Fibrianto","doi":"10.25002/tji.2018.682","DOIUrl":"https://doi.org/10.25002/tji.2018.682","url":null,"abstract":"Materials and Methods: Twenty-four of 3-month old male rats that were divided into 6 groups (3 control groups and 3 treatment groups) with different evaluation time (month 2, month 3, and month 4). Treatment in each group was repeated 5 times. The cartilage defect was induced manually and mechanically at the medial condyle of rats’ right femur using a Kirschner Wire (D=1.0 mm; h=1.0 mm). The umbilical cord stem cell cultures were obtained from pregnant rats (19 days). Rats in treatment groups were injected with mesenchymal stem cell-conditioned medium (1 mL/kg BW) 5 times with interval a week after cartilage defect. Histopathological examination of chondrocytes formation and fibrosis tissues was done through Haematoxylin Eosin staining. Increased P. acnes colonization causes a Toll-like receptor (TLR)-2-dependent increase in the production of interleukin (IL)-12 and cathelicidin (LL-37), and a Th-17-dependent increase in interleukin (IL)-17. This study aimed to investigate the relationship between IL-12, IL-17, and LL-37 from patient’s serum and various severities of AV. Materaials and Methods: This study was an analytic observational cross-sectional study. Subjects were enrolled using the consecutive sampling method and assigned according to the Global Acne Grading System (GAGS) criteria. Statistical analysis was performed with one-way analysis of variance and Kruskal-Wallis tests. Results: Mean levels of IL-12, IL-17, and LL-37 in the serum in mild AV were 50.65±6.38, 119.07±24.61, and 180.26±112.92 IU/mL, respectively. The mean levels of IL-12, IL-17, and LL-37 in moderate AV were 47.82±6.51, 132.52±19.41, and 165.91±82.08 IU/mL, respectively. The mean levels of IL-12, IL-17, and LL-37 in severe AV were 48.78±4.93, 208.34±35.38, and 259.50±130.88 IU/mL, respectively. In very severe AV IL-12, IL-17, and LL-37 levels were 39.63, 251.29, and 113 IU/mL, respectively. There were no significant differences between the serum levels of IL12 (p=0.157) and LL-37 (p=0.434) in the different severities of AV, whereas there was a significant association between the serum levels of IL-17 and the severity of AV (p<0.001). Materials and Methods: True experiment post test only control group design was used as research method. Treatments used were control (adjuvant in Tris HCl), and Mab-hZP3 with dosages of 20 μg, 40 μg and 60 μg which tested on day 10, 15 and 20. Luteineizing hormone (LH) receptor, estradiol level and the number of de Graaf follicles measurement were done by immunohistochemical test, ELISA method, and hematoxilin eosin staining, respectively. Results: The results showed that there was no significant difference found on Mab-hZP3 interaction between 20 μg-60 μg dosages and 10–20 days of observation time towards LH receptor expression, estradiol level and and the number of de Graaf follicles. This was related to specificity of monoclonal antibody used. Conclusion: Mab-hZP3 did not lower LH receptor expression, estradiol level and the number of de Graaf follicles quantity, which means that Mab-hZP3 did not interfere at folliculogenesis and not altering hormones profile. Matarial and Methods: In the study, we analyzed the findings of 27 pair pathologic specimens of patients with CRS. Concha nasalis media smear was used to determine the IL-4 and IFN-g expression by real-time polymerase chain reaction (RT-PCR). Data were analyzed by cytokine gene absolute log copy number (CN). Results: Both IL-4 and IFN-g in patients with CRS with nasal polyps CN mean Log of CRS with nasal polyps were higher (15.28±3.92 and 8.54±2.96, respectively) than those of CRS without polyps (14.65±4.65 and 7.57±1.82, respectively), but this was not statistically significant (p=0.596, 0.346 respectively).
