{"title":"Characterization of Heading- and Yield-related Gene Loci in the Cheongcheong/Nagdong Doubled Haploid Line using Rice QTLs","authors":"Yoon-Hee Jang, Jae-Ryoung Park, Kyung-Min Kim","doi":"10.7740/KJCS.2019.64.1.001","DOIUrl":"https://doi.org/10.7740/KJCS.2019.64.1.001","url":null,"abstract":"A quantitative trait loci (QTL) analysis of traits related to heading and yield was performed develop rice cultivars that are both early maturing and panicle weight type varieties. Our analysis included 120 strains of the Cheongcheong Nagdong doubled haploid (CNDH) variety. An observational growth experiment was conducted to identify genetic agronomic traits of CNDH. Heading date, ten plant weight, moisture, thousand grain weight, and yield had a normal distribution based on the frequency distribution table of the observational growth data. The QTL analysis found one heading-related and nine yield-related QTLs. The LOD of 2.85 was the largest in QTLs for heading date (QHD), 5.39 in QTLs for ten plant weight (QTPW), 3.92 in QTLs for moisture (QM), 4.80 in QTLs for thousand grain weight (QTGW), and 3.7 in QTLs for yield (QY). Genomic analysis detected 58 candidate genes on chromosome 2, 3, 7, 8, and 10. Among those, we found Rcd1 protein and OsERF3 gene in QM, MtN3 and zinc finger protein genes in QTGW, and OsNAC3 protein gene in QY. Increasingly frequent abnormal weather occurrences could include severe drought, which could cause water stress to rice during the seedling stage. This experiment was conducted to clarify the effects of drought during the seedling period on yield and quality of rice. Drought conditions were created in a rain shelter house facility. The drought treatment was conducted at 3, 10, and 20 days after transplanting. Soil water content was measured by a soil moisture sensor during the whole growth stage. In this study, we have chosen 3 rice cultivars which are widely cultivated in Korea: ‘Haedamssal’ (Early maturing), ‘Samkwang’ (Medium maturing), and ‘Saenuri’ (Mid-late maturing). The decrease in yield due to drought treatment was most severe 3 days after transplanting because of the decrease in the number of effective tillers. The decrease in grain quality due to drought treatment was also most severe 3 days after transplanting because of the increased protein content and hardness of the grains. The cultivar ‘Haedamssal’ was the most severely damaged by water stress, resulting in about a 30% yield loss. Drought conditions diminished the early vigorous growth period and days to heading in early-maturing cultivars. The results show that drought stress affects yield components immediately after transplanting, which is a decisive factor in reducing yield and grain quality. In WHS, rice standing percentage and weedy rice occurrence are the most important factors considered to secure a rice yield. We investigated the optimum seeding date of WHS in the Southern Plain area of South Korea. Weedy rice needed two weeks at 15°C to show over 80% emergence. Germinated rice seed grown at 20°C needed over for 10 days to achieve a shoot length over 3 cm. In field cultivation, the mean temperature for ten days after seeding showed a highly positive correlation with rice standing rate, spikelet number per square meter and yield index that favorably compared to machine transplanting. TP seeds were inoculated with Herbaspirillum spp. at concentrations (%v/v) of 0 (control), 10, 20, and 40% at 20, 25, and 30oC in a growth chamber for seven days. It was observed that the phenolic content (PC) including flavonoid, rutin, and tanin increased with an increase in inoculant rate at 20oC, whereas the PC content increased with an increase in temperature regardless of the inoculant rate. In Tunisia, salt stress is one of the main problems that limit crop production. ‘Mahmoudi’ was selected as the most salt-sensitive out of 11 Tunisian durum cultivars. Using the salt-tolerant cultivar ‘Om Rabia’, resistant and susceptible cultivars were evaluated to compare genetic responses under salt stress. At the fully expanded third leaf stage, salt stress was applied by submerging the pots in 500 mM NaCl for 5 min every day for saline water irrigation in the greenhouse. The treatment was applied for 1 week and salt stress tolerance was determined by changes of growth parameters to the control condition. The salt tolerance trait index and salt tolerance index were calculated and used as selection criteria. The expression levels of TdHKT1;4, TdHKT1;5, and TdSOS1 were examined using qPCR. For further evaluation of physiological responses, salt stress (150 mM NaCl) was additionally applied for 48 h at the fully expanded third-leaf stage. The heading date for the water-broadcast seeded silage rice was September 11-18, which was delayed by 11-17 days when compared to that for the machine-transplanted silage rice. On average, water-broadcast seeded silage rice had a shorter plant height, more panicles per area, and a greater biomass production because of the increased straw dry weight. However, the difference in dry weight of each plant organ between the two cultivation practices exhibited yearly variation. When the data were pooled across experimental years, cultