Südüpak","doi":"10.35193/bseufbd.928182","DOIUrl":"https://doi.org/10.35193/bseufbd.928182","url":null,"abstract":"Mitogen-Activated Protein Kinases (MAPKs) function in signaling pathways as modules cascading between stimulus activated sensors and response mechanisms.ZmMAPK14, a group C final MAPK of this cascade was identified as a differentially expressed message in cDNA-AFLP studies of both susceptible and resistant genotypes, where a gradual induction was displayed in the resistant genotype while a clear repression occurred in the susceptible genotype in response to Puccinia sorghi (Ps) inoculation. RT-qPCR verification studies, however, did not reveal the same pattern of expression in that both displayed inductions at different levels. Ps inoculation induced a limited expression increase fluctuating between 1.5 and 2.5-fold in the susceptible genotype while a dramatic upregulation starting at 12 h with a 149-fold and gradually increasing to a maximum level of 477 folds at 72 h in resistant genotype was observed. To obtain further evidence about its role in plant defense, ZmMapk14 expression in response to applications of chitin, Salicylic Acid (SA) and H2O2 at six time points covering a 0-24 h interval were studied in both genotypes. All treatments induced ZmMapk14 expression in the resistant genotype significantly at different levels while the expressional changes in the susceptible were more complex and limited similar to the Ps inoculation levels in the susceptible. Overall, the results show that ZmMAPK14 display differential expression in resistant and susceptible genotypes in response to Ps inoculation and applied defense compounds, and it presumably functions in plant defense to Ps. In this context, urban water management in Merzifon district has been evaluated by examining water resources, drinking water transmission and distribution network, water subscriber data, wastewater system and storm water system. It has been determined that the drinking water resources in the district are quite limited, but there are difficulties in reaching the result in the studies on both increasing the drinking water capacity and renewing the drinking water network, while the demand for water is increasing day by day. It has been observed that if the necessary measures are not taken in the short-term period, a major drinking problem awaits the district, and this cannot be solved by the measures taken by the local government alone. It is an applied project work aimed at solving the problem. Literature equivalents of basic concepts, such as production, product complexity, factory layout, assembly lines, line balancing, and ergonomics were examined and related to the subjective application project topic. After analysing the customer expectation, the working steps are laid out step by step in a way to respond to it. In the research, the mathematical modelling difficulty of the operational structure that will respond to the increase in capacity and product variety has been revealed within the framework of sector-specific constraints. Because of this difficulty, the importance of industry-specific solutions based on experience and intuition has been emphasized. GİRİŞ Uluslararası Motorlu Araç Üreticileri Organizasyonu (OICA) verilerine göre araç satışları son on yılda ciddi bir artış göstererek yüz milyon sınırına yaklaşmıştır [1]. Bu artışlara paralel olarak Tablo 1’de gösterildiği gibi ticari araç satışlarında da yükselme gözlenmektedir. Global eğilime paralel şekilde, Otomobil Sanayi Derneği (OSD) verilerine göre 2019 yılında Türkiye’de 982.642’si binek otomobili olmak üzere 1.461.244 araç üretildi. Otomotiv toplam ihracatımız 31,2 milyon dolara ulaştı. Bu finansal değer otomotivin Türkiye ekonomisindeki önemini gözler önüne sermektedir. Yine OSD verilerine göre otomotiv, toplam ihracatımızın %15’ini oluşturmaktadır ve son on beş yıldır en büyük ihracat sektörümüzdür. Üretilen araçların %85 ihraç edilmektedir. GSYH’mizin %3’ü bu sektörden kaynaklanmaktadır. Türkiye özellikle hafif ticari araç üretiminde Avrupa’nın en büyüğü haline gelmiştir ve dünyanın en büyük 14ncü otomotiv üreticisi konumundadır [2].
