The purpose of this research is to analyze the Length-weight relationship of Bigeye scad caught by purse seine in Manado Bay. as well as to evaluate the results of the analysis of the Length-weight relationship and the catchment feasibility of the catch to support the sustainability of the potential of bigeye scad resources. The method used in this study is a quantitative method. conducted at Tumumpa Manado Coastal Fisheries Port, using fork length measurement data (FL) and weight of bigeye scad taken by the author under the guidance of lecturers in May 2021 and November 2021. Length-weight relationship of bigeye scad caught in May 2021 follows the equation W = 0,0249 L2.8057 with K = 1.01 and in November 2021 W = 0,0207 L2.9151. The type of bigeye scad in both seasons is categorized as negative allometric. This fish is categorized as unfit to catch (Lc < Lm). The structure of the catch in May 2021 obtained an average length of 10.26 ± 0.26 cm and an average weight of 18.03 ± 1.51 g where 51% were caught at a size of 9.8-10.8 cm and 46% had a weight of 11.0-16.0 g. while in November 2021 the average length was 10.90 ± 0.28 cm and the average weight was 22.53 ± 1.76 g where 50% were caught at a size of 10.0-11.0 cm and 31% g weight 21.0-26.0 g.Keywords: Bigeye Scad; Length Weight Relationship; Decent Catch; Purse Sein; Manado BayAbstrakIkan Selar bentong memiliki cita rasa enak, disukai masyarakat, dan harganya pun murah. Intensitas ketertangkapan jenis ikan ini, diduga kelak akan menimbulkan masalah ketersediaan sumberdaya ikan ini. Tujuan penelitian ini menganalisis hubungan panjang berat ikan selar bentong hasil tangkapan kapal pukat cincin (purse seiners) di Teluk Manado, serta Mengevaluasi hasil analisis hubungan panjang berat ikan selar bentong dengan kelayakan tangkap hasil tangkapan untuk menopang keberlanjutan potensi sumberdaya ikan selar bentong ini. Metode yang digunakan pada penelitian ini yaitu metode kuantitatif, dilakukan di Pelabuhan Perikanan Pantai (PPP) Tumumpa Manado, dengan menggunakan data ukuran panjang cagak (fork length, FL) dan berat ikan selar bentong yang diambil oleh penulis di bawah bimbingan dosen pada Mei 2021 (musim Pancaroba I) dan November 2021 (musim Pancaroba II) yang tertangkap purse seiners di Teluk Manado. {"title":"Evaluation of 16S rRNA Gene Sequence for DNA Barcoding of Tuna Fish","authors":"B. This is due to its requirement of a small amount of tissue sample in order to conduct a full analysis for species identification. This research aimed to conduct an assessment for the use of 16S rRNA gene sequence for tuna fish identification through DNA barcoding. Previous in silico studies using the COI gene and CYB gene were conducted using the same tuna fish specimens. A comparison of 16S rRNA gene sequence between Bluefin tuna (five species), Yellowfin tuna (three species), and another group of tuna (five species) showed the reliability of this gene in differentiating all species represented by the respective specimens. The multiple sequence alignment of 1695 bp in this research is reliable for accurate identification. All specimens of Thunnus (Bluefin group and Yellowfin group) were able to be differentiated with other genera (Auxis, Euthynnus, and Katsuwonus) group in 27 sites. The other tuna fish genera group members are similar in 27 sites and the member Thunnus group has a polymorphism in the same location. The similarity among the Bluefin group is 99.3% to 99.8%. The similarity among Yellowfin group is 99.6% to 99.9%. The similarity among the other group is 97.5% to 99.5%. In conclusion, the 16S rRNA gene is a reliable marker for DNA barcoding of tuna fish.Keywords: DNA barcoding; 16S rRNA gene; tuna fishAbstrakUntuk kepentingan autentikasi produk-produk dari ikan, DNA barcoding