The results showed that the nitrogen content, total protein and total amino acids in linseed were 3.27%, 20.39%, 92158.22 μg/g FW, respectively, and 2.79%, 17.43%, 92246.59 μg/g FW in sesame seeds correspondingly. The content of unsaturated fatty acids such as linolenic acid and linoleic acid in linseed was 290.57 μg/g, 0.55 μg/g, and 3326.69 μg/g、0.24 μg/g in sesame seed oil. In addition, the contents of palmitic acid, palmitoleic acid, arachidic acid and heptadecanoic acid in sesame were significantly higher than those in linseed. The contents of ergosterol, β-sitosterol and stigmasterol in linseed were 6.57 μg/g, 972.66 μg/g, and 123.27 μg/g, and 5.62 μg/g, 930.57 μg/g, and 145.67 μg in sesame, which were significantly different. The measurement results of metal elements showed that the metal content of linseed d and sesame seed were both Mg>Ca>Fe>Zn, the content of Fe, Mg and Zn in linseed were both higher than that in sesame seed, the Ca element in sesame seeds was higher than that in linseed. It can be seen that linseed oil and sesame seed oil have their own advantages and have different nutrition and health functions.","PeriodicalId":359482,"journal":{"name":"Bioscience Methods","volume":"54 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2021-02-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"114827255","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In order to obtain the paraffin section technology and study the cytological mechanism of morphological formation for fruit tumor in cucumber, we used the samples of warty fruit line (S52) and non-warty fruit lines (S42 with big fruit spine and S06 with small fruit spine) as materials, and improved some key links such as dehydration, dyeing and dewaxing etc., based on traditional paraffin section technology. Results demonstrated that increasing ethanol concentration gradient during dehydration can shorten the treatment time of each stage to 30 minutes. Samples were stained with Ehrlich's hematoxylin for three days, which was convenient for subsequent operation. Optimal thickness of the section was 8 μm. After twice pure xylene dewaxing, sections could be examined and selected by microscope, which shortened the time for obtaining high-quality section. High quality sections with complete tissue and clear structure can be obtained by the optimized paraffin section technology, which is an effective method to observe the cytological structure of fruit tumor. This study provides a set of complete and efficient operation steps of paraffin section technology for cucumber fruit tumor, which will provide technical reference for anatomical researches on other organs in cucumber and structural similar characteristics of other plants.","PeriodicalId":359482,"journal":{"name":"Bioscience Methods","volume":"1 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2021-01-28","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"131149906","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

First, the expression characteristics analysis of miR393 and target gene Ve were performed on the root, stem, leaf, flower and fruit of Solanum torvum Swartz. during flowering and fruiting stage. The strong Verticillium dahliae Kleb. were inoculated into Solanum torvum Swartz. during 5-6 leaf stage, and the control group was treated with ultrapure water. The Solanum torvum Swartz. roots were taken at 0 h, 3 h, 6 h, 12 h, 24 h, 36 h, 48 h and 72 h for expression analysis of infection by Verticillium dahliae Kleb. . The results showed that under normal growth conditions, miR393 and target gene Ve had specific expression patterns in different tissues of Solanum torvum Swartz., and the differential expression was significant. Both miR393 and target gene Ve had the highest expression levels in roots, followed by stems, and had the lowest levels in fruits. After infection by Verticillium dahliae Kleb., both miR393 and target gene Ve had certain expression in different infection periods. The expression of target gene Ve showed an upward tendency with the extension of infection time, and the highest expression level appeared at 48 h, followed by 72 h. While the expression of miR393 showed a downward tendency with the extension of infection time, and the highest expression level appeared at 6 h. The expression of miR393 and target gene Ve showed opposite tendencies in different infection periods. These results provided a foundation for further research on the interaction mechanism between miR393 and target gene Ve and the molecular mechanism of resistance to Verticillium wilt.","PeriodicalId":359482,"journal":{"name":"Bioscience Methods","volume":"91 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"1900-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"114222998","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Genomic DNA of whole plant of P. ternata was used as template, UBC818 primer was used in ISSR-PCR system, L 16 (4 5 ) orthogonal test and single factor experiment were used to optimize the system. Based on the visual analysis of orthogonal experiment, we found the following are the influences of various factors on the experimental results in turn: dNTPs, primer, Mg 2+, Taq DNA polymerase, DNA template.
