{"title":"Exploring Metabolites Of Green Algae Caulerpa Spp. To Discover Putative Inhibitors Of Dpp-4, A New Antidiabetic Target Protein","authors":"Walter Balansa","doi":"10.35800/jip.v11i2.48061","DOIUrl":"https://doi.org/10.35800/jip.v11i2.48061","url":null,"abstract":"Diabetes mellitus (DM) remains a serious global health threat, claiming a million lives every year and affecting nearly 9% of the adult population who suffer from this impaired insulin sensitivity disease. The Dipeptidyl peptidase-4 (DPP-4) enzyme has recently attracted attention because of its crucial role in insulin signaling, making this enzyme an interesting and emerging target for antidiabetic drug discovery. This study aimed to explore reported metabolites from marine algae of the genus Caulerpa as DPP-4 inhibitors through computational studies using CB-dock 2, Protein-Ligand Interaction Profiler, SwissAdme, and pkCMS. Molecular docking allowed the identification of 7 hit compounds with strong binding affinities (8.4 kcal to 9.3 kcal/mol) against DPP-4 target enzyme PDB ID: 3G4I. Four hits showed stronger binding affinity than two DPP-4 specific inhibitors and FDA-approved antidiabetic drugs, sitagliptin (8.4 kcal/mol) and linagliptin (9.0 kcal/mol). Following a molecular modification of the hit compounds using a bioisosterism-like approach and ADMET evaluation with pkCMS, three putative DPP-4 inhibitors were identified. They showed either stronger binding affinities or better ADMET profiles than sitagliptin and linagliptin, suggesting their promising potential as DPP-4 inhibitors. However, further optimized bioisoterism, in silico and in vivo studies, and clinical-based trials are required to confirm their antidiabetic activity. The practicality, user-friendliness, and short-time analysis of BLM make this method a good choice for spicule analysis. However, its effectiveness and accuracy are yet to be confirmed. This study aimed to validate the effectiveness of BLM by comparing its usefulness to both TLM and the gold standard methods. BLM was first subjected to measuring megascleres and microscleres of 2 sponges. Then, by using the If function built-in Excell and t-test in SPSS 16.0, the compatibility of BLM was evaluated against SEM by measuring the length of spicules from 4 Sangihe sponges and their counterpart species from different locations. Furthermore, the t-test analysis was used to validate the compatibility and effectiveness of our method to the TLM by measuring the spicules of four sponges. Both the F-function and the t-test analysis proved BLM was compatible with SEM with both measurements showing a perfect match for megascleres typed spicules of 4 compared sponges. This new technique also showed a perfect match with SEM (p = 0.367, t-test) and with TLM (p = 0.963, t-test). This study aimed to determine the physicochemical characteristics of trash fish nuggets added with Eucheuma cottonii seaweed. The physicochemical characteristics include pH, proximate levels such as protein, fat, and carbohydrates, water content and ash content, and the essential mineral content of zinc (Zn). Trash fish nuggets with the addition of wheat flour alone have characteristics, including pH of 6.8-7.1, water content of 40.5%, ash content of 1.04%, protein content of 40.3%, fat content of 7 0.07% and carbohydrate content of 10.12%, zinc content of 2.55 g/g nugget weight, while trash fish nuggets with the addition of a mixture of wheat flour and E. cottonii had a pH of 6.8-7.1, water content of 42.5%, ash content of 1.22%, protein content of 42.9%, fat content of 5.97% and carbohydrate content of 4.11% and zinc content of 4.56 g/g nugget weight. Based on the organoleptic examination, trash fish and E. cottonii nuggets were chewy with a delicious and savory taste. Penelitian ini bertujuan untuk menentukan karakteristik fisikokimia dari nugget ikan rucah yang ditambahkan dengan rumput laut Eucheuma cottonii. Karakteristik fisikakimia meliputi pH, kadar proksimat seperti protein, lemak dan karbohidrat, kadar air dan kadar abu serta kandungan mineral essensial seng (Zn). Nugget ikan rucah dengan penambahan tepung gandum saja memiliki karakteristik antara lain pH sebesar 6,8-7,1, kandungan air sebesar 40,5 %, kadar abu sebesar 1,04 %, kadar protein sebesar 40,3 %, kadar lemak sebesar 7,07 % dan kadar karbohidrat sebesar 10,12 %, kandungan seng sebesar 2,55 μg/g bobot nugget, sedangkan