merupakan perangkat yang dapat diandalkan. Ini disebabkan karena metode ini hanya membutuhkan sedikit sampel jaringan untuk analisis identifikasi spesies. Penelitian ini bertujuan untuk melakukan asesmen penggunaan sekuens gen 16S rRNA untuk identifikasi ikan tuna melalui DNA barcoding. Studi in silico sebelumnya untuk gen COI dan gen CYB dilaksanakan menggunakan spesimen ikan tuna yang sama. Perbandingan sekuens gen 16S rRNA antara kelompok ikan tuna Bluefin (lima spesies), kelompok ikan tuna Yellowfin (tiga spesies), dan kelompok ikan tuna jenis lain (lima spesies) menunjukkan kemampuan gen ini dalam membedakan semua spesies. Dari hasil yang diperoleh, penjajaran multisekuens dari 1695 bp dapat diandalkan untuk indentifikasi yang akurat. Semua spesimen genus Thunnus (kelompok Bluefin dan kelompok Yellowfin) dapat dibedakan dengan spesimen lainnya pada 27 titik nukleotida. Kelompok tuna jenis lain juga memiliki nukleotida yang seragam di 27 lokasi dibandingkan dengan yang dari genus Thunnus. Tingkat kesamaan antar spesimen kelompok tuna Bluefin yaitu 99,3% sampai 99,8%. Tingkat kesamaan antar spesimen kelompok tuna Yellowfin yaitu 99,6% sampai 99,9%. Identification of mangroves and sea slugs refers to the identification book. The results obtained were L. pallescens species with elongated and tapered morphology at the end of the shell measuring 0.3-2.7 cm. The operculum is purple. The color of the shell obtained was 66.85% consisting of dark colors (black, black, orange, brown and gray spots), occupying the stems and roots of the mangrove, while the light colors (yellow, yellow, dark spots and red) were found to be 33.15%, occupying the leaves and stems of mangroves. The high survival rate of L. pallescens was found in the mangrove roots. This species was found in R. mucronata by 65.26% while in A. marina only 34.74%, this could be caused by differences in the shape of the mangrove roots.Keywords: L. pallescens; Mangrove; Shell color; MorphologyAbstrakTujuan penelitian ini adalah mengindentifikasi siput laut L. pallescens yang diambil dari dua mangrove berbeda yaitu Rhizopora mucronata dan Avicennia marina di perairan Tongkaina, Kecamatan Bunaken, Kota Manado berdasarkan morfologi dan anatomi serta warna cangkang. Identifikasi mangrove dan siput laut merujuk pada buku identifikasi. Hasil penelitian yang diperoleh yaitu spesies L. pallescens dengan bentuk morfologi memanjang dan meruncing, pada bagian ujung cangkang berukuran 0,3-2,7 cm. Operculum berwarna ungu. Warna cangkang yang diperoleh 66,85% yang terdiri dari warna gelap (hitam, hitam bercak orange, coklat dan abu-abu), menempati bagian batang dan akar mangrove sedangkan warna terang (kuning, kuning bercak gelap dan merah) didapat 33,15%, menempati bagian daun dan batang mangrove. Tingginya kelangsungan hidup L. pallescens berada pada bagian akar mangrove. Spesies ini ditemukan pada R. mucronata sebesar 65,26% sedangkan pada A. marina hanya 34,74%, hal ini dapat disebabkan oleh karena perbedaan bentuk akar mangrove. Kata Kunci: L. Tidal research is useful for providing information about the components and types of tides and predicting tides and for mitigating natural disasters. Tidal measurements were carried out in the Nasik Strait, Bangka Belitung using the Valeport 106 tool. Tidal data was processed using the Admiralty Method. The Admiralty method is a method used to calculate tidal harmonic constants from observations of water levels. With this method, it is possible to know the amplitude and phase difference of the tides, the value of the Formzahl number, and the type of tide. From the results of data processing and analysis, the value of the Formzahl number in the waters of the Nasik Strait is 11,454 and the tidal type is a single daily tidal type that only