By considering a multi-story building resting on a piled raft foundation as a typical SSI system, subjected to an earthquake (i.e., Chi Chi earthquake, Taiwan 1999), the time-dependent responses of specified locations in the foundation are recorded and analyzed, both in-time domain and frequency domain. Referring to prior knowledge about system identification and damping, this study suggests an overwhelming approach to determining the damping ratio for analyzing more thoroughly a system of SSI systems. Results are a general procedure to establish the damping matrix, namely the Caughey damping matrix which takes several frequencies into account. A different viewpoint that the connection of superstructure and infrastructure in series could help to estimate the damping, and contribute to a wide range of system identification. All these procedures are used to predict more properly the damping characteristics of a system of structures. Suggestions over the findings would be expected to contribute a more rigorous component to the analysis of high-rise buildings in the future.","PeriodicalId":34742,"journal":{"name":"Ho Chi Minh City Open University Journal of Science Engineering and Technology","volume":"64 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2022-11-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"87804492","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Purpose: This study is designed to establish a protocol for detecting methylation of UCHL1 gene’s promoter in nasopharyngeal carcinoma (NPC) in a Vietnamese population. Materials and methods: 10 samples of NPC biopsy tissues and 10 samples of non-cancerous swabs were collected from the local hospital. Chloroform/Phenol method and Nested-MSP assays were established to detect methylation of a target gene. Results: The isolated DNA reached purity and high concentration which were confirmed by the method of absorbance measurement at 260nm and 280nm. Additionally, the Nested-MSP products of methylation or unmethylation were analyzed and visualized in the agarose gel with the band of 169bps and 210bps, respectively. By sequencing, it was confirmed that the two sets of primer could distinguish the status of methylation and unmethylation of UCHL1 gene’s promoter. Conclusion: Our data suggested that the current protocol could successfully identify the status of methylation and/or unmethylation of UCHL1 gene’s promoter.","PeriodicalId":34742,"journal":{"name":"Ho Chi Minh City Open University Journal of Science Engineering and Technology","volume":"62 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2022-11-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"91141293","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Therefore, an alternative approach should be taken into account. Recently, recombinant monoclonal antibody technology has emerged as the best choice to cure hybridoma related drawbacks. However, recombinant antibodies require known genes for their generation. The purpose of this study is to collect the full-length genes encoding the anti-human CD45 antibody derived from the hybridoma cell line 16E8-F2. In this research, we designed specific primer pairs to amplify the light and heavy chain genes of the antibody through the PCR method. Afterward, the genes were separately cloned into a cloning vector called pJET1.2/blunt. The generated recombinant pJET1.2 vectors will serve as the main material source to manufacture the recombinant monoclonal antibody recognizing human CD45 protein tomorrow.","PeriodicalId":34742,"journal":{"name":"Ho Chi Minh City Open University Journal of Science Engineering and Technology","volume":"11 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2022-11-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"75388742","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The global dissemination of Mobile colistin resistance mcr-harboring bacteria is threatening public health. Such isolates have been detected in various reservoirs including animals, food products, the environment, and humans. From these reservoirs, the isolates are approximate: 53% from animals; 39% from humans; 5% from the environment; and 3% from food. Bacterial distributions were: E.coli 91%; Salmonella 7%; and Klebsiella 2%. Among the 10 mcr variants, mcr-1 and mcr-9 are the most prevalent (up to 95% and 64.53%, respectively, in different studies) while other variants account for 5%. The coexistence of mcr and other antibiotic resistance genes in single isolates is a significant concern; mcr variants carried by different plasmid types increase antibiotic resistance and transfer of mcr genes to other bacteria. The hypothesis that the food chain is intimately involved in mcr gene transfer is supported by the presence of mcr-harboring isolates in animals and foods relative to humans. Cheaper, quicker, and more effective diagnostic tools for detecting colistin-resistant bacterial phenotypes and genotypes are essential and urgent. Currently, high quality tests