It is required to determine whether the most common plant miRNAs can interact with human genes and what consequences this interaction may have.","PeriodicalId":33060,"journal":{"name":"ExRNA","volume":"45 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-09-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"134994865","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

As such, the development of non-invasive tools to predict risk following AMI remains a clinical priority. MicroRNAs (miRNAs) are small-noncoding RNAs that regulate gene expression and have come to prominence as a novel class of circulating biomarkers in AMI. In this narrative review, we summarise the current evidence that links circulating miRNAs with parameters of LV remodelling and MACE following AMI, and investigate clinical challenges that must be overcome to facilitate the translation of miRNAs into clinical use.","PeriodicalId":33060,"journal":{"name":"ExRNA","volume":"16 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-09-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"135433187","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

These nanovesicles transport proteins, lipids, and nucleic acids, including microRNAs (miRNAs). miRNAs are small non-coding RNA molecules that regulate gene expression. Exosomal miRNAs (exo-miRNAs) modulate many pathways involved in tumor progression, growth, metastasis, and angiogenesis. Therefore, in this review we investigated the role of exosomal miRNAs in both solid and hematological tumors, with the purpose of highlighting their potential role as disease biomarkers. Methods: Published peer-reviewed papers used as references for this narrative review were selected from the PubMed database using “exosomes”, “microRNA” and “cancers” as keywords. The time frame adopted in the bibliographic research covers an 11-year period, from January 2012 to January 2023. Key Content and Findings: This review explores the role of exosomal miRNAs as disease biomarkers. An excursus on exosomes and miRNAs is initially made to then illustrate the mechanisms of miRNAs sorting in exosomes. Exo-miRNAs can communicate with cells and can modulate their actions. Their half-life is extended thanks to the protection provided by exosomes. The role of exosomal miRNAs in different types of solid and hematological tumors is summarized highlighting the potential of disease staging and progression. Conclusions: All manuscripts studied suggest that exosomal miRNAs can be used as diagnostic and prognostic biomarkers iwn cancer. The lack of appropriate and usable biomarkers for cancer diagnosis and prognosis makes exosomal miRNAs good candidates to provide non-invasive information to guide disease management in different types of cancer.","PeriodicalId":33060,"journal":{"name":"ExRNA","volume":"9 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"135447216","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Such biopsies contain various biomarker analytes, including extracellular RNA (exRNA) with the potential to reflect dynamic changes in the tumor. However, it is challenging to distinguish tumor-derived exRNA from normal RNA. To overcome this limitation, xenograft models serve as a practical tool. In a mouse engrafted with human tumor cells, human exRNA is by definition originating from the tumor, whereas murine exRNA is host-derived. To study treatment response by monitoring tumor-derived exRNA, xenograft models with a high release of tumor exRNA into the circulation are desirable. Methods: The aim of this study was to evaluate whether and to what extent the cell line, its engraftment site, or the tumor size influence the amount of tumoral exRNA detected in blood plasma. To that end, four different neuroblastoma cell lines were engrafted in nude mice, either subcutaneously in the flank or orthotopically in the adrenal gland. Tumor sizes were monitored by caliper measurements (subcutaneous grafts) or MRI scans (orthotopic grafts) and blood was collected via terminal cardiac puncture to evaluate the tumoral exRNA fraction. Results: We demonstrate that the tumoral exRNA levels are correlated with the size of the subcutaneous tumor grafts. These levels are also highly dependent on the engrafted cell line. Furthermore, orthotopic engraftment potentially results in superior levels of tumoral exRNA, likely because of higher vascularity of the tumor tissue. Conclusions: Factors as cell line, tumor size and injections site should carefully be considered when performing experiments to study circulating RNA biomarkers.","PeriodicalId":33060,"journal":{"name":"ExRNA","volume":"1 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"135447208","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
