Methods: Hep2C (human carcinoma cancer cell line, ATCC:CCL-23) cells were cultured. For cytotoxicity evaluation Hep2C cells exposed to curcumin at different concentrations of 30 µg/ml, 15 µg/ml, 7.5 µg/ml, 3.7 µg/ml, 1.9 µg/ml, 0.9 µg/ml, 0.45 µg/ml for 24 hours These Hep2C cells are evaluated with MTT assay. The IC 50 value of the agent for 24 h of exposure was detected. The graph of the absorbance data obtained by the Spectramax I3 device. Viability values of Hep2C cells calculated from the absorbances obtained from MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay are gained. The preparations were observed based on changes in nuclei and structures using an inverted microscope (Leica Microsystems). Nontreated cells were used as negative control and for positive control Hep2C cells were exposed to ammonium molibdate (1mg/ml) for the above given incubation period. hücre kültürü Results: High doses of curcumin µg/ml, 15 µg/ml, 7.5 µg/ml) showed high antiproliferative and cytotoxic effects on Hep2C cells. The antiproliferative and cytotoxic effects were not observed on cervical cancer Hep2C cells treated with lower concentrations of curcumin. Conclusion: Curcumin has been shown that it is non-toxic, can be used as a highly antioxidant and anti-inflammatory agent and has multifaced therapeutic-pharmacological effects. However, researches on the antiproliferative, anti-cancer effects of curcumin in cervical cancer cells is not sufficient. The present study evaluates the antiproliferative and cytotoxic effects of curcumin on human cervical cancer Hep2C cells as the first time. The results of our study support these effects of curcumin on Hep2C cells in a concentration-dependent","PeriodicalId":35553,"journal":{"name":"Turk hijiyen ve deneysel biyoloji dergisi. Methods: 317 patients who presented with gastrointestinal complaints were included in the study. A questionnaire including; socio-demographic characteristics of the patients, lifestyle and digestive system complaints was applied. Cellophane band method, nativ-lugol, sedimentation and Modifiye kinyoun acid-fast methods were used for the diagnosis of fecal parasites. After the samples were prepared and stained, they were examined under a microscope. Results: Parasite positivity was detected in 205 (64.66%) of 317 patients. Blastocystis spp. constitutes 34.1%, 34.7% of Cryptosporidium spp and 16.1% of Entamoeba coli . The incidence of helminths was 2.8% and the incidence of protozoa was 97.2%. 73.7% of using city water, 93.7% were eating vegetables predominantly. In 9 (4.4%) of the patients with parasite positivity, no parasite was detected at the first examination, and it was detected in the 2nd and 3rd Bakers. The complaints of salivation in Entamoeba coli , Blastocystis spp. and Cryptosporidium spp, and allergy, rectal itching, drooling, increased appetite were significantly higher in Enterobius vermicularis ( p <0.05). Conclusion: It has been observed that there is a high rate of parasites in patients presenting with digestive complaints. Mostly Blastocystis spp., Cryptosporidium spp species were observed in Ordu Province. Patients who present with the complaint of digestion, especially those whose complaints do not improve, should definitely be evaluated in terms of parasites and the preliminary diagnosis of parasitosis should not be immediately ruled out, even if the first stool examination is negative.","PeriodicalId":35553,"journal":{"name":"Turk hijiyen ve deneysel biyoloji dergisi. This study aimed at investigating the antibacterial and anticarcinogenic activities of methanol and hexane extracts derived from I. viscosa . Methods: The antibacterial activity of different concentrations of I. viscosa methanol (1.56-800 mg/ml) and hexane (0.19-100 mg/ml) extracts was tested using the disc diffusion method. Listeria the hexane extract were 400 mg/ml for L. monocytogenes , E. coli ve K. pneumoniae , and 100 mg/ml for M. luteus . MBC values were similar to MIC values. IC50 values for MDA-MB-231 and HT-29 cells were 25 mg/ml and 200 mg/ ml for the methanol extract, and 6.25 mg/ml and 12.5 mg/ml for the hexane extract, respectively. Depending on increased concentration levels, the presence of necrotic and early apoptotic cells was observed among MDA-MB-231 and HT-29 cells. Conclusion: Methanol and hexane extracts of I. viscosa were found to have antibacterial effects on the studied bacteria and anticarcinogenic activity on MDA-MB-231 and HT-29 cells. C. difficile is the commonest cause of nosocomial and antibiotic-associated diarrhea in North America and Europe. It can produce two toxins called A and B. Strains producing toxins can lead to a more serious disease picture that causes megacolon, perforation or septic shock. The use of molecular methods with high specificity and SONUÇLARININ DEĞERLENDİRİLMESİ method. The data obtained are interpreted as positive, negative or invalid results. Results: A total of 109 samples were tested. 66 (61%) of the stool samples belonged to male and 43 (39%) female patients. Twenty-five (23%) of the patients were under the age of 18 and 36 (33%) under the age of 18 years. Of the samples, 26 (24%) were from intensive care units, 60 (55%) were from services and 23 (21%) were from polyclinics. It was found 109 from stool samples. 9 (8%), C. difficile PCR test toxin B positive, 100 (92%) negative. Conclusion: The most important cause of nosocomial diarrhea is C. difficile . The main predisposing factor of infection is the use of antibiotics. Prophylaxis or even one dose of antibiotic therapy may be sufficient for the development of C. difficile infection.