include RPNP (99.3% specificity, 96.7% sensitivity) and MRPNP (95.4% specificity, ~100% sensitivity). LBJMR, CHROM agar, COL-APSE, and Super Polymyxin are now the best media to screen the bacteria, with near 100% selectivity. Multiplex PCR is a suitable method to quickly and accurately detect mcr genes in E. coli and Salmonella . Multi-drug resistant Gram-negative bacteria remain a global burden and need to have continuous and effective surveillance.","PeriodicalId":34742,"journal":{"name":"Ho Chi Minh City Open University Journal of Science Engineering and Technology","volume":"29 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2022-11-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"85211184","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This study aims to test the quality of DNA isolation extracted from samples of processed food products from chicken meat (chicken nuggets). The novelty of this research lies in the DNA isolation technique modified from the manual kit which is used to produce good-quality DNA isolation results. The extraction method used was the modified extraction method and the standard extraction method of the DNeasy Mericon Food kit. Analysis of DNA quality was measured using a nanophotometer, where the analysis of DNA quality was analyzed based on the parameters of concentration values and purity values. Based on research data, it is known that the modified DNA extraction method produces a concentration value of DNA isolation with an average concentration value of 1295.1 ng/µL. Meanwhile, the value of the purity of the isolated DNA was 2.10 on average. while the standard DNA extraction method resulted in an average DNA isolation concentration value of 111.53 ng/µL. While the value purity of the isolated DNA was at an average value of 1.926. From the research data analyzed, it was found that the modified DNA isolation method has the advantage that the concentration of isolated DNA obtained is higher than the standard method, while the purity value of the DNA isolated from the standard method has better purity when compared to the modified method.","PeriodicalId":34742,"journal":{"name":"Ho Chi Minh City Open University Journal of Science Engineering and Technology","volume":"72 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2022-11-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"83438491","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This has been shown to improve patient survival substantially. The current study is aiming to locally produce an anti-HER2 monoclonal antibody (named Trastuzumab) which has an equivalent biological properties in comparison with the original version, Herceptin®). In silico design and construction of recombinant vectors, as well as the establishment of transfected cell lines with high expression of Trastuzumab were performed. Based on the protein sequences obtained from the Drugbank, the DNA sequences encoding for the light chain (Tras-Lc) and heavy chain (Tras-Hc) of Trastuzumab were optimized and integrated into pNanogen-Hygro and pNanogen-Puro vectors, respectively. The Neon Transfection System was used to co-transfect the pNanogen-Tras-Lc-Hygro and pNanogen-Tras-Hc-Puro constructs into CHO cells. Different co-transfected single-cell-colonies selected on media supplemented with hygromycin and puromycin were used for ELISA and SDS-PAGE assays to identify the CHO cell lines which highly express Trastuzumab. Based on the present results, 30μg of both constructs were suitable for DNA co-transfection. After 07 days of culture, the highest amount of Trastuzumab (561 µg/ml) was obtained from the H06LD68 cell line.","PeriodicalId":34742,"journal":{"name":"Ho Chi Minh City Open University Journal of Science Engineering and Technology","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2022-11-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"79751667","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Since the Khmer language does not always use explicit separators to split words, the definition of words is not a natural concept. Hence, tokenization and part-of-speech tagging of these languages are inseparable because the definition and principle of one task unavoidably affect the other. In this study, different approaches using in Khmer word segmentation and part-of-speech are reviewed and experimental study using a single long short-term memory network is described. Dataset from Asia Language Treebank is used to train and test the model. The preliminary experimental model achieved 95% accuracy rate. However, more testing to evaluate the model and compare it with different models is needed to conduct to select the more higher accuracy model.","PeriodicalId":34742,"journal":{"name":"Ho Chi Minh City Open University Journal of Science Engineering and Technology","volume":"73 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2022-04-20","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"74641255","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
