MBC prognosis is known to be weak, frequent recurrent, and MBC patients have only a survival rate of about 5 years. One of the proteins that causes breast cancer metastasis is Human Epidermal Growth Factor 2 (HER2). Pentagamaboron-0 (PGB-0), a newly curcumin analogue performed cytotoxic effect on HER2-positive breast cancer cells but it is practically water-insoluble. The aims of this study are to determine anti-metastatic activity of a more soluble form of PGB-0 namely PGB-0 fructose complex (PGB-0-F) toward HER2 positive cancer (MCF-7/HER2) cells. PGB-0-F was obtained from Cancer Chemoprevention Research Centre Faculty of Pharmacy Universitas Gadjah Mada. Based on scratch wound healing assay result, PGB-0-F inhibited cell migration especially in combination with doxorubicin at the concentration 15 μM. Under gelatin zymography assay, PGB-0-F in combination with doxorubicin decreased Matrix Metalloproteinases 9 (MMP-9) expression compare to the doxorubicin. One strategy to target the cancerous cell is Boron Neutron Captured Therapy which showed high affinity toward cancer cells and reported to have anti-proliferative as well as antimetastatic activities. Cancer Chemoprevention Research Center Faculty of Pharmacy Universitas Gadjah Mada, has developed boron-containing substance namely pentagamaboronon-0 (PGB-0) which is known to exhibit anticancer activity towards breast cancer cell. The purposes of this research are focused to explore the anti-migratory activities of PGB-0-So against triple negative breast cancer cell. The MTT cytotoxicity assay of PGB-0-So against 4T1 breast cancer cell line were found to exert potential effect in dose-dependent manner with IC50 values of 39 μM. The study of cell migration inhibition using in vitro wound healing assays and gelatin zymography on highly metastasis breast cancer cell line 4T1, following the treatment of sub IC50 doses of PGB-0-So complex slightly inhibited cell migration through the inhibition of matrix metalloproteinase-9 expression. Liberica coffee was one of the most widely cultivated types of coffee in Indonesia, especially in Tanjung Jabung Barat Jambi Province. Liberica coffee known as the coffee with highest levels of caffeine if compared with Arabica and Robusta coffee but the research about Liberica coffee was still very little if compared to the other two types of coffee. In the previous research, caffeine shown to be increasing alanine aminotransferase enzyme. Otherwise, coffee also contain chlorogenic acid which is a potent antioxidant and play a role in the protection of liver. The aim of this study are to investigate the effect of dark roasted Liberica coffee consumption on liver histological appearance, macroscopic anatomy and physiological function (serum glutamic-pyruvic transaminase (SGPT) level). This was an experimental study with posttest only control group design, using 45 male white mice and divided into 4 groups: control (with no coffee treatment), P1, P2 and P3 which were given coffee treatment consecutively 8000 ppm; 10.000 ppm and 12.000 ppm for 28 days. The result showed that liver histological damage level associated with the extent coffee intake. Chloramphenicol is an antibiotic widely used,despite of its toxicity potentials. The enhancement of free radicals in the body could suppress antioxidant activity. Propolis of Trigona sp. has been known to contain very high amount of antioxidants. The enhanced serum glutamic oxaloacetic transaminase (SGOT) and alkaline phosphatase (ALP) levels in serum is used as marker of liver damage due to the increase of free radicals. The purpose of this study was to observe the effect of Trigona sp. propolis ethanolic extract on SGOT and ALP levels in rats (Rattus novergicus) pretreated by chloramphenicol to induce liver damage. Test animals used for this research were male rats aged 8-12 weeks divided into five treatment groups: negative controlgroup (normal), positive control group (induced by 400 mg/kgBW chloramphenicol), first therapy group, second therapy group, third therapy group induced by chloramphenicol with and propolis extract with the dose of 8 mg, 16 mg, and 24 mg, respectively. Chloramphenicol was injected subcutaneously for 14 days, whereas propolis extract were administered orally for 21 days. The level of SGOT and ALP was determined using spectophotometry. The results showed that propolis extract could reduce levels of SGOT and ALP. Dose of 24 mg/kg was the effective dose to decrease levels of SGOT and ALP significantly (p<0.01). Pentagamaboronon-0 Sorbitol (PGB-0-So) is an aquoeous formulation of curcumin analog, PGB-0, with sorbitol. This compound has been developed as an anti-cancer chemotherapeutic agent and a boron carrying pharmaceutical for boron neutron capture therapy (BNCT). The aims of this study are to investigate antimetastatic activities of PGB-0-So against HER2-overexpressed MCF-7 breast cancer (MCF-7/HER2) cells. The MTT cytotoxicity