nugget ikan rucah dengan penambahan campuran tepung gandum dan E. cottonii memiliki pH sebesar 6,8-7,1, kandungan air sebesar 42,5 %, kadar abu sebesar 1,22 %, kadar protein sebesar 42,9 %, kadar lemak sebesar 5,97 % dan kadar karbohidrat sebesar 4,11 % serta kandungan seng sebesar 4,56 μg/g bobot nugget. Berdasarkan pemeriksaan organoleptis menunjukkan produk nugget ikan rucah dan E. cottoni lebih kenyal dengan rasa yang lezat dan gurih. The location of the Water Tourism Park Conservation Area of North Minahasa Regency has been reserved by the North Sulawesi Provincial Government through Governor Decree Number 407 of 2018 covering an area of 25,838.91 hectares. The purpose of this research is to formulate a strategy for the sustainable development of the Water Tourism Park Conservation Area in North Minahasa Regency. This research was conducted for 6 months starting from June - December 2022. The method used in this research is a descriptive research method with survey techniques, namely by collecting data through interviews with relevant stakeholders as respondents. The results of this study obtained 10 (ten) priority strategies for the sustainable development of Water Tourism Park Conservation Area in North Minahasa Regency which will be recommended to the local government, in this case the Marine and Fisheries Office of North Sulawesi Province and related stakeholders as suggestions and materials that can be proposed in the context of making public policy in the field of marine and fisheries. Key words: Conservation; North Minahasa; Sustainable Development; Strategy; Water Tourism Park Abstrak Pemerintah Indonesia telah menargetkan 32,5 juta hektar kawasan konservasi perairan atau 10% dari luas perairan Indonesia pada tahun 2030. Lokasi Kawasan Konservasi Taman Wisata Perairan Kabupaten Minahasa Utara telah dicadangkan oleh Pemerintah Provinsi Sulawesi Utara melalui Surat Keputusan Gubernur Nomor 407 Tahun 2018 seluas 25.838,91 hektar. Tujuan penelitian ini untuk merumuskan strategi pengembangan berkelanjutan Kawasan Konservasi Taman Wisata Perairan di Kabupaten Minahasa Utara. Penelitian ini dilakukan selama 6 bulan dimulai dari Juni - Desember 2022. Metode yang digunakan dalam penelitian ini adalah metode penelitian deskriptif dengan teknik survey, yaitu dengan cara pengumpulan data melalui wawancara dengan para stakeholders terkait sebagai responden. This research was conducted with the aim of knowing the types of seagrass and the condition of the seagrass beds in the waters near Mokupa Village, where the beach is between Mokupa Resort and Lotus Resort Manado. The method used is a quadrant transect (vertically to the shoreline), the coordinates point 1°24'53\" N 124°42'22\" E is transect 1, 1°24'54\" N 124°42'22\" E is transect 2, and 1°24'56.7\" N 124°42'23.5\" E is transect 3. In this study, 5 species of seagrass were found, namely: Syringodium isoetifolium, Cymodocea rotundata, Thalassia hemprichii, Enhalus acoroides, and Halophila ovalis. Cymodocea rotundata had the highest average seagrass cover per species, 15.349%, and Enhalus acoroides had the lowest average seagrass cover, 0.053%. The condition of the seagrass beds at the study site was included in the unhealthy category with a seagrass cover of 31.432%. Penelitian ini dilakukan dengan tujuan untuk mengetahui jenis-jenis lamun dan kondisi padang lamun di perairan sekitar Desa Mokupa, dimana pantai berada di antara Mokupa Resort dan Lotus Resort Manado. Penelitian ini mengikuti Pedoman Status Padang Lamun KEPMEN LH 200/2004. Metode yang digunakan yaitu transek kuadran (tegak lurus garis pantai), dengan titik koordinat Transek 1 1°24'53\" N 124°42'22\" E, Transek 2 1°24'54\" N 124°42'22\" E, Transek 3 1°24'56.7\" N 124°42'23.5\" E. Pada penelitian ini ditemukan 5 jenis lamun yaitu: Syringodium isoetifolium, Cymodocea rotundata, Thalassia hemprichii, Enhalus acoroides dan Halophila ovalis. For high FAD usefulness, the light used must be able to collect fish that are at great distances both horizontally and vertically. The purpose of this study was to obtain the composition of the fish caught and how much the fish was determined to be when the raft was without lights in the water and when the raft was equipped with lights in the water. The method used was an experiment using 2 (two) types of lights, namely electric surface lights and LED lights in water, and 3 (three) types of