{"title":"Effects of different extracts of Eugenia caryophyllata on pentylenetetrazole-induced seizures in mice.","authors":"Mahmoud Hosseini,&nbsp;Taha Jafarianheris,&nbsp;Navid Seddighi,&nbsp;Mohammad Parvaneh,&nbsp;Ahmad Ghorbani,&nbsp;Hamid Reza Sadeghnia,&nbsp;Hassan Rakhshandeh","doi":"10.3736/jcim20121222","DOIUrl":"https://doi.org/10.3736/jcim20121222","url":null,"abstract":"<p><strong>Objective: </strong>To investigate the possible anticonvulsant effect of different extracts of Eugenia caryophyllata (clove) on pentylenetetrazole (PTZ)-induced seizures in mice.</p><p><strong>Methods: </strong>The animals were divided into saline, 50, 100, 250 and 500 mg/kg of aqueous extract, 50, 100, 250 and 500 mg/kg of ethanolic extract, and 50, 100, 250 and 500 mg/kg of chloroformic extract of clove groups. The extracts or saline were injected 60 min before each PTZ injection. Latency to the first minimal clonic seizure (MCS) and generalized tonic-clonic seizure (GTCS) and the percent of mortality were recorded.</p><p><strong>Results: </strong>Aqueous extract of clove at doses of 50, 100, 250 and 500 mg/kg significantly extended the MCS and GTCS latency (P<0.05). The MCS latency in mice treated with 50, 100 and 250 mg/kg of the ethanolic extract was significantly increased (P<0.05). The GTCS latency in mice treated with 50, 100, 250 and 500 mg/kg of ethanolic extract was significantly higher than that of the saline-treated group (P<0.05). There were no significant differences in MCS and GTCS latency between mice treated with different chloroformic extract of clove or saline.</p><p><strong>Conclusion: </strong>The aqueous and ethanolic extracts of clove could inhibit the PTZ-induced convulsion, and this plant has the potential to be used as a new therapeutic agent for control of seizures. The connective tissue around the needle-tip of LU acupoints in continuous thin-section CT image sets was observed. The LU acupoints and part of the meridian were three-dimensional reconstructed in vivo and displayed with digital medical technology based on the CT-scan data. Then, the images were compared with traditional meridian records.</p><p><strong>Results: </strong>The connective tissue structure around the needle-tip of the eight LU acupoints was observed as follows: 5 acupoints were closely related to periosteum and 3 of them were closely related to blood vessels and nerve sheath. Three-dimensional reconstruction of anatomical position of the Lung Meridian of Hand-Taiyin was successfully completed in vivo, and the traveling course of meridian was similar to classical meridian illustration in traditional Chinese medicine.</p><p><strong>Conclusion: </strong>There is no specific entity of anatomical structure of LU acupoints and the meridian. Yinchen Wuling San (YCWLS) and Zhizi Baipi Decoction (ZZBPD) on liver fibrosis induced by dimethylnitrosamine (DMN) in rats and explore the formula-syndrome relationship.\u0000\u0000\u0000METHODS\u0000Forty-eight rats were weighted, and divided into five groups: normal control, DMN, YCHD, YCWLS and ZZBPD groups using stratified random method. Liver fibrosis in rats was induced by intraperitoneal injection of DMN for 4 weeks at the dosage 10 mg/kg body weight, once per day for 3 consecutive days in each week. Each groups was gavaged with distilled water (control or model group), YCHD, YCWLS and ZZBPD respectively in the last 2-week modeling period. At the end of the 4th week, rats were sacrificed and their blood sample and liver tissue were taken for detecting pathology and analyzing of hydroxyproline (Hyp) and liver function.\u0000\u0000\u0000RESULTS\u0000Compared with the normal control group, DMN model group showed liver damage and liver fibrosis with remarkably increased Hyp content (P<0.01) and abnormal liver function (P<0.01). Liver fibrosis induced by DMN in rats was improved by administration of YCHD. Compared with DMN model group, content of Hyp was decreased remarkably (P<0.01) and liver function and hepatic histology were improved significantly after 2 weeks of YCHD treatment (P<0.05 or P<0.01)., and content of hepatic Hyp was decreased in the ZZBPD groups. Comparison of various parameters between groups showed that content of serum total bilirubin was decreased significantly in the YCWLS group (P<0.01), and content of hepatic Hyp was decreased in the ZZBPD group.\u0000\u0000\u0000CONCLUSION\u0000Efficacy of YCHD in inhibition of liver fibrosis induced by DMN in rats is far better than YCWLS and ZZBPD. The pathogenesis of the liver fibrosis induced by DMN is mainly damp-heat. In addition, both damp and heat pathogenic factors play very important role in this model. The proliferation of primary chondrocytes was assessed in second-passage cultures using cell counting kit-8 and the growth curve was drawn. Type II procollagen gene (Col2a1) expression in chondrocytes was also identified using cell immunofluorescence assay. The second-passage chondrocytes were divided into five groups, including control group and osthole groups at 6.25, 12.5, 25 and 50 μmol/L. The growth property of rat chondrocytes was observed after 24, 48 and 72 h of culture with osthole at corresponding dose. Both protein and mRNA expression of proliferating cell nuclear antigen (PCNA) and cyclin D1 was measured by Western blot and polymerase chain reaction methods.</p><p><strong>Results: </strong>The second-passage chondrocytes were viable and showed Col2a1 expression in the cytoplasm. The proliferation of rat chondrocytes was inhibited by osthole in a dose-dependent manner. It is important to take effect by following the principle of formulas corresponding to syndromes. However, some medical practitioners always feel that the actual clinical effect is far less than expected. Six errors in the use of classic prescriptions as well as the theory of formulas corresponding to syndromes are the most important causes to be considered, i.e. paying attention only to the local syndromes while neglecting the whole, paying attention only to formulas corresponding to syndromes while neglecting the pathogenesis, paying attention only to syndromes while neglecting the pulse diagnosis, paying attention only to unilateral prescription but neglecting the combined prescriptions, paying attention only to classic prescriptions while neglecting the modern formulas, and paying attention only to the formulas but neglecting the drug dosage. Therefore, not only the patients' clinical syndromes, but also the combination of main syndrome and pathogenesis simultaneously is necessary in the clinical applications of classic prescriptions and the theory of prescription corresponding to syndrome. Seventy-five commonly used clinical markers were selected, including liver and kidney function, clotting function, the quantitative detection of hepatic B virus (HBV) markers, HBV-DNA, blood count, hormones levels, cellular immunity indicators, humoral immunity indicators, lipid panel, protein electrophoresis, alpha-fetoprotein and liver fibrosis indicators. Receiver operating characteristic (ROC) curve was used to detect the diagnostic efficiency of single differential indicators, and stepwise discriminant analysis model was used to explore the discrimination efficiency of differential indices between two TCM syndromes in CHB.</p><p><strong>Results: </strong>The differential indices between two CHB Chinese syndromes were albumin, prothrombin time, immunoglobulin A, immunoglobulin M, blood urea nitrogen, blood uric acid, basophils, basophil percentage and mean platelet volume. Network biology and network pharmacology were built by applying network-based approaches in biomedical research. The cardiovascular system may be regarded as a complex network, and cardiovascular diseases may be taken as the damage of structure and function of the cardiovascular network. Although Chinese medicine (CM) is effective in treating cardiovascular diseases, its mechanisms are still unclear. With the guidance of complex network theory, network biology and network pharmacology, network-based approaches could be used in the study of CM in preventing and treating cardiovascular diseases. A new discipline-network cardiovasology of CM was, therefore, developed. In this paper, complex network theory, network biology and network pharmacology were introduced and the connotation of \"disease-syndrome-formula-herb\" was illustrated from the network angle. Network biology could be used to analyze cardiovascular diseases and syndromes and network pharmacology could be used to analyze CM formulas and herbs. The zebrafish (Danio rerio) has recently become a popular model in the field of drug screening, specifically emerging as an important vertebrate model for in vivo high-content drug