This study aimed to investigate the effects of High-Intensity Interval Training (HIIT) and probiotic use on the expression of interleukin-10 and interferon-gamma genes in intestinal tissue in an animal model of fatty liver. Methods & Materials: In this experimental study, 40 male Wistar rats (weighing 200-250 g) were selected. The study rats were randomly divided into 5 groups, including healthy control, fatty liver, fatty liver + HIIT, fatty liver + probiotic, and fatty liver + HIIT + Probiotics. Oral tetracycline was administered at a dose of 140 mg/kg of body weight (as a solution in 2 mL of water) for 7 days with fatty liver. HIIT was performed on tape 5 times a week for 5 weeks. The obtained data were analyzed using One-way Analysis of Variance (ANOVA) and Tukey post-hoc test at the significant level of P<0.05. Findings: The present research results suggested that the expression of interleukin-10 and interferon-gamma genes in the intestinal tissue of steatosis mice in the fatty liver group was significantly higher than in the healthy control group (P=0.001). HIIT and probiotic use significantly reduced the expression of interleukin-10 and interferon-gamma genes in the intestinal tissue, compared to the fatty liver group (P=0.001). Conclusion: According to the obtained data, HIIT and probiotic consumption can help modulate the expression of genes involved in the innate immune system of intestinal tissue caused by fatty liver.","PeriodicalId":340278,"journal":{"name":"Quarterly of the Horizon of Medical Sciences","volume":"31 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2021-06-10","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"115033594","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Methods & Materials: Male rats, in groups of 7, received different doses of the methanol extract of harmal seed (100, 200, and 400 mg/kg/d body weight) or placebo via a gastric tube for 42 days. At the end of the experiment, the weights of the body, liver, spleen, and heart, and also the levels of some serum parameters, including glucose, cholesterol, triglyceride, low-density lipoprotein, high-density lipoprotein, insulin, growth hormone, and thyroid hormones, as well as the composition of the carcass were investigated. Findings: The mean weight gain, food intake, and the serum level of triiodothyronine (T3) significantly declined in the rats receiving the extract at 400 mg/kg. In contrast, the serum level of cholesterol in the group increased (P<0.001). The serum levels of glucose were significantly lower in all experimental groups compared to the control. Furthermore, the groups treated with the methanol extract dose-dependently showed higher serum lactate dehydrogenase levels compared to the control (P<0.05). No significant differences were observed regarding other studied parameters compared with the control group. Conclusion: This research suggests positive effects for harmal extract on some metabolic parameters and weight. At least one of the involved mechanisms seems to be declined appetite and hence decreased feed intake.","PeriodicalId":340278,"journal":{"name":"Quarterly of the Horizon of Medical Sciences","volume":"39 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2020-10-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"131649528","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This study aimed to investigate the effect of cellular extract of co-cultured Lactobacillus casei on the expression of BAX and human β-defensin 2 (hBD-2) genes in HT29 cells. Methods & Materials: This experimental study was conducted in the Research Center for Pharmaceutical Nanotechnology of Tabriz University of Medical Sciences in 2017. The HT29 cell line was obtained from the Pasteur Institute of Iran, and cells were assessed using Microculture Tetrazolium Test (MTT) after culturing. DNA was extracted from the treated cells, and then the DNA ladder assay was carried out. After preparing cDNA, the expression levels of BAX and hBD-2 genes in the HT29 cell line were measured using a real-time Polymerase Chain Reaction (PCR) method. Findings: The results of the MTT assay indicated that Lactobacillus casei inhibited the proliferation of HT29 cells and induced apoptosis in these cells. Results of DAPI staining and DNA ladder assay obtained from treating HT29 cells by Lactobacillus casei showed qualitative changes in cell apoptosis. Moreover, realtime PCR results indicated that Lactobacillus casei bacteria significantly increased the expression of the hBD-2 gene in HT29 colon cancer cells within 12-24 hours (P= 0.023), while BAX gene expression showed no significant change in the first 24 hours (P= 0.37). Conclusion: The extract of Lactobacillus casei can be used to stimulate cancer cells to produce β-defensins, inhibit pathogens, prevent the stimulation of cellular signaling, and fight antibiotic-resistant bacteria.","PeriodicalId":340278,"journal":{"name":"Quarterly of the Horizon of Medical Sciences","volume":"58 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2020-10-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"121314031","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The main objectives of this study were to assess antibiotic resistance outlines and detection of antibiotic resistance and virulence genes, including the blaCTX-M and pap among the E. coli isolates recovered from the urine of patients referred to Shaheed Madani Hospital (Bejestan, Northeast of Iran). Methods & Materials This cross-sectional study was conducted from April 2016 to March 2018. A total of 100 non-duplicate isolates of E. coli were recovered from the urine of patients referred to Shaheed Madani Hospital (Bejestan, Northeast of Iran). Antimicrobial susceptibility test and extended-spectrum-beta-Lactamases (ESBLs) production were performed according to Clinical and Laboratory Standards Institute (CLSI) guidelines. The polymerase chain reaction was carried out for the detection and distribution of the pap virulence gene and blaCTX-M antibiotic resistance gene. Findings About 100 E. coli isolates were recovered from the urine sample of patients (21 male, and 79 female). Carbapenems were the most effective antibiotic against isolates. Four strains (4%) were resistant to colistin. Twenty-seven strains (27%) were ESBL-positive. Carbapenems were the most effective antibiotic against ESBL-positive strains. Also, 82% and 89% of isolates were blaCTX-M and pap gene positive, respectively. Conclusion This study has shown that ESBL-positive strains with a high level of drug resistance and virulence factors are a potential risk for hospital wards. Colistin resistant isolates found in our study are quite alarming.","PeriodicalId":340278,"journal":{"name":"Quarterly of the Horizon of Medical Sciences","volume":"99 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2020-10-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"132827969","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
