Hence, extensive research is required to find out the mechanisms of action as well as bioactivity of the various phytochemicals and efficacy of the medicinal values of plants. The plant Euphorbia hirta is commonly called as Asthma weed. It belongs to the family Euphorbiaceae. The present study is detailed out to complete fluorescence analysis. The study will provide proper information about the presence of bioactive molecules in the crude drug i.e. Euphorbia hirta plant. The method showed a linear response for concentration of range of 5-15μg/ml using Potassium Phosphate Buffer(PH 7): Acetonitrile solution in the ratio of (35:65) as the mobile phase with detection at 240 nm and flow rate 1ml/min and the retention time for Metformin HCl and Anagliptin was found to be 4.227 and 5.893 respectively. The method was validated for the Specificity, Precision, Robustness, Accuracy, LOD, LOQ, Assay. The drug undergoes degradation under Acidic, Basic, Oxidation, Photolytic and Thermal degradation. All the peaks of degraded product were resolved from active ingredient with significantly different retention time. Bedarkar","doi":"10.20959/WJPR201711-9641","DOIUrl":"https://doi.org/10.20959/WJPR201711-9641","url":null,"abstract":"IntroductionPowder of fruit pulp of Indian gooseberry (Amalaki) levigated multiple times with its juice i.e. Amalaki swarasa bhavita Amalaki churna (ASBAC) is commonly regarded as Amalaki Rasayana (AR) is recommended in Ayurvedic classics and practiced in the management (treatment, prevention of complications) of Madhumeha (Diabetes). Alpha amylase and alpha glucosidase activities of Amalaki Rasayana has not been evaluated till the date. Hence, its in-vitro study was conducted. Aims and objectives-To evaluate amylase and glucosidase activities of Amalaki Rasayana. Materials and methodsAmalaki Rasayana was prepared with 16 times levigation (Bhavana) of fine powder of fruit pulp of Indian gooseberry (Amalaki, Emblica officinalis Gaertn.) with its fresh fruit pulp juice equal in quantity for each levigation leading to 36.33% of powder and 63.66% of total solid content derived from 581.39% of fresh fruit pulp juice. Methanolic extract of Amalaki Rasayana was tested for Porcine pancreatic amylase inhibition and intestinal glucosidase activities at 4 concentrations (40, 80, 120 and 160 μg/ml) through starch as base and chromogenic DNSA (3, 5-dinitrosalicylic acid) as colouring agent, in 2 different solutions by recording absorbance with UV VIS Spectrophotometer. Results-Amalaki Rasayana exhibited comparatively better amylase inhibition and -glucosidase inhibition by acetic acid buffer and inhibition was dose dependent at all concentration. ConclusionAmalaki Rasayana exhibited inhibition of Pancreatic Alpha amylase and intestinal alpha glucosidase in dose dependent manner in the concentration of 40, 80, 120 and 160 μg/ml thus have potential to reduce post prandial hyperglycemia. There are few studies in the literature about the bioactivities of Algerian Ulva lactuca marine algae extracts; that remain largely unexploited, thus this study aimed to examine whether polysaccharide or lectine fraction from Algerian U. lactuca are believed to promote anti-inflammatory and immunomodulatory properties. Methods: the polysaccharide or lectine fraction extracted from Algerian U. lactuca were evaluated in vivo by testing the carrageenan induced acute inflammatory in Wistar rats and the in vivo immunomodulatory effect. The U. lactuca metabolites were also examined for their capacity to inhibit the synthesis of TNF-α, and the release of NO in mouse macrophages RAW 264.7 stimulated with LPS. Results: The results showed that the both U. lactuca polysachrides (ULPF) and lectines (ULLF) phases at 100 mg/kg proved a significant anti-edema effect. The ULPF phase was significantly more effective in decreasing MPO activity and MDA level in rat paw than ULLF phase. Both ULPF and ULLF phases enhanced the in vivo macrophage phagocytic activity, this effect is more pronounced in the ULLF-group than that in ULPF-group. At 60 μg/mL both ULPF or ULLF fraction from U. lactuca were weakly cytotoxic on RAW cells. Colon-specific drug delivery has gained increased importance not just for the delivery of the drugs for treatment of local diseases associated with the colon but also for its potential for the delivery of proteins and therapeutic peptides. Different approaches are designed based on prodrug formulation, pH-sensitivity, time-dependency (lag time), microbial degradation and osmotic pressure etc to formulate the different dosage forms like tablets, capsules, multiparticulates, microspheres, microbeads, liposome for colon targeting. The efficiency of drug delivery system is evaluated using different in vitro and in vivo release studies. Acharya Sushruta mentioned prognosis of Madhumehaj vrana as “Krichrasadhya” (difficult to treat), which is experienced even in present day practice. Slight injury to glucose laden tissues of diabetic patients may result in ulcer formation and they become at high risk for major complications, including infection and amputation. PriyangvadiTaila contains the drugs possessing Shodhan and Ropana properties, thus considered as a good curative measure for local treatment of Vrana. A case report of Madhumehajanya Vrana successfully treated by local application of Priyangvadi-Taila has been presented here. The plant Zanthoxylum rhetsa has a long history in herbal medicine in many countries. Experiments were conducted following standard procedures. The extract was evaluated for their invitro antioxidant, antimicrobial and total phenol and flavonoid content. The AQZR ointment were administered topically, for evaluating the wound healing potential in excision wound model for twenty one days. Povidone iodine ointment was the standard for excision wound model. Extracts treated group showed in vitro antioxidant, anti microbial properties as compared with standard and control group. AQZR exhibited similar wound healing activity that of the standard but with lesser magnitude. The result may be attributed to the phytoconstituents such as flavonoids and phenolics present in it which may be due to their individual or cumulative effect that enhanced wound healing and provided scientific evidence to the ethnomedicinal futures of Zanthoxylum rhetsa.
