{"title":"[The enzymic and immunological determination of coeruloplasmin: sex- and method-dependent differences during the administration of oestrogens (author's transl)].","authors":"H Ebeling","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>After 14 days of 1 mug ethinyloestradiol per kg body weight per day, both sexes show highly significant increases in serum coeruloplasmin which are independent of the determination method. Expressed on a percentage basis, the concentrations of coeruloplasmin under oestrogen medication determined by the p-phenylendiamine oxidase reaction are significantly higher than those determined by the immunological-nephelometric method. This is because several coeruloplasmin components, mainly C and DO, take part in the determination with p-phenylendiamine oxidase, whereas the immunological method measures chiefly the coeruloplasmin DO component. The significant sex differences under oestrogen medication shown by the determination with p-phenylendiamine oxidase, can therefore be explained by the presence of C- and D-coeruloplasmins in women, while the serum of men contains almost exclusively D-coeruloplasmins. Under oestrogen medication, the increase occurs primarily in the DO- coeruloplasmin component, and women also show an increase in coeruloplasmin-C. The significant higher enzymatic concentrations in the serum of the women in contrast to the men are thus plausibly explained. The method is based on the determination of inorganic phosphorus released by the action of the two enzymes on adenosine-5'-monophosphate at pH 9.5 (200 mmol/l tris-buffer) in the presence and absence of L-cysteine. {"title":"[New protease-inhibitors with broad specificity in the polychaet Sabellastarte indica (Savingny), I (author's transl)].","authors":"C Gauwerky, G Corman, G Uhlenbruck","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>In this paper we describe new and so far unknown protease inhibitors present in the tentacles of the annelid Sabellastarte indica Savingny. At least five different isoinhibitors with inhibitory activity towards trypsin, plasmin, chymotrypsin and kallikrein can be separated electrophoretically. Our protease inhibitor active material differs from the other well known protease inhibitors found in invertebrates in its high molecular weight, in that it is heat-labile and in the occurrence of the isoelectric point in the weakly acid region. On the other hand, the new protease inhibitors have some similarities to the soybean inhibitor described by Kunitz, and to ovomucoid. Oestriol is detected as a derivate of 1-dimethylaminonaphtalene-5-sulphonylchloride (DANSYLchloride). The method has the following characteristics: 1. The recovery of oestriol-16-glucuronide is 94%. 2. For the interpretation of the results obtained with this method mean values and 95% probability bounds of urinary oestriol excretion for each week of the last three months of pregnancy are given. (e.g. 49--125 mumol/24 h at 32 weeks of gestation, 108--238 mumol/24 h at 40 weeks). 3. The coefficient of variation of the method is 4% (from day to day). 4. The lowest detectable concentration is 3 mumol/l. 5. Influence of glucose can be eliminated. 6. From one sample this instrument determines the value for platelets (PBC), white blood cells (WBC), red blood cells (RBC), hemoglobin (Hb) and the hematocrit simultaneously. Moreover the system calculates the derived values: the mean cell volume (MCV), mean cell hemoglobin (MCH) and the mean cell hemoglobin concentration (MCHC). The values were investigated for cross sample contamination, coincidence, reproducibility, and compared with other methods. In place of a radioisotope, an enriched form of the stable isotope 46Ca was administered orally, and the proportion of the applied dose of 46Ca appearing in the serum and urine was measured. 46Ca was measured by neutron activation over 47Sc. The clinical picture showed a close correlation with the determined resorption capacities. {"title":"[A comparative study of total protein and albumin in man, monkey, dog and rat employing different analytical methods (author's transl)].","authors":"A Metz, A Schütze","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>In the present study, serum albumin was determined colorimetrically in three animal species and in man with the help of four currently used dye-reagents (protein binding reagents). The results were compared with those obtained from corresponding electrophoretic and biuret determinations, using Versatol as a standard-control throughout. Using 2-(4-hydroxyazobenzene)-benzoic acid and bromocresol purple as reagent for the albumin determinations, species-specific differences from the electrophoretic results were found. No such differences occurred with bromo-cresol green, except in rats. There was no significant difference in albumin concentrations between man, monkeys and rats.