occurs at one high tide and one low tide in one day.Keywords: tides; tidal types; admiralty method; formzahl numberAbstrakPasang surut adalah naik turunnya muka air laut yang disebabkan oleh gaya tarik benda-benda di langit, terutama bulan dan matahari terhadap massa air laut yang ada di bumi. Penelitian pasang surut bermanfaat untuk memberikan informasi mengenai komponen dan tipe pasang surut serta memprediksi pasang surut serta untuk mitigasi bencana alam. Pengukuran pasang surut dilakukan di Perairan Selat Nasik, Bangka Belitung dengan menggunakan alat Valeport 106. Data pasang surut diolah dengan menggunakan Metode Admiralty. Metoda Admiralty merupakan metode yang digunakan menghitung konstanta harmonik pasang surut dari pengamatan ketinggian air. Dengan metoda ini dapat diketahui amplitudo dan beda fase pasang surut, nilai bilangan Formzahl dan tipe pasang surutnya. The role of seagrass in shallow marine waters is as a primary producer, as a habitat for biota, catching sediments, and a nutrient recycler. The existence of seagrass is influenced by several factors, namely: temperature, salinity, depth, brightness, nutrients, and salinity. The purpose of this study was to determine the Relative Density, Relative Abundance, Relative Dominance, Frequency, Relative Frequency, Important Value Index, Diversity Index, and Dominance Index, to determine the types of seagrass and to determine the condition of the aquatic environment. This research uses the quadratic methods and line transect. This research was conducted on May 28, 2021, at Meras Beach, Bunaken District, Manado City, North Sulawesi. The number of stands of seagrass species in the study area ranged from 23-320 individuals, species density (8.36-116.36 individuals/m2), relative density (3.62-50.47%), frequency of presence (0.037- 0.50 ), relative frequency (3.62- 50.47%), dominance index (0.072-1.009), the diversity index (1.236), index of the importance of seagrass in Meras Coastal Waters showed that Cymodocea rotundata had the highest important value index among the 5 seagrass species, namely 151.41%. There are 5 species of seagrass found in Meras Coastal Waters, namely, Enhalus acoroides, Syringodium isoetifolium, Halophila ovalis, Cymodocea rotundata, Thalassia hemprichii. The environmental conditions in Meras Beach are temperature 29°C, salinity 35‰, the brightness is quite clear and has a substrate of sand, muddy, sand mixed with mud, muddy mixed with sand, and coral fragments.Keywords: Meras Beach; Seagrass; Community Structure.AbstrakLamun (seagrass) adalah tumbuhan berbunga (Angiospermae) yang tumbuh dan berkembang biak pada dasar perairan laut dangkal, mulai daerah pasang surut (zona intertidal) sampai dengan daerah sublitoral. Keberadaan lamun dipengaruhi oleh beberapa faktor yaitu: suhu, salinitas, kedalaman, kecerahan, nutrient dan salinitas. Tujuan penelitian ini adalah untuk mengetahui Kepadatan Relatif, Kelimpahan Relatif, Dominasi Relatif, frekuensi, Frekuensi Relatif, Indeks Nilai Penting, Indeks Keanekaragaman, dan Indeks Dominasi, untuk mengetahui jenis-jenis lamun dan untuk mengetahui bagaimana kondisi lingkungan perairan. WPPNRI 716 is a management area in Indonesian waters which includes the waters of the Sulawesi Sea and the northern part of Halmahera Island. This research was carried out by the purse seiner KM. Rebert carried out fishing operations in the waters of the Sulawesi Sea at WPPNRI 716. Data collection was carried out for 4 months, namely from October 2021 - to January 2022, with the aim of knowing the distribution area of catching and KM. Rebert catches. KM. Rebet catchment area based on the GPS point is located at WPPNRI 716, which is 81.5 miles from the fishing base (Tumumpa Beach Fishery Port). The number of catches in October (trip 1) was 6350 kg, in October (trip 2) was 4807 kg, in November (trip 3) was 10245 kg, in December (tip 4), was 4234 kg, in December (trip 5) as much as 4280 Kg, in January (trip 6) as many as 2645, and in January (trip 7) 4350 Kg. This study used the quadrat transect method. This study found differences in the number of ascidian species with water depth, 11 species of 5 families at 15 M depth, and 8 species of 3 families at 7 M depth. The diversity index ranged from 0.868 to 1.844 at 15 M depth and 0.965 to 1.864 at 7 M depth, the evenness index was 0.533 – 0.839 at 15 M depth and 0.600 – 0.897 at 7 M depth, the dominance index was 0.254 – 0.745 at 15 M depth and 0.254 – 0.708 at 7 M depth. Ascidian in Manado Bay had two distribution patterns, a uniform distribution pattern and a clustered distribution pattern. Environmental parameters had a water temperature of 27 oC – 31 oC, the salinity of 30 0/00 – 32 0/00, the brightness of 12 m – 14 m, and pH of 8 – 10.*Keywords: Ascidian; diversity; ecological index; distribution patternAbstrakTujuan dari penelitian ini adalah untuk mengetahui distribusi dan keanekaragaman jenis ascidia di perairan Teluk Manado meliputi: komposisi jenis, kepadatan individu, keanekaragaman, dan dominansi. Serta mengetahui pola distribusi ascidia. Penelitian ini menggunakan metode transek kuadran. Pada penelitian ini ditemukan perbedaan jumlah spesies ascidia menurut kedalaman, 11 spesies dari 5 family pada 15 M dan 8 spesies dari 3 famili pada kedalaman 7 m. Nilai indeks keanekaragaman ascidia di kedalaman 15 m = 0.868 – 1.844 dan 7 m = 0.965 - 1.864, indeks keseragaman 15 m = 0.533 – 0.839 dan 7 m = 0.600 – 0.897, indeks Dominasi15 m = 0.254 – 0.745 dan 7 m = 0.254 – 0.708. Ascdia di perairan Teluk Manado memiliki dua pola distribusi yaitu pola distribusi seragam dan pola distribusi mengelopok. The aim of this research is to identify the morphology, of meristic crabs and the effect of the moon phase on species diversity. Sampling locations were on the coast of Pondang Village and Lopana Village, sampling using the roaming method and carried out at the two lowest low tide phases during the dead moon and full moon phases, during the lowest low tide crabs were generally found with walking legs. Based on the identification of the morphology of crabs found on the coast of Pondang Village and Lopana Village, East Amurang District, South Minahasa Regency, there are six species of them Ocypode ceratophthalmus (Stimpson, 1858), Grapsus albolineatus (Latreille in Milbert, 1892), Eriphia sebana (Shaw and Nodder, 1803), Atergatis floridus (Linnaeus, 1767), Pilumnus vespertilio (Fabricius, 1793), Episesarma mederi (Edwards, 1853). Tujuan penelitian mengidentifikasi secara morfologi, meristik kepiting dan pengaruh fase bulan terhadap keanekaragaman spesies. Lokasi pengambilan sampel di pesisir pantai Kelurahan Pondang dan Desa Lopana, pengambilan sampel menggunakan metode jelajah serta dilakukan pada dua fase surut terendah saat fase bulan mati dan purnama, saat surut terendah umumnya dijumpai kepiting yang memiliki kaki jalan. Corals are invertebrates belonging to the phylum Coelenterata (hollow animals) or Cnidaria. In order to preserve the coral reef ecosystem in the future in the Bahowo area, quantitative data is needed that can explain/describe the condition of coral reefs. The purpose of this study was to determine the condition of coral reefs, in this case, data on coral cover and associated biota in Bahowo waters. The data collection of this research used the UPT (Underwater Photo Transect) method. Analysis of the data in the form of research images using the CPCe (Coral Point Count with Excel extensions) application. The results of the analysis of the condition of coral reefs in Bahowo waters are