Breeding conventional methods for generating of genetic variability are of low efficiency. We showed that treatment of seeds of rice(Hashemi cultivar) with 0.8% EMS for 8 h caused visible phenotypic variations on M2 rice mutant genotypes including flowering date, plant height, number of fertile tiller, panicle length, number of filled and unfilled grains per panicle, grain width and length, 100 grain weight and grain yield. The phenotypic variation coefficients of most traits found to be more than the genetic variation coefficients. The number of filled grains per panicle and seed length had the highest and lowest general heritability, respectivly. The seed yield had also high heritability. Analysis of correlation between different characteristics in the mutant genotypes showed that the number of fertile tillers and the number of unfilled grains per panicle had positive correlation with yield. Also, grain yield exhibited positive and significant correlation with panicle length, number of tillers and number of filled grains at genotypic level. In multiple regression analysis by stepwise method, number of tillers, number of filled grains per panicle, 100-grain weight, and grain width entered into the model, respectively, that explained 96 percent of grain yield variations. Results of grain yield and its components path coefficient analysis showed that the number of tiller had the highest direct effect (0.77) through than other traits on grain yield and, therefore it can be considered as major trait in grain yield improvement in rice. Also, based on results of this research and by using optimal selection index, mutant genotypes EM 18-17-5 and EM 15-14-1 were selected as superior mutant genotypes. Asghari","doi":"10.29252/pgr.6.2.157","DOIUrl":"https://doi.org/10.29252/pgr.6.2.157","url":null,"abstract":"To investigate the effect of exogenous brassinosteroid application on grain yield, catalase, chlorophyll content, membrane mtability index and gene expression of some genes involving in brassinosteroid signaling pathway ( BES1 and BRI1 ) under drought stress, a split-split plot on randomized complete block design with three replications was conducted at the experimental field of Seed and Plant Improvement Institute, Karaj, Iran in 2019. The main factor was two irrigation treatments (normal irrigation and water holding after 50% flowering stage), the subplots were four concentrations of brassinosteroid (0, 0.25, 0.625 and 1 mg/l) and seven genotypes (Mehregan, Paris, 2858, 3505, 3737, 4228 and 4056) were considered as sub-sub plots. Samples were taken at 30 days after 50% flowering stage (zadoks 89) from flag leaves. The results showed that drought stress significantly reduced grain yield, chlorophyll content, membrane stability index and increased catalase in all genotypes. Genotype 4228 was identified as the most tolerant genotype among unknown wheat genotypes based on grian yield, chlorophyll content, membrane stability index and catalase. Also, the result revealed that applied epibrassinolide could reduce the destructive effects of drought stress on wheat thus grain yield was enhanced under drought stress in all genotypes by increasing the aforementioned traits. Forethermore, grain yield was increased by rising the epibrasinolide concentration. The aim of this experiment was to investigate the genetic diversity of tall fescue ( Festuca arundinacea ) using agronomic traits such as plant height, spring growth score, days to flowering, days to pollination, flag leaf length and width, panicle length, weight of 100 seed and seed yield per plant. The experiment was carried out in a randomized complete block design with three replications. Based on the results, the ecotypes were significantly different for all of the evaluated traits with the exception of 100 seed weight. The highest phenotypic variation coefficient (58.66), genetic variation coeficient (58.32) and heritability (0.97) were related to plant height. The correlation between traits showed that ecotypes with higher height, panicle length and flag leaf height and spring growth score have higher seed yield. The results of stepwise regression analysis showed that in the regression model for seed yield per plant, the panicle length, alone, justified 60.95% of the variation. Cluster analysis grouped 25 ecotypes into three groups. Analysis of variance of agronomic traits as well as yield and related traits showed that grouping caused mean squares between groups in all traits (except flag leaf width). The molecular markers are the tools that have provided a new perspective for plant breeding advancements. This article has reviewed the various advantages and uses of molecular markers and the utilization of the high potential of natural polymorphisms within communities, combined with the abilities of conventional plant breeding methods. The marker attributes are not subjected to environmental influences, and their high frequency number and high structural diversity are as part of their benefits in identifying identities, determining the genetic diversity of species and studying relationship between populations. They may aid in discovering more information about protecting and maintaining genetic stock collections, identifying varieties, determining genes with chromosomal location and the number of genes controlling traits. Genome sequencing, the preparation of physical and genetic maps and genomic fingerprinting of plants are some of the other applications of this tool in plant breeding. The high efficiency of selection with the help of markers in selection of genotypes has been emphasized as the parent of crosses and selection with the help of a marker in breeding programs and genomic selection. New technologies offer new opportunities to shape genetic variation in the improvement of specific plant breeding programs. Nowadays, development of next-generation sequencing technology, genome sequencing and high throughput approaches for markers have facilitated EST-derived simple sequence repeat (EST-SSR) marker development as well as single nucleotide polymorphism (SNP) marker.
