The bacterial community structures and predominant bacterial species were investigated using PCR-Denaturing Gradient Gel Electrophoresis(DGGE),and the correlation between the bacterial community and the main environmental influencing factors were analyzed by using redundancy analysis(RDA). The results of PCR-DGGE showed the dominant bacterial phyla in sediment environment in Xihu area were Proteobacteria, Firmicutes, Bacteroides, Chloroflexi and Acidobacteria. In the bottom part, the dominant bacteria was not obvious in August 2009, but Lactobacillales, Pseudomonadales, Alteromonadales, Chloroflexi and Burkholderiales appeared as alternate dominant groups during the following 5 periods with time going on. For the middle-east part,Lactobacillales, Pseudomonadales, Pseudomonadales, Vibrionales, Pseudomonadales, Lactobacillales were observed as alternate dominant bacteria during the 6 periods. During April to May 2012, an investigation was carried out on the bacterial community structure in ten lakes on northeastern Qinghai-Tibetan Plateau. With 16S rRNA sequencing and PCR-DGGE technology, twelve bacteria genus level libraries were constructed with a total of 117 operational taxonomic unit including 27 Betaproteobacteria, 8 Gamaproteobacteria, 21 Alphaproteobacteria, 7 Deltaproteobacteria,16 Acidobacteria, 2Deinococci, 5 Sphingobacteria, 6 Flavobacteria, 5 Nitrospira, 11 Actinobacteria, 6 Gemmatimonadetes and 3 unclassified bacteria. Most sequences in these studied lakes were closer to aquatic bacteria than any other bacterial clusters. The number of operational taxonomic unit in northeastern Qinghai-Tibetan Plateau was 32.1 ± 11.2, Shannon-Wiener diversity index 0.97± 0.29, Pielou's evenness index 0.19 ± 0.04, and Sorenson similarity index between lakes 0.31 ± 0.11, all comparatively low.Notably, the environment factors played an important role in bacteria community, with temperature positively related to OTUs(R = 0.763, P 0.05), to Shannon-Wiener diversity index(R = 0.708, P 0.05), and to Pielou's evenness index(R = 0.641, P 0.05); the lake area was also positively related to OTUs(R= 0.754, P 0.05) and Shannon-Wiener diversity index(R = 0.668, P 0.05). Water chemistry also played an important part in bacteria community. Hong","doi":"10.3724/SP.J.1145.2013.00781","DOIUrl":"https://doi.org/10.3724/SP.J.1145.2013.00781","url":null,"abstract":"In order to investigate the resistance of Casuarina equisetifolia L. to autotoxicity and to provide scientific basis for continuous planting of C. equisetifolia, clones of ‘Huian No.1’ C. equisetifolia were water cultivated to study the effects of chlorophyll fluorescence in branchlet under allelochemical stress (of seven concentrations: 0, 12.5, 25, 50, 100, 200 and 400 mg L) induced by quercetin-3-α-araboside (allelochemical A) and quercetin-3-β-glucoside (allelochemical B). The membership function of fuzzy mathematics was used to analyze the tolerance abilities of C. equisetifolia seedling under different allelochemicals stress. The results showed slight variations in F0, Fm, Fv/F0 and Fv/Fm of the branchlet under slight allelochemicals stress (12.5, 25, 50 mg L). The Fv/Fm was observed to maintain 78% of normal photosynthesis reaction. The F0, qN, Drate, LPAR of the branchlet significantly (P < 0.05) increased with the concentration of allelochemicals(100, 200, 400 mg L), while the values of Fm, Fv/Fm, Fv/F0, Prate, ΦPSII, qP decreased significantly (P < 0.05). Allelochemicals stress damaged the function of PSII reaction center and decreased the photochemical electron transfer rate, which led to the structural damage of photosynthetic organs and functional degradation in photosynthetic ability. The range of chlorophyll fluorescence parameters in branchlet of C. equisetifolia varied with different allelochemicals,the seedlings had better stress tolerance against allelochemical A than allelochemical B. To study the composition of bacterial community in housefly midgut and assess the role of housefly in the epidemiology of diseases, 16S rRNA gene clone library of bacteria in housefly midgut was constructed. The library compromised of 88 sequences, which were grouped into 22 OTUs, including Proteobacteria(12 OTUs), Firmicutes(9 OTUs), and Bacteroidetes(1 OTU). Twelve OTUs of Proteobacteria belonged to Betaproteobacteria(1 OTU), Epsinoproteobacteria(1 OTU)and Gammaproteobacteria(10 OTUs) respectively. No Alphaproteobacteria or Deltaproteobacteria was found. The 9 Firmicutes included Lactobacillales(6 OTUs), Clostridia(1 OTU) and Bacillales(2 OTUs). In the library, Gammaproteobacteria was the dominant phylotype, which compromised 71 sequences(80.7%). Some species in Enterobacter, Klebsiella, Providencia, Proteus,Aeromonas, and Staphylococcus in the library were pathogens or opportunistic pathogens to human and animal. Lai, Wei Wei, Yuanrun Zheng","doi":"10.3724/SP.J.1145.2013.00096","DOIUrl":"https://doi.org/10.3724/SP.J.1145.2013.00096","url":null,"abstract":"Soil microbial process plays an important role in the conversion of soil organic carbon into inorganic carbon,which is significant for the global carbon cycle.The aim of this study was to investigate the microbial functional diversity of carbon source utilization in three vegetation types under natural conditions along Sangong River basin in Fukang,Xinjiang,China.The three typical vegetation types were dominated by Haloxylon ammodendron,Reaumuria soongorica,and Cleistogenes chinensis,respectively.The soil type is saline soil in this area with varied alkalinity.BIOLOG method was used to determine capability of soil microorganisms in utilizing 31 types of single carbon source.Correlation between soil physicochemical property(pH,water content,total soluble salt) and AWCD(the average well color development),Shannon diversity index,richness value were analyzed.Microbial AWCD in H.ammodendron plot was significantly higher than that of the other two plots(P0.01).pH value(pearson correlation=0.798,P0.01) and total carbon content(pearson correlation=0.655,P0.05) of the soil were closely associated with the value of AWCD,while the effect of water content and total soluble salt was not significant.In this study,the H.ammodendron vegetation had the highest soil total carbon content as well as the highest soil alkalinity while the ability of utilizing carbon source of its soil microbial community was higher than the other two vegetation types.The analysis of normalised optical density(OD) data with PCA(principal component analysis) showed that eleven carbon sources(five types of carbohydrates,two types of amino acid,four types of carboxylic acids) were well used by the soil microbes of H.ammodendron vegetation,six carbon sources(one of the carbohydrates,three of amino acid,two of carboxylic acids) were better metabolized in R.soongorica community.In addition,the soil microbes in C.chinensis vegetation only effectively utilized five out of thirty-one single carbon sources.It was assumed that higher total carbon content and alkalinity would result in higher utilization in the soil microbial community.This result will be benefit for the management of desertification and will contribute to the understanding of the underground carbon process in the saline soil in arid area and the research works on carbon cycle.","PeriodicalId":39895,"journal":{"name":"应用与环境生物学报","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2013-12-13","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"69808017","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