fishing gear, namely fishing rods, ring seines and nets). The types of fish caught were dominated by selar fish (Selaroides sp), moonfish (Lampris guttaus), Trevally fish (Caranx sp) and squid (Loligo sp). The fish's interest in the light was observed since the first 15 minutes the lights were turned on, and were around 4 to 6 meters and the longer they were getting closer to the lights for about 1 hour the fish would cluster around the raft for about 2-4 meters.Keywords: FAD, Underwater lamp, Manado Bay.AbstrakPerairan Teluk Manado merupakan salah satu kawasan perairan di Sulawesi Utara yang memiliki sumberdaya perikanan yang potensial, dengan usaha penangkapan ikan dengan alat bantu rakit yang dipasang lampu listrik permukaan. Untuk daya gunanya rumpon yang tinggi, maka cahaya yang digunakan harus mampu mengumpulkan ikan yang berada pada jarak yang jauh baik horisontal maupun vertikal. Tujuan penelitian ini adalah mendapatkan komposisi hasil tangkapan ikan dan berapa dalam ketetapan ikan saat rakit tanpa lampu dalam air dan saat rakit dilengkapi lampu dalam air Metode yang digunakan adalah experimen dengan menggunakan 2 (dua) tipe lampu yakni lampu listrik permukaan dan lampu LED dalam air, dan 3 (tiga) jenis alat tangkap yakni pancing, pukat cincin dan jaring). Jenis ikan yang tertangkap didominasi berturut-turut oleh ikan selar (Selaroides sp), ikan moonfish (Lampris guttaus), ikan bobara (Caranx sp) dan cumi-cumi (Loligo sp). It has an important function in the effort to absorb carbon levels through the photosynthesis process, which is better than other forest vegetation. The purpose of this study, namely to identify mangrove species, examine vegetation and determine the value of biomass and estimated carbon stock in Budo Village, Wori District, North Minahasa Regency, North Sulawesi. Data collection in this study used the line transect method with the transect position being on the left, middle, and right of the mangrove ecosystem of Budo Village, which was pulled along 100 meters per transect. Then the quadrants were measured 10 m x 10 m using raffia rope to limit each quadrant with an interval of 10 iddle, and right of the mangrove ecosystem of Budo Village, which was pulled along 100 meters per transect. Then the quadrants were measured 10 m x 10 m using raffia rope to limit each quadrant with an interval of 10 m, and the distance between transects is about 50 m. Based on the results of the study, we found four types of mangroves, namely: Rhizophora mucronata, Rhizophora apiculata, Sonneratia alba, and Bruguiera gymnorrhyza. The highest species density and relative density values were on transect 3 with a value of 900 trees/ha and 86.53% by Rhizophora apiculata. The species frequency and relative species frequency were highest in each transect owned by Rhizophora apiculata with values of 0.8 and 55.66%. The highest species cover was Rhizophora apiculata with a value of 2.56 cm/m and a relative species cover of 72.21%. The highest important value index was Rhizophora apiculata on transect 3 with a value of 214.31%. The highest biomass content was on transect 3 with a value of 592.27 tons/ha. Based on the research results, the CPUE value of large pelagics has decreased every year, where the CPUE value in 2017 was 25.42%, in 2018 it was 25.08%, in 2019 it was 17.71%, in 2020 it was 17.10%. , the last in 2021 was 14.66% and for small pelagics, the CPUE value decreased, where the CPUE value in 2017 was 21.70%, in 2018 it was 17.92%, in 2019 it was 17.19%, in 2018 it was 17.19%. in 2020 is 23.13%, and the last in 2021 is 20.04%. Utilization rates in large and small pelagics are in a state of concern as CPUE decreases yearly.Keywords: CPUE, production data, purse seine, utilization rate.AbstrakPenelitian ini bertujuan untuk menganalisis perkembangan lima tahun CPUE (catch per unit effort) dan mengetahui upaya penangkapan perikanan pukat cincin di PPP Tumumpa serta pengaruhnya terhadap CPUE secara periodik. Berdasarkan hasil penelitian nilai CPUE pelagis besar mengalami penurunan setiap tahun, dimana nilai CPUE pada tahun 2017 yaitu 25,42 %, pada tahun 2018 yaitu 25,08 %, pada tahun 2019 yaitu 17,71 %, pada tahun 2020 yaitu 17,10 %, terakhir pada tahun 2021 yaitu 14,66 % dan untuk pelagis kecil nilai CPUE mengalami penurunan, dimana nilai CPUE pada tahun 2017 yaitu 21,70 %, pada tahun 2018 yaitu 17,92 %, pada tahun 2019 yaitu 17,19 %, pada