assay of PGB-0-So exhibited cytotoxic effect with an IC50 value of 35 μM. The testing of anti-migration activity using the scratch wound healing assay demonstrated that PGB-0-So inhibited the closure of the wound on MCF-7/HER2 cells compare to the control. Furthermore, PGB-0-So was able to suppress matrix metalloproteinase (MMP)-9 activities, based on the gelatin zymography assay. Pumpkins contain rich unsaturated fatty acids, phytoestrogens and vitamins E in their seeds that have potential pharmaceutical, nutraceutical, and cosmeceutical properties. Information regarding their nutritional components and therapeutic properties of pumpkin seeds has expanded dynamically in the recent years and this review focus on the three main components of pumpkin seeds that described before. Several types of unsaturated fatty acids are the dominant composition in pumpkin seeds which can play a role in the disease prevention and promote health. Pumpkin seeds also contain the important phytoestrogen compounds, i.e., secoisolariciresinol and lariciresinol that have estrogenic-like effect such as preventing hyperlipidemia and osteoporosis for menopausal women. Phytoestrogens in pumpkin seeds also could be related to a reduced hormone-dependent tumor. Pumpkin seeds are rich in vitamin E contents as an emerging free radical scavenger, antiaging and antioxidant such as a-tocopherol and g-tocopherol. Scientifically, the phytochemical content of the fingerroot rhizome showed some therapeutical effects such as antibacterial, anti-inflammatory, anti or pro-oxidant, and also anticancer. In this article, we summarize some studies especially about anticancer activity of fingerroot and its constituent coumpound. We found that fingerroot is capable of inhibiting various pathways of cell physiology processes. One potential pathway to be inhibited by fingerroot is Poly (ADP-ribose) polymerase (PARP) which has role in apoptotic induction. Boesenbergia pandurata known as medicinal plant possessing anticancer potential due to the cytototoxic and antimetastatic characteristic of its active compound. The aim of this study is to observe the inhibitory of migration of Boesenbergia pandurata ethanolic extract (BPEE) in combination with doxorubicin (Dox) on MCF-7/HER2 cells. BPEE was prepared by 96% ethanol maceration. Under MTT assay, BPEE decreased the cells viability with IC50 value of 23 ± 3.9 μg/mL. Lamellipodia and wound healing assay analysis showed that BPEE (5 μg/mL) and its combination with doxorubicin (10 nM) inhibited cells migration after 48 hours observation, while gelatin zymography analysis showed that this combination did not affect the expression of MMP2 and MMP9 proteins, but single treatment of BPEE was more able to decrease the expression of both MMP. The combination with those concentrations inhibited the cells migration but not with the cells viability. Thus, BPEE is potential to be developed as antimetastatic agent. The aim of this study is to examine the estrogenic effects of ethanolic extract of papaya peels (EEPP), on the development of mammae gland and the increasing of uterine weight. The in vivo test was performed with ovariectomy in Sprague Dawley female rats that caused the rats to be in an estrogen deficiency state. After 30 days of treatment, animals are sacrificed to take the uterus and mammae glands. Measurement of uterine weight and mammae gland are observed by hematoxylin-eosin staining method to know the lobulus development and AgNOR staining to determine the proliferation level of mammae gland epithelial cells. The test results showed that EEPP concentration of 500 and 1000 mg/kgBW were able to increase uterine weight and proliferation of mammae gland. One of which is to inhibit bone resorption by targeting osteoclastogenesis through inhibition of the Cathepsin K, p38 Mitogen-activated Protein Kinase (MAPK), Nuclear Factor of Activated T-cells c1 (NFATc1) and Nuclear Factor kB (NFkB). Besides p38 MAPK, the c-Jun N-terminal kinase (JNK) / stress-activated protein kinases (SAPK), another member of MAPK family, has been reported to play important roles in osteoclastogenesis. Hence, current study was undertaken in order to investigate mechanism of Caffeic Acid towards JNK/SAPK pathway. Tartrate Resistant Acid Phosphatase (TRAP) staining was performed on caffeic acid-treated and RANKL-TNFα-induced RAW-D cells. Western blot analysis was performed to detect JNK/SAPK and phosphorylated-JNK/SAPK. Protein bands were quantified and statistically analyzed. Treatment of 10 μg/mL Caffeic Acid inhibited 20 ng/mL RANKL and 1 ng/mL TNFα-induced RAW-D differentiation into TRAP+ osteoclast-like polynuclear cells. Induction of 20 ng/mL of RANKL and 1 ng/mL of TNFα for 0.2 or 1 hour, significantly increase phosphorylation of JNK/SAPK as compared with control. Treatment of 10 µg/mL Caffeic Acid significantly inhibited the 20 ng/mL of RANKL and 1 ng/mL of TNFα-induced phosphorylation of JNK/SAPK.
