts its effects on OC by acting through HIF-1.Conclusion: The study underscores the role of MAFA-AS1 in promoting aerobic glycolysis and chemical resistance in OC by up-regulating HIF-1 expression, suggesting that targeting MAFA-AS1 holds promise as a therapeutic strategy for OC patients undergoing chemotherapy.","PeriodicalId":48815,"journal":{"name":"International Journal of Radiation Research","volume":"23 1","pages":"201-210"},"PeriodicalIF":0.0,"publicationDate":"2025-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"http://ijrr.com/files/site1/user_files_fad21f/admin-A-10-1-1312-bafa462.pdf","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147904150","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Yin, Yingying Wang, Hongli Ji","doi":"10.61186/ijrr.22.2.277","DOIUrl":"https://doi.org/10.61186/ijrr.22.2.277","url":null,"abstract":"Background:The presence of tertiary lymphoid structures (TLS) is related with good prognosis of various cancers including bladder cancer.The majority of bladder tumors are identified as non-muscle-invasive bladder cancer (NMIBC) when they are discovered in their early stages.We aimed to evaluate the prognostic value and clinical relevance of TLS in NMIBC.Materials and Methods: This cohort included 130 NMIBC samples, with 39 (30%) having TLS, as confirmed by hematoxylin and eosin and immunohistochemistry staining.The chi-square test was utilized to examine the relationship between TLS and the biomarkers and clinicopathologic characteristics of NMIBC.Results: It was found that TLS has significant association with pT stage, Ki-67 expression, and COX-2 expression.Using Kaplan-Meier analysis and Gehan-Breslow-Wilcoxon test, we found that the presence of TLS is associated with longer recurrencefree survival and overall survival of NMIBC patients.Moreover, we used bioinformatics to analyze the association of TLS marker L1CAM and bladder cancer.L1CAM is downregulated in bladder cancer, linked to an advanced stage of the disease, and indicates a poorer prognosis for those who have bladder cancer.In bladder cancer, there is a positive correlation between its expression and B cell infiltration.We also indicated that NMIBC with TLS have a higher level of TLS than that without TLS.
