This method can promote drug therapy and overcome some traditional treatment challenges in different ways that are discussed at this paper. \"One-size-fits-all\", Large-scale production, and less patient and physician acceptability are some limitations that we will encounter in traditional therapy. Three-dimensional printing of pharmaceutical products, is a versatile technology that needs specific attention. droplet-based, extrusion-based, and laser-assisted 3D printers are three main techniques that can be used in this field. The limitations and advantages of this method are also discussed, highlighting potential innovative pathways towards the possibility of drug carriers’ usage in ink formulas. The administration pathway of drug-loaded composites is another important issue in drug treatment strategies that has been discussed here. Oral drug delivery as a convenient method of systemic drug administration with great patient preference, is introduced as a most prevalent pathway that has been studies about 3D printed medicines. MSCs show great promise in cancer therapy due to their unique features. MSC secrete various cytokines with multifunctional properties, although their roles are unclear. Methods: We have investigated the influence of secreted cytokines from MSCs on KG-1 cells as a cell model of acute myeloid leukemia (AML). For this purpose, following the culture and characterization of MSCs, a trans-well system was used for co-culturing MSCs and KG-1. To determine apoptosis induction Ki/Caspase-3 assay was conducted for cultured KG-1 alone and in co-culture with MSCs (10:1) on day 7. In the following step, the protein was isolated from both groups (control and experimental) and western blotting was done for investigating the BAX and BCL-2 proteins expression. Results: It was found that MSCs significantly enhanced caspase-3 activity in KG-1 cells (*P<0.05). Besides, A significant increase in protein expression of BAX was detected, while BCL-2 displayed a dramatic reduction (*P<0.01). Corticosteroids may down-regulate wound healing. This work aims to develop and characterize nanofiber eluting dexamethasone and minoxidil and investigate their effect on wound healing in a rat model. Method: Minoxidil and dexamethasone-loaded wound dressing were developed and characterized with SEM and FT-IR. In vitro, drug release studies were performed for 12 days. To model the wound healing activity of the developed formulations, excisional wounds were created on the dorsal section of male rats. Results: Histological results of Hematoxylin & Eosin and Masson’s trichrome staining indicate wound healing suppression in the dexamethasone-treated group and good healing activity in the minoxidil-treated group. Here we identified that the application of topical minoxidil can be effective for wound healing probably driven by the anagen hair growth while dexamethasone suppresses collagen production and prevents scar formation. The telomere is a noncoding DNA sequence including \"TTAGGG\" at the ends of chromosomes of vertebrates. The stability of telomere length is an important factor as a survival signal for cells and cancer prevention. Telomerase is a multi-subunit DNA polymerase that plays a crucial role in maintaining the telomere length, which is critical for the age-related pathogenesis of breast neoplasm. Some regulatory factors interfere with telomerase activity and therefore promote breast tumorigenesis. High telomerase activity and restoring telomere lengths are determined as key factors in progressing tumors to advanced stages of malignancies, which are highly estrogen-dependent in breast carcinogenesis. Melatonin is a hormone-like substance secreted by the pineal gland and has been reported to downregulate telomerase. It may therefore control telomere length in cancer cells. Certain malignancy-related biological pathways have recently been linked to telomere length, and this review provides new insights regarding the effects of melatonin on telomere length by reviewing the anticarcinogenic mechanisms underlying melatonin in relation to telomerase activity in breast carcinogenesis. Delay in wound healing owing to high levels of oxidative stress is a major difficulty in various metabolic illnesses, including diabetes mellitus. Several therapeutic wound dressing materials and methods, such as hyperbaric oxygen treatment and negative pressure wound therapy, have been developed to speed up wound healing and restore cellular homeostasis. A significant advance has been made in locating transcriptional regulators involved in wound healing. The redox sensitive transcription factor nuclear factor erythroid 2-related factor 2 (Nrf2) is the major regulator of antioxidant defense regulation, inducing the expression of cyto-protective genes and increasing the generation of antioxidants that scavenge free radicals.Activators of Nrf2 have been shown to reduce oxidative stress and improve wound healing in a variety of pathophysiological situations, including diabetes and its consequences such as diabetic foot ulcers, chronic kidney disease, and diabetic nephropathy. Several therapeutic chemicals have been discovered to alleviate oxidative stress and consequently increase cell proliferation. Angiogenesis results in tissue healing through activating the transcription factor Nrf2. Studies have shown that epithelial ovarian cancer initially responds to platinum-based therapy, however, the recurrent type is often resistant to treatment and is