CDDP ototoxicity is caused by damage of the organ of Corti, spiral ganglion cells or lateral wall stria vascularis and spiral ligament . Mesenchymal stem cells MSCs were shown to differentiate into neurogenic and auditory hair cells in vitro. In this study, effect of MSCs in CDDP ototoxicity model of HEI-OC1 cochlear cells was evaluated. Method: The cochlear cells were exposed to 50 and 100 microM CDDP for 24, 48, 72 hours with and without MSCs as coculture. The viability of the cells was analyzed with trypan blue dye and the percentage of apoptosis with Annexin-V by flow cytometer. The differentiation of MSCs to immature cochlear cells were shown by Math1, Calretinin and Myosin IIa immunohistochemistry. Results: At 100 microM dose, CDDP caused cytotoxicity on cochlear cells predominantly via necrosis. In co-culture, MSCs decreased cochlear cell damage of CDDP. In co-culture the ratio of Math1 and calretinin positive cells were increased supporting the idea of differentiation of MSCs into immature hair cells. Conclusion: In this in vitro study, our data support that MSCs protects cochlear cells from CDDP cytotoxicity. MSC therapy might be a candidate cellular therapy approach to overcome CDDP ototoxicity. The mechanism seems to be via differentiation of MSCs into immature hair cells. Our next step is to plan in vivo nude mice neuroblastoma animal model comparing CDDP therapy with and without systemic MSC administration and check ototoxicity.","PeriodicalId":245385,"journal":{"name":"The Journal of Basic and Clinical Health Sciences","volume":"64 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2020-09-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"131487531","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

It was aimed to assess the frequency of falls and fractures in early period after stroke. Materials and Methods: This study which was conducted between 2011-2013, included 50 patients who were admitted to and hospitalized at the rehabilitation clinic between the first two weeks and six months since the date of the stroke, who had sitting balance or were suitable to undergo ambulation. The demographic and clinical characteristics, complications and previous histories of the patients were recorded and physical examinations were performed. The patients were called for follow-up at the sixth month and first year after discharge; the number of falls and presence, number and location of fractures associated with falls were queried. Results: 76% of the patients returned for follow-up at the sixth month;70% for follow-up at the first year. Fall rates were 2% during the clinical follow-up; 26.3% during the first sixth months; 25.7% between the 6 and 12 months. 1 of 35 patients had wrist fracture on the hemiplegic side during the first six months. The patients were divided into two groups as fallen and not fallen. Conclusion: Fall after stroke is common during acute period, rehabilitation phase and chronic period. Early rehabilitation after stroke may increase balance and upper limb motor functions leading to lower rates of falls and fractures. Progressed osteoporosis and weakened bones leading to increased risk of fractures may be observed with longer follow-up periods.","PeriodicalId":245385,"journal":{"name":"The Journal of Basic and Clinical Health Sciences","volume":"24 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"1900-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"127963070","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Type 2 diabetes mellitus (T2DM) associated with dysfunctions in glucose and lipid metabolisms is known as a metabolic disorder. The aim of this study was to investigate whether asprosin levels were changed in subjects with newly diagnosed T2DM (nT2DM) comparing to subjects with normal glucose tolerance (NGT) and to determine if asprosin levels were linked to metabolic parameters. Method: This case control study included 68 subjects with nT2DM and 68 NGT age-, body mass index (BMI)and gender-matched subjects. We used ELISA method for measuring the circulating asprosin levels. A 2-h 75-g oral glucose tolerance test was used for diagnosing of T2DM. Metabolic parameters of enrolled subjects were determined. Results: Circulating asprosin levels were elevated in nT2DM subjects when compared to controls. (6.75 ± 1.54 vs. 4.05 ± 1.28 ng/ml, P < 0.001). Moreover, asprosin levels were found to be higher in nT2DM subjects with metabolic syndrome comparing to those without metabolic syndrome. Correlation analysis revealed that asprosin levels positively correlated with insulin, fasting blood glucose, insulin resistance and triglycerides whereas it displayed negative correlation with HDL cholesterol in only nT2DM subjects. Linear regression analyses showed an independent association between asprosin and insulin resistance. According to the logistic regression analyses, the subjects with the highest asprosin levels increased the probability of nT2DM prevalence. Conclusions: Increased asprosin levels associated with insulin resistance and unfavorable outcomes of metabolic parameters in subjects with nT2DM.","PeriodicalId":245385,"journal":{"name":"The Journal of Basic and Clinical Health Sciences","volume":"2_OS 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"1900-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"133093164","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Therefore, individual-specific arrangements in the early period can prevent these diseases. This study was aimed to assess the suitability of school desk and chair for Turkish population with current anthropometric measurements and find out the impact of this on the same participants’ life quality. Method: The students of the School of Health Science (N=153) made up the sample for this study. 