PHENETIC ANALYSES OF TENGKAWANG (SHOREA SPP., DIPTEROCARPACEAE) Tengkawang(娑罗双树科)的表型分析

Yulita","doi":"10.14203/bkr.v19i1.120","DOIUrl":"https://doi.org/10.14203/bkr.v19i1.120","url":null,"abstract":"Tengkawang is referred to a group of approximately 16 species of Shorea producing illipe butter from their kernels.  The name Tengkawang mostly referred to the Bornean Illipe Nut, even though they are not restrictly distributed in Borneo.  Nine of the 16 species belonging to Shorea Section Pachycarpae , two species of Section Brachypterae , three species of Section Mutica and two species of Section Shorea . Recent phylogenetic studies based on molecular markers considered Section Pachycarpae tend to form a monophyletic group.  This present study was aimed to investigate the nature of groupings within Tengkawang and their relatedness based on phenotypic traits.  Thirteen species of Shorea and one species of Hopea as an outgroup were selected to perform a phenetic analysis. An UPGMA dendrogram of Tengkawang was obtained using average taxonomic distance.  Cluster analysis indicated that genetic distance (Dist coefficient) varied from 0.92 to 0.24.  Two main groups were formed, first group contained exclusively Shorea Section Pachycarpae and second group consisted of the remaining species from various Sections  The principal Component Analysis (PCA) was also performed and showed the relative position of 13 of Tengkawang in two-dimension.  The PCA has also identified 27 morphological traits having important roles in the grouping of Shorea section Pachycarpae . Induksi poliploidi dilakukan dengan meneteskan kolkisin pada pucuk bibit P. amabilis .  Tujuan penelitian ini adalah untuk mendapatkan konsentrasi kolkisin yang efektif untuk induksi poliploidi bibit P. amabilis dan  menghasilkan bibit P. amabilis poliploid. Percobaan disusun dalam rancangan kelompok lengkap teracak dengan satu faktor, yaitu konsentrasi kolkisin. Pucuk bibit P. amabilis ditetesi 0,01 ml kolkisin (0, 1000, 2000, 3000, 4000, dan 5000 mg L -1 ). Hasil percobaan menunjukkan bahwa peningkatan konsentrasi kolkisin dari 1000 sampai 5000 mg L -1 tidak berpengaruh nyata terhadap persentase hidup dan pertumbuhan bibit pada 24 minggu setelah perlakuan (24 MSP). Bibit P. amabilis poliploid dapat dihasilkan pada penetesan kolkisin 1000, 3000, 4000, dan 5000 mg L -1 dengan konsentrasi kolkisin paling efektif adalah 5000 mg L -1 . Hal ini terjadi juga pada usaha konservasi tumbuhan secara ex situ . Proses menentukan jenis tumbuhan dan lokasi pengoleksian yang tepat selalu menjadi permasalahan yang dihadapi sebelum kegiatan pengoleksian dilaksanakan. Hal ini karena penentuan keputusan perlu mempertimbangkan berbagai hal seperti status konservasi, ketersediaan dana, ketersediaan informasi, waktu,  dan resiko keberhasilan tumbuh. Dalam tulisan ini akan dijelaskan kerangka pengambilan keputusan untuk  menentukan prioritas spesies tumbuhan yang akan dijadikan koleksi ex situ . Tiga puluh spesies Rhododendron asli Indonesia yang terancam punah digunakan sebagai contoh. Metode yang digunakan adalah skoring terhadap 11 kriteria. Kriteria tersebut mencakup status spesies (meliputi: status konservasi, status keberhasilan introduksi ex situ , representasi kelompok taksa unik); status lokasi (status dalam area Biodiversity Hotspot dan Global 200 ecoregion, serta perlindungan legal habitat); kemudahan propagasi (meliputi: bentuk hidup, ketinggian habitat, dan jarak antara lokasi dan lembaga ex situ ); efektivitas (jumlah spesies kongenerik simpatrik yang terancam kepunahan); dan biaya pengoleksian. Interpretasi hasil skoring dilakukan dengan sistem peringkat. Flushing yaitu pertumbuhan tunas/ flush secara serempak. Tujuan penelitian ini adalah mengamati pertumbuhan tunas daun dalam merespon intensitas cahaya dengan posisi tumbuh berbeda dan faktor lingkungan yang mempengaruhinya. Metode yang digunakan adalah deskriptif dengan pengambilan purposive sampling, flush dibedakan posisi tumbuhnya yaitu terminal dan aksilar dalam kondisi terpapar dan ternaungi. Laju tumbuh Amherstia nobilis Wall. dan Brownea capitella Jacq. di daerah terpapar cahaya lebih cepat dibandingkan daerah ternaungi. Laju pertumbuhan diiringi dengan perubahan warna. Perubahan warna pada