associated with high mortality. Fucoxanthin, a natural component found in marine algae, possesses various pharmacologic properties. This study evaluated the cytotoxicity and resistance reversal activity of fucoxanthin on multidrug resistance-associated protein 2 (MRP2)-overexpressing, cisplatin-resistant ovarian cancer cells (A2780RCIS) and their parental cells (A2780). Methods: Cell viability was evaluated in the presence of different concentrations of fucoxanthin or cisplatin or fucoxanthin/cisplatin combination using the MTT assay. Propidium iodide staining and sub-G1 analysis were used to evaluate fucoxanthin potential for cell cycle modification and apoptosis induction in cancer cell lines. Results: The results showed that fucoxanthin was able to cause similar cell toxicity in both cell lines via apoptosis induction. Co-treatment of cells with cisplatin (3.125 to 100 µM) and nontoxic concentrations of fucoxanthin (1 and 2.5 µM) did not reverse resistance to cisplatin in A2780RCIS cells. Copper complexes are highly active in biological applications and have high bioactivity. Because of the above description and applications, in the present work, synthesis and characterization of pyrazolyl borate derivative ligands and their Cu(II) complex were reported. The structure of the synthesized complex was determined by X-ray crystallography. In addition, the antimicrobial activity of the synthesized compounds along with the molecular docking of them was investigated. Methods: N-donor pyrazolyl borate derivative ligands abbreviated as K[HB(PzMe2)3] and K[H2B(PzMe2)2] and their Cu(II) complex were synthesized and characterized. The synthesized ligands and complex were evaluated for antibacterial activities against the gram-positive (B. subtilis) and the gram-negative (S. enterica) bacteria. Also, their molecular docking with B. subtilis SMC head domain (PDB ID: 5H67) as the possible targets was investigated. Results: The in vitro and in silico results showed, among the investigated compounds, complex [Cu(H2B(PzMe2)2)(HB(PzMe2)3)] indicated the highest antibacterial activity. Also, the Statistical analysis showed that the difference between the obtained data was significant. Conclusion: We have synthesized N-donor pyrazolyl borate derivatives and their copper (II) complex. Single X-ray results indicated the Cu(II) complex adopted an N5 environment around the metal center with a distorted square pyramidal geometry. The obtained binding energy of molecular docking studies is in direct correlation with the in vitro antibacterial studies. In Persian medicine, Alkanna orientalis (Boraginaceae) has been used for centuries as a naturally derived remedy for managing lesions. A cross-sectional pilot clinical trial was conducted to assess the wound healing effect of an ointment made of chloroform extract of roots of A. orientalis (CERAO). Methods: Sixty patients (36 men and 24 women) diagnosed with bedsore staging 1-2 entered the study for one year. They were divided into two groups of control and treatment with equal proportions. The control group received conventional treatment from the hospital, including irrigation serum, mupirocin, phenytoin ointments, and gauze dressing. After rinsing and cleansing with normal saline, in the intervention group, patients received a thin layer of CERAO once daily for four weeks. Clinical outcomes were measured at weeks 2 and 4. Results: Recovery assessment was carried out by measuring wound area, days of epithelia formation, and complete wound closure. The difference between the two groups was statistically significant (P-value <0.05) in terms of the mentioned criteria. The recovery percentage was 26.7% and 60% for the control and treatment groups, respectively. In the control group, 16.7% of the study population experienced the development of wounds, while in the intervention group, wound progression was not observed. Ostadrahimi","doi":"10.34172/ps.2022.40","DOIUrl":"https://doi.org/10.34172/ps.2022.40","url":null,"abstract":"Background: Clinical studies, investigating the effect of β-Alanine (BA) supplementation on recovery biomarkers in physically active individuals, have generated inconsistent results. This systematic review and meta-analysis study aimed to clarify the clinically relevant dietary effects of BA supplementation. Methods: A comprehensive search was done in the electronic databases of Scopus, PubMed, ISI Web of Science and Embase from inception to 2020. Meta-analysis was done using the random-effects model. Pooled effect size was evaluated using standard mean difference (SMD) and 95% confidence intervals (CI). Heterogeneity of between-study was evaluated according to Cochran’s Q test and I2. Subgroup analysis was conducted to identify the potential sources of heterogeneity. Results: Overall, 32 studies were included in the current study. The results suggested that BA supplementation increases carnosine level significantly (SMD: 0.22mmol/L, 95%CI: -0.17,0.61, P=0.27) but no effect was shown about lactate, fatigue, VO2, PH and bicarbonate (HCO3-) (P>0.05). Subgroup analysis revealed a significant association of VO2, carnosine and fatigue with supplementation dosage, gender and duration of administration respectively. Conclusion: BA supplementation emerged its beneficial effects on enhancing carnosine level which highlights its ergogenic effects. In contrast, no significant effects had been shown in term of fatigue delay and blood levels of lactate, HCO3-, PH, and VO2 value.
