This is particularly important for the production of (new) analytical-grade standards, used as reference materials, and for the execution of clinical trials to test the biological activity of the single compounds that could potentially be used as new drugs or active pharmaceutical ingredients (APIs). To date, various purification strategies, usually based on several steps, have been proposed with different outcomes. However, all these methods have both advantages and disadvantages. The critical step in the chemical analysis is sample preparation. Sorptive sample preparation techniques are among the new generation of microextraction approaches, and are compliant with green analytical chemistry principles. A recent intercontinental collaboration between two academic research laboratories—the Aristotle University of Thessaloniki, Greece, and the Florida International University, USA—has yielded a significant number of analytical/bioanalytical methods using fabric phase sorptive extraction (FPSE), magnet integrated fabric phase sorptive extraction (MI-FPSE), and capsule phase microextraction (CPME) for the isolation of various analytes from different complex sample matrices. Alongside their use as vaccines, they also play a role in protein replacement therapy to ensure therapeutic protein is synthesized within the patient. Structural elements, such as the 5’ cap, UTR regions, reading frame, and poly A tail are considered as critical quality attributes (CQAs) that are subject to a range of analytical techniques. However, chromatography and other separation methods are commonly used for characterization and quantification of the drug substance and drug product. An alternative instrumental method is proposed that uses dynamic extraction in combination with gas chromatography (GC) equipped with a simple flame ionization detector (FID) to achieve these extremely low detection limits. The extraction process was fully automated by means of online solid-phase extraction (SPE). A rational screening strategy for pharmaceutically important peptides has been developed that uses combinations of reversed‑phase ultrahigh-pressure liquid chromatography (UHPLC) columns and mobile phases that exhibit complementary reversed-phase chromatographic selectivity using either UV- or mass spectrometry (MS)-compatible conditions. Numerous stationary and mobile phases were categorized using the chemometric tool of principal component analysis (PCA), employing a novel characterization protocol utilizing specifically designed peptide probes.