14 different anthropometric measurements were taken from the participants and they were required to fill in the WHOQOL-Bref Life Quality Scale for the last month in the study. Result: Statistically no significant difference was determined in regards of general and sub-parameters of life quality points such as physical and environmental points between sample female (n=81) and male (n=72) population (p>0.05). Some fit and unfit measurements for desk dimensions were determined when anthropometric measurements of desk dimensions were analyzed minimally and maximally (5–95%) in terms of gender. Conclusion: In this study musculoskeletal disorders owing to the long term use of ergonomically unsuitable desks by determining fit and unfit measurements in desk dimensions and the effect of this on life quality are explained. Also norm values for new furniture designs were determined by calculating anthropometric measurements that must be updated termly for the continuously changing population.","PeriodicalId":245385,"journal":{"name":"The Journal of Basic and Clinical Health Sciences","volume":"18 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"1900-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"132705756","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Drug-induced QT interval prolongation (DI-QTP) is closely associated with severe ventricular arrhythmias [especially torsade de pointes (TdP)] and sudden cardiac death. In particular, development of DI-QTP is generally associated with multiple risk factors. Cardiac and non-cardiac drugs may cause QT interval prolongation (QTP) and TdP. Most of the QT-prolonging drugs act by blocking the rapid component of the delayed rectifier potassium channel whereas a smaller number of drugs act by modifying Ca2+ and Na+ currents. In addition, pharmacokinetic drug interactions are among the reasons of DI-QTP. The corrected QT interval (QTc) according to heart rate by Bazett’s formula is the most commonly used. Genetic susceptibility is another important issue in predicting DI-QTP and TdP risk. Silent mutations and/or polymorphisms associated with cardiac ion channels may cause a risk for DI-QTP. Firstly, for treatment, drugs that cause QTP should be stopped rapidly, electrolyte abnormalities and other pathologies should be rapidly corrected. Intravenous magnesium sulphate, overdrive pacing, isoproterenol and plasma alkalinisation via sodium bicarbonate are the main useful treatments for DI-QTP and related TdP therapy. Class 1B antiarrhythmic drugs and intravenous potassium are thought to may be effective in TdP. The purpose of this article is to review the underlying mechanisms of QTP, risk factors and genetics of DI-QTP, how to measurement of QT interval and treatment of acquired LQTS. Key-words: Drugs, QT interval, QT prolongation and torsade de pointes. Sudden cardiac death which is caused by mostly (% 80–85) acute ventricular arrhythmias, is a common cause of mortality. Prolongation of ventricular repolarization is an important cause of ventricular arrhythmias (1). Long QT syndrome (LQTS) characterized by QT interval prolongation (QTP) which represents ventricular depolarization and repolarization, may occur as congenital form or acquired form which induced by drugs. Many drugs have QT-prolonging effects and they may cause severe ventricular arrhythmias. Drug-induced QT interval prolongation (DI-QTP) is more common than congenital QT prolongation and particularly important in people with multiple risk factors (1–4). The purpose of this article is to review the underlying mechanisms of QTP, risk factors and genetics of DI-QTP, how to measurement of QT interval and treatment of acquired LQTS. Drugs that have QT-prolonging effects Cardiovascular and non-cardiovascular many agents may cause QTP and severe ventricular arrhythmias [especially torsade de pointes (TdP) which is defined as ventricular polymorphic tachycardia] (4). Drugs that have been associated with QTP and TdP, are shown in Table 1 (1, 5–11). TdP is important in routine clinical practice because it may degenerate into ventricular fibrillation and drug-induced TdP incidence is n","PeriodicalId":245385,"journal":{"name":"The Journal of Basic and Clinical Health Sciences","volume":"183 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"1900-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"134240920","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Manual calipers are widely used for this purpose. But the measurements with manual calipers may be inaccurate. Contrarily, three-dimensional (3D) ultrasonographic measurements give reliable and accurate tumor volume calculation. We aim to; evaluate the accuracy of common four formulas given in the literature to estimate xenograft tumor volumes based on manual caliper measurements and offer a new coefficient for a better estimation of the tumor volumes. Patients and Methods: Detailed manual diameter measurements of xenograft tumors were in 14 nude mice performed using Vernier caliper. Tumor volumes were calculated using the suggested formulas in the literature based on manual measurements. 