screening of multiple efficacy parameters and whole-organism toxicity. The authors also discussed the advantages of the zebrafish model for evaluating drug metabolism. {"title":"Ameliorative potentials of Syzygium jambolanum extract against arsenic-induced stress in L6 cells in vitro.","authors":"Asmita Samadder,&nbsp;Jayeeta Das,&nbsp;Sreemanti Das,&nbsp;Raktim Biswas,&nbsp;Anisur Rahman Khuda-Bukhsh","doi":"10.3736/jcim20121114","DOIUrl":"https://doi.org/10.3736/jcim20121114","url":null,"abstract":"<p><strong>Objective: </strong>To determine the ameliorative potentials of Syzygium jambolanum (SJ) extract in L6 skeletal muscle cells in regard to arsenic-induced impairment of optimum glucose homeostasis and improper functioning of mitochondria.</p><p><strong>Methods: </strong>Several study parameters like glucose level and mitochondrial functioning through indexes of pyruvate kinase, glucokinase and mitochondrial membrane potential were assessed. The expression of the relevant marker proteins and mRNAs like glucose transporter 4 (GLUT4), insulin receptor substrate 1 (IRS1), IRS2 and glucokinase for tracking down the signalling cascade were critically analyzed.</p><p><strong>Results: </strong>Introduction of SJ extract could bring about positive modulation of various markers, by acting on GLUT4, thereby bringing about an attenuation of the arsenite-induced toxic conditions in L6 cells.</p><p><strong>Conclusion: </strong>Syzygium jambolanum extract has considerable ameliorating potentials against arsenic-induced glucose imbalance and stress and has possibility of therapeutic use in the management of arsenic-induced toxicity including hyperglycemia.</p>","PeriodicalId":23993,"journal":{"name":"Zhong xi yi jie he xue bao = Journal of Chinese integrative medicine","volume":"10 11","pages":"1293-302"},"PeriodicalIF":0.0,"publicationDate":"2012-11-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"31052169","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

{"title":"Antihepatotoxic activity of Euphorbia neriifolia extract against N-nitrosodiethylamine-induced hepatocarcinogenesis in mice.","authors":"Veena Sharma,&nbsp;Pracheta Janmeda,&nbsp;Ritu Paliwal,&nbsp;Shatruhan Sharma","doi":"10.3736/jcim20121115","DOIUrl":"https://doi.org/10.3736/jcim20121115","url":null,"abstract":"<p><strong>Objective: </strong>To scrutinize the protective role of hydroethanolic extract of Euphorbia neriifolia leaves (HEEN) against N-nitrosodiethylamine (DENA)-induced hepatocarcinogenesis in male Swiss albino mice.</p><p><strong>Methods: </strong>Experimental mice were pretreated with 150 and 400 mg/kg of HEEN, or 0.5% and 1% of butylated hydroxyanisole (BHA) as a standard for 14 d prior to the administration of a single dose of 50 mg/kg of DENA. Levels of xenobiotic metabolizing enzymes such as cytochrome (Cyt) P450 and Cyt b5, activities of reduced glutathione (GSH), glutathione-S-transferase (GST), aspartate amino transaminase (AST), alanine amino transaminase (ALT) and alkaline phosphatase (ALP), and total protein and cholesterol content in the liver tissues were measured to determine the hepatotoxicity caused by DENA.</p><p><strong>Results: </strong>The levels of Cyt P450 and Cyt b5 were significantly increased, and GST and GSH were significantly depleted after DENA treatment (P<0.01). The activities of AST, ALT and ALP, and the total protein content were also significantly dropped off (P<0.01). The total cholesterol level was markedly increased by DENA as compared with the normal group (P<0.01). However, the pre-supplementation of HEEN showed a remarkable amelioration of these abnormalities, and the levels of the antioxidant enzymes in the liver were significantly restored, which exhibited the dose-dependent protective effect against DENA-induced hepatotoxicity.</p><p><strong>Conclusion: </strong>HEEN exerts its chemopreventive effects by alleviating the xenobiotic enzymes and enhancing the levels of antioxidants and biochemical assays in DENA-induced carcinogenesis by reducing the formation of free radicals.</p>","PeriodicalId":23993,"journal":{"name":"Zhong xi yi jie he xue bao = Journal of Chinese integrative medicine","volume":"10 11","pages":"1303-9"},"PeriodicalIF":0.0,"publicationDate":"2012-11-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"31052170","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
