The five began therapy with insulin produced by bacteria into which the human insulin gene had been introduced. The recombinant insulin (also known as biosynthetic insulin), which is being produced by Eli Lilly and Co, had previously been administered to healthy human volunteers in the United States, Great Britain, Greece, and West Germany and found to be free of toxic effects. All carbohydrate loads were calculated to contain 50 g of glucose. The data demonstrate that 1) dextrose and potato elicited similar plasma glucose responses whereas rice, and bread elicited lower responses with corn intermediate; 2) dextrose and potato elicited similar plasma insulin responses whereas rice gave lower responses, with bread and corn intermediate; 3) all of the carbohydrate loads suppressed plasma glucagon with dextrose causing the greatest suppression. Combined estrogen/ progestogen pills were associated with an increased risk of thromboembolic disease . Thromboembolic complications included cerebral ischemia, myocardial infarction, and vein thrombosis. In contrast, the progestogen-only pill was found to be successful and associated with only menstrual irregular ities. Intrauterine devices were assoc iated with a high failure rate (11 of 30 became pregnant during the first year) . They concluded that the combined estrogen/progestogen pill and intrauterine devices were potentially dangerous and/or unacceptable forms of contraception and the progestogenonly pill was the preferred method of contraception in insulin-dependent diabetic women . All patients were found to have substantial degrees of depression during psychiatric interview and by Kupfer-Detre test scores (8.1 +/- 0.6, as compared with control values of 4.0 to 4.3 +/- 0.2). Treatment with imipramine hydrochloride or amitriptyline hydrochloride resulted in complete remission of lower extremity pains in all patients in 10 +/- 2 weeks, with concomitant relief of depression and return of depression test scores to 3.8. There were twice as many women as men. One hundred twenty-nine patients had spells of light-headedness, shakiness, diaphoresis, weakness, and fatigue. Hypoglycemic symptoms occurring during the test were not related to level of plasma glucose nadir or to rate of descent of glucose level. Hypoglycemia was not found when glucose levels were measured during occurrence of spontaneous symptoms in 86 patients. MMPI scores were significantly different from those of general medical patients. Both men and women evinced a conversion V profile.