3D ultrasound volume measurements were performed on same xenograft tumors using high resolution Vevo 2100 imaging system. To propose a new coefficient; means of ratio between manual and ultrasound volume measurement values were used. Also, data set was divided into two subgroups as tumor volume under 800 mm3 and over 800 mm. New coefficients for each subgroup were defined. Results: Only with prolate ellipsoid formula there was no statistically significant difference between volume measurements with two methods (p=0,24). Our proposed formula (0,45 L*W*H) could estimate tumor volumes as good as prolate ellipsoid formula. Coefficient 0,35 and 0,81 in the same formula were found efficient in the selected subgroups. Conclusion: Using one common coefficient/formula for tumor volume estimation in any tumor size can be inaccurate. Appropriate coefficient should be chosen according to the dataset worked with.","PeriodicalId":245385,"journal":{"name":"The Journal of Basic and Clinical Health Sciences","volume":"83 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"1900-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"134477186","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Patients and Methods:This study was conducted within the framework of a national multicentre project. Patients with either stage IIIB or IV primary lung cancer diagnosed after April 2010were included in thisstudy. A classical confirmatory approach was used for both the reliability and validity analyses. Internal consistency was tested using Cronbach’s α value, and the validity analysis was performed using construct validity and clinical validity. Results:276 (92.3%) patients were male and the average age was 60.8 ± 9.4 years. The most frequently observed histological type was squamous cell carcinoma (36.8%), and 61.5% of these stage IV tumours. Cronbachs α values for the subscales ranged from 0.60 to 0.84. The majority of the FACT-L subscales revealed inter-scale correlation coefficients greater than 0.35. All sub-dimensions, except that of the social/family well-being scale, are able to significantly discriminate between stages IIIB and IV. Significantly lower scale scores were detected in patients with stage IV than stage IIIB. The comparative fit index was 0.917, and the root mean square error of approximation was 0.091. Dyspnoea, haemoptysis, chest pain, weight loss, anorexia, localised pain, and fever symptoms had a significant correlation with the FACT-L, trial outcome index, and lung cancer subscale. Conclusion:Turkish version of the psychometric properties of the original FACT-L scale is regarded as a valid and reliable tool and can be used safely in a clinical context when managing patients with lung cancer in Turkey.","PeriodicalId":245385,"journal":{"name":"The Journal of Basic and Clinical Health Sciences","volume":"47 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"1900-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"126059009","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Their quality of life, in general, is negatively affected. The aims of this study were to determine changes in employment status after diagnosis of CHD and sociodemographic, clinical and work-related factors affected. Methods: This is a retrospective cohort study. Participants were selected from the Manisa Public Health Directorate database; of Turkey. Inclusion criteria encompassed residence in inner Manisa Districts, employment at an income-generating job, and a diagnosis of CHD for the first time (n=41) by a doctor between 01.12.2012–01.12.2013. A cohort of without CHD (n=73) was also recruited for the study for comparison. Chi-square tests and logistic regression analyses were performed to exam the relationship between dependent and independent variables. Results: It was found that 76% of patients returned to work within a year after receiving a diagnosis of CHD. However, the lay-off rate was found to be significantly higher among employees with CHD than others (p<0.05). There was no relationship found between being laid off and having a comorbidity (p>0.05). In the evaluation of all study participants, blue-collar and private sector employees had significantly higher lay-off rates compared to white-collar and public sector employees. Conclusion: After a CHD diagnosis, 24% of CHD patients cannot return to work. People in a lower occupational class experience a higher rate of lay-off, which may make life economically worse for these individuals. Social life and working conditions should be taken into consideration when evaluating whether CHD patients should return to work.","PeriodicalId":245385,"journal":{"name":"The Journal of Basic and Clinical Health Sciences","volume":"42 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"1900-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"122872201","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