in the damaged/bad category with live coral cover percentage data of 16.33%.Keywords: Live coral cover; Underwater photo transect (UPT); Coral reef condition; Bahowo watersAbstrakTerumbu karang merupakan ekosistem pesisir dengan tingkat keanekaragaman tertinggi dengan jumlah sekitar satu juta spesies di seluruh dunia dan merupakan habitat bagi kumpulan dari berjuta-juta hewan polip yang menghasilkan zat kapur membentuk skeletonnya dan berkembang menjadi hamparan koloni yang luas. Karang adalah hewan tak bertulang belakang yang termasuk dalam filum Coelenterata (hewan berongga) atau Cnidaria. Dalam rangka pelestarian ekosistem terumbu karang ke depan di daerah Bahowo, maka dibutuhkan data kuantitatif yang dapat menjelaskan/menggambarkan tentang kondisi terumbu karang. Tujuan dari penelitian ini adalah untuk mengetahui kondisi terumbu karang, dalam hal ini data tutupan karang dan biota asosiasi di perairan Bahowo. Pengambilan data penelitian ini menggunakan metode UPT (Underwater Photo Transect). Analisis data berupa gambar penelitian menggunakan aplikasi CPCe (Coral Point Count with Excel extensions). Juvenile coral is the result of metamorphosis and growth of coral planula measuring 5 cm and attached to certain substrates. There are two types of substrates in the waters, stable substrates and unstable substrates such as rubble. The purpose of this study was to study hard coral recruitment, both in terms of density, the composition of juvenile coral colony types, size, and the type of substrate occupied by juvenile corals. This research was conducted in August 2021 in the waters of Molas, Bunaken District, Manado City. The method used for site selection is the purposive sampling method and data collection using a sampling method with quadrant transects. The results showed that the density of coral recruitment at the study site was 8,43 colonies/m2. The percentage of juvenile coral attachment on stable substrates is 90% and 10% on unstable substrates. The average size of juvenile corals at the study site was 2.4cm. The composition of the juvenile coral genus found at the research site were corals of the genus Acropora, Anacropora, Coeloseris, Cycloseris, Cyphastrea, Echinopora, Favites, Fungia, Galaxea, Goniastrea, Isopora, Leptastrea, Leptoseris, Lobophyllia, Merulina, Montastrea, Montipora, Oulophyllia, Pachyseris, Pavona, Platygyra, Plasiastrea, Pocillopora, Porites, Psammocora, Scolymia, Seriatopora, Symphyllia, and Trachyphyllia. Of the entire genus, corals of the genus Porites were dominated by the number of juveniles as many as 36 colonies.Keywords: Molas, coral, recruitment, ScleractiniaAbstrakRekrutmen karang adalah masuknya individu karang baru pada populasi terumbu karang dikarenakan reproduksi ataupun migrasi. Juvenil karang merupakan bentuk hasil metamorphosis dan pertumbuhan planula karang yang berukuran ≤ 5 cm dan menempel pada substrat tertentu. Terdapat dua tipe substrat di perairan, substrat yang stabil dan substrat yang tidak stabil seperti pecahan karang (rubble). Tujuan penelitian ini adalah untuk mempelajari tentang rekruitmen karang keras, baik dari densitas, komposisi jenis koloni juvenil karang, ukuran, hingga tipe substrat yang ditempati oleh juvenil karang. Penelitian ini dilakukan pada bulan Agustus 2021 di perairan Molas Kecamatan Bunaken Kota Manado. Metode yang digunakan untuk pemilihan lokasi adalah metode purposive sampling dan pengumpulan data menggunakan metode sampling dengan transek kuadran. Hasil penelitian menunjukan bahwa densitas rekruitmen karang pada lokasi penelitian sebesar 8,43 koloni/m2. Persentase penempelan juvenil karang pada substrat stabil sebesar 90% dan pada substrat yang tidak stabil sebesar 10%. Ukuran rata-rata dari juvenil karang pada lokasi penelitian adalah 2,4cm.