tahun 2020 yaitu 23,13 %, dan terakhir pada tahun 2021 yaitu 20,04 %. Tingkat pemanfaatan pada pelagis besar dan kecil berada dalam keadaan mengkhawatirkan karena CPUE mengalami penurunan setiap tahun.Kata Kunci: CPUE, data produksi, pukat cincin, tingkat pemanfaatan. Seagrass sampling was carried out using a random sampling method (randomly) with a quadratic transect drawn perpendicular to the shoreline. Thus, 6 species of seagrass were obtained, namely Enhalus acoroides, Halophila ovalis, Thalassodendron ciliatum, Thalassia hemprichii, Syringodium isoetifolium and Halodule pinifolia, with various aquatic environmental conditions. The results of the analysis of the status of the seagrass beds obtained that the seagrass cover value was 50.20% which was included in the \"moderate\" category, while the seagrass cover per species was Thalassia hemprichii species with the highest cover found was 30.08%, Enhalus acoroides was 21.49% , Halophila ovalis was 6.84%, Thalassodendron ciliatum was 17.39%, Syringodium isoetifolium was 12.31% and Halodule pinifolia was the least common type was 2.35%. The density value of Enhalus acoroides seagrass is 48.375 ind/m2, Halophila ovalis is 14.5 ind/m2, Thalassodendron ciliatum is 35,475 ind/m2, Thalassia hemprichii species with the highest density is 67.25 ind/m2, Syringodium isoetifolium is 27.875 ind/m2 and Halodule pinifolia species density is at least 5.25 ind/m2. Hasil dari analisis status padang lamun diperoleh nilai tutupan lamun adalah 50,20% termasuk ke dalam kategori “sedang”, sedangkan tutupan lamun per jenis yaitu Thalassia hemprichii jenis dengan tutupan yang tertinggi dijumpai adalah 30,08%, Enhalus acoroides adalah 21,49%, Halophila ovalis adalah 6,84%, Thalassodendron ciliatum adalah 17,39%, Syringodium isoetifolium adalah 12,31% dan Halodule pinifolia jenis yang paling sedikit ditemukan adalah 2,35%. Generally, the commodities targeted for fishing are types of reef fish and seagrass beds that have a high selling value, such as snapper, grouper, baronang, yellowtail, lobster, octopus, and others. Bubu fishing gear or traps called bebihe local people are one of the fishing gear used to catch demersal fish or reef fish. The success of the bubu fishing operation is greatly influenced by the construction of the bebihe fishing gear itself and the treatment of the bait used. The use of seagrass bait, allegedly can increase the catching ability of the bebihe. However, scientific information on the use of seagrass as a bait for bebihe for fishing is not yet available. Therefore, the purpose of this study is to determine the effectiveness of seagrass baits with the comparison of kitefish in the operation of bebihe; and identify the types of fish caught. This research was carried out in the waters of Beeng Laut island, South Central Tabukan District, Sangihe Islands Regency; is based on experimental methods. Six bebihe units were operated during nine trips to collect data; where three units use seagrass bait of the type Thalassia hemprichii, and the other three units use kite fish bait; and then the catch data is analyzed by a t-test. The catch of bebihe during the study amounted to 363 heads, as many as 345 were caught with seagrass baits, and 18 were caught with kite baits. Analysis of the t-test showed that at t count = 5.480 > t table 0.05;8 = 2.306; where this explains that the use of seagrass bait on bebihe, gives a noticeablely different catch compared to kitefish bait.Keywords: Bebihe, Seagrass, Bait, Seagrass Meadow Fish AbstrakUsaha perikanan bubu telah berkembang dengan baik di seluruh perairan Indonesia, terutama di wilayah pesisir yang memiliki habitat terumbu karang dan padang lamun. Umumnya komoditi yang menjadi target penangkapan adalah jenis ikan karang dan padang lamun yang memiliki nilai jual tinggi, seperti ikan kakap, kerapu, baronang, ekor kuning, lobster, gurita, dan lain-lain. Alat tangkap bubu atau perangkap yang oleh masyarakat lokal dinamakan bebihe adalah salah satu alat tangkap yang digunakan untuk menangkap ikan demersal atau ikan karang. Keberhasilan operasi penangkapan perikanan bebihe sangat dipengaruhi oleh konstruksi alat tangkap bebihe itu sendiri dan perlakuan umpan yang digunakan. Penggunaan umpan lamun, diduga dapat meningkatkan kemampuan tangkap dari bebihe. Namun informasi ilmiah tentang penggunaan lamun sebagai umpan bebihe untuk menangkap ikan belum tersedia.