daun  A. nobilis berlangsung selama dua bulan lebih cepat dari B. capitella . Hingga tahun 2010, empat kebun raya yang dikelola oleh LIPI hanya mampu mengkonservasi sekitar 21% dari seluruh tumbuhan terancam Indonesia. Oleh karena itu, diperlukan pengembangan Kebun Raya Daerah (KRD) untuk mengkonservasi tumbuhan pada tiap daerah di Indonesia. Makalah ini bertujuan untuk untuk mengetahui capaian KRD dalam konservasi tumbuhan dan menentukan strategi pengelolaan koleksi tumbuhan tiap daerah. Pada akhir 2012, KRI yang didukung KRD telah berhasil mengoleksi 24% dari tumbuhan terancam Indonesia versi IUCN Red List 2013 , dan berhasil membudidayakan 25% termasuk koleksi pembibitan. Beberapa strategi untuk pengembangan koleksi KRD antara lain: penguatan sistem data base koleksi; pengembangan koleksi tumbuhan di setiap KRD dengan mengacu pada IUCN Red List ; dan penentuan spesies prioritas untuk konservasi terkait dengan kebijakan dan kondisi alam di Indonesia. Those information based on Ristoja 2012 research. The objective of Ristoja was to provide a database of local ethnomedicine knowledge, herbal formula and medicinal plant in Indonesia. Inter Simple Sequence Repeats (ISSR) markers were used to assess the genetic diversity of Dringo from 20 selected ethnic groups in Indonesia. Ten selected ISSR primers generated 82 amplified fragments with 51,2% were polymorphic. Dice coefficient was used to calculate similarity index and UPGMA was used to construct a dendogram. The genetic similarity index among accessions ranged from 76,7-100 % thus indicating that low level of genetic diversity in dringo. Seiring dengan program pemerintah untuk melakukan diversifikasi pangan, tanaman minor yang potensial sebagai sumber karbohidrat dapat dipopulerkan kembali dengan tujuan sebagai upaya konservasi tanaman tersebut. Terkait hal tersebut, kentang hitam ( Plectranthus rotundifolius ) dapat menjadi sumber pangan alternatif bagi masyarakat yang menempati daerah kering. Kentang hitam tidak hanya berperan sebagai sumber karbohidrat dengan indeks glikemik rendah, tetapi juga kaya vitamin dan sejumlah mineral penting. Informasi mengenai teknik budidaya menjadi hal penting yang perlu dipersiapkan dalam rangka pengembangannya. Penelitian ini dilakukan untuk menjawab pertanyaan mengenai teknik budidaya yang perlu dilakukan untuk meningkatkan kualitas hasil panen kentang hitam di pasaran. Penelitian dirancang berdasarkan Rancangan Acak Kelompok (RAK) dua faktor, yaitu aksesi dan teknik budidaya. Empat aksesi kentang hitam yang digunakan: Nganjuk, Sangian, klon 6G dan O3; dan empat jenis teknik budidaya: bumbun, pangkas, jerami, and pengangkatan tajuk) Setiap perlakuan terdiri dari empat ulangan, dengan tiga tanaman/ulangan. Pengamatan dilakukan pada parameter pertumbuhan dan hasil. Hasil penelitian  menunjukkan aksesi kentang hitam yang berbeda memerlukan teknik budidaya berbeda untuk meningkatkan hasil dan ukuran umbi. Both species are drought resistant and have very high nutritional and medicinal properties. Detection of genetic diversity is of great value for the improvement of nutritional and medicinal value of these plants. The aim of the present study was to characterize a new biotype Moringa observed in Al Bahah Region, Saudi Arabia. We used 11 RAPD and 15 ISSR primers to characterize and compare the new biotype with M. peregrina and M. oleifera . Level of polymorphism generated by each marker was calculated. We also calculate Nei and Li’s coefficient to measure the genetic distance between the studied species. Level of polymorphism generated by RAPD and ISSR was 46% and 57%, respectively. RAPD and ISSR primers revealed that the new biotype shared 55 amplicons (45.08%) with both M. peregrina and M. oleifera , 28 amplicons with M. peregrina (22.95%), 21 amplicons (17.21%) with M. oleifera , and displayed 18 unshared amplicons (14.75%). Based on RAPD data, genetic distance between M. oleifera and M. peregrina was 0.32, whereas genetic distance between the new biotype and M. oleifera and M. peregrina was 0.21 and 0.29, respectively. For ISSR data, genetic distance between M. oleifera and M. peregrina was 0.5, whereas genetic distance between the new biotype and M. oleifera and M. peregrina was 0.36 and 0.34, respectively.
