El-beltagy","doi":"10.21608/mjab.2020.110256","DOIUrl":"https://doi.org/10.21608/mjab.2020.110256","url":null,"abstract":", ABSTRACT: Twelve dissimilar plant associated bacterial colonies were isolated from five different tomato seedlings, the Non Ribosomal Lipopeptides genes were detected by degenerated primers technique in the twelve tomato endophytic isolates, only the long rode bacilli endophytic strains were found to harbour the lipopeptides synthetases genes, these three strains were given laboratory names of (BMG100, BMG101 and BMG102) for further pursuit. Partial sequencing of 16s RNA gene and protein profile analysis for the three bacilli isolates showed that all isolates belonged to Bacillus subtilis. The lipopeptides production by HPLC analysis revealed that all the endophytic isolates from tomato stems had the ability to produce lipopeptides except the third isolate named B. subtilis BMG102 showed no production. different levels of volumetric oxygen transfer coefficient (K L a) were used as cursors for lipopeptides production. The maximum concentration of surfactin type was about 1431, 529 and 437 mg.L -1 , the plipastitin (fengycin type ) was 360 198 mg.L -1 and zero while the Mycosubtilin (iturin type) was zero, 228 and 414 mg.L -1 for the strains B. subtilis ATCC21332, BMG100 and BMG101, respectively. Surfactin production is visibly optimal at high oxygen transfer (k L a) and showed more than 0.07 S -1 , while the optimum production of the fengycin and iturin could be observed at mild values of O 2 transfer (k L a) which equal 0.01 and 0.015 S -1 respectively. The antibacterial - antifungal effects of the three B.subtilis extracts collected during lipopeptides productivity regimes showed inhibition activity against Pseudomonas syringae, Ralstonia solanacearum and Erwinia amylovora, as will as the antifungal activity against Aspergillus niger, Phanerochaete chrysosporium and Alternaria alternata. These results indicate potential use of B.subtilis strains as biocontrol agents, especially when considering commercial implementation. The fruits were applied by the recommended doses of chlorpyrifos. Residues of pesticides in fruits were determined used liquid chromatography. The results showed that the residual chlorpyrifos decreased gradually due to time factor and the process of the fruit washing with water had a clear effect in reducing the concentration of the presence of chlorpyrifos in the applied fruits. El-beltagy","doi":"10.21608/mjab.2020.110273","DOIUrl":"https://doi.org/10.21608/mjab.2020.110273","url":null,"abstract":", , ABSTRACT : Samples collected from patient stool during their course of investigation at hospitals ministry of health, Egypt as well as solid and liquid food samples collected from local markets were examined for presence of Salmonella spp. Isolation and serological identification showed that 92.5% of collected samples contained Salmonella spp belonged to six serovars; Salmonella , enteritidis, Newport, derby, senftenberg and Virchow. These six serovars were checked by PCR for presence of three genes involved in virulence and toxin production, invA, avrA and stn. The gene invA was detected in all serovars, while avrA and stn genes were found in only 4 serovars. In this regard, S.typhimurium was found to harbour the three tested genes. When Salmonella typhimurium grew on normal synthetic medium and natural animal cells (Red blood; RBC and murine epithelial cells; IEC-6) and incubated separately at different temperatures 5, 25, 35 and 40 C for 60 h., they showed regular increase in growth rate with increasing temperature. Also, total RNA concentrations increased up to 24 h. then slightly decreased at each examined temperature specially at 25, 35 and 40 C, but the highest RNA concentration was observed at 40 C. indicating its preparedness in the form of metabolic and cellular activities to cope with temperature stress. Twenty male albino rats were used over 30 days period. The animals were divided into (4) groups, wherein groups number (1) represent control which were fed basal diet, while group number (2) was received high fat diet to serve as hyperlipidemic group. Other two groups allowed to feed high fat diet supplemented with jamun seeds and leaves, group number (3) treated simultaneously with jamun seeds, fourth group treated with jamun leaves. At the end of the experimental period, blood samples were collected to determine lipid profile, liver functions and determine the kidney functions include urea nitrogen and creatinine. From the obtained results we concluded that group of rats fed on high fat diet were considered as a major risk factor for hyperlipidemia disease. Our results could be summarized that jamun seeds and jamun leaves were considered the best for causing a reduction of TC, TG and LDL. Also, liver functions has been improved and there were significant reduction GOT, GPT and ALP activity than that of positive control group. Anise seeds contains: total carbohydrates 51.4 %, crude protein 18.1 %, total lipids 13.7 % and total ash 14.9 % in dry sample. While mint leaves contains contains total carbohydrates 55.7 %, crude protein 19.8 %, total lipids 4.9 %, and total ash 18.6 % in dry sample. Total phenolic in anise seeds were 216.6-237.8 mg/100g in aqueous and ethanolic extracts, respectively and total flavonoids were 148.1-156.4 mg/100g in aqueous and ethanolic extracts, respectively comparing with 234.3-278.9 mg/100g in aqueous and ethanolic extracts, respectively and 157.5-186.3 mg/100g in aqueous and ethanolic extracts, respectively of total phenolic compounds and flavonoids in mint leaves. HPLC results showed that anise seeds were found to contain 19 of phenolic compounds, amoung them catechein, cinnamic, ellagic, gallic, chlorogenic, ferulic and catechol were the major active in anise seeds, while mint leaves contained rosmaric, cinnamic, gallic, ferulic, and savianolic as the major of phenolic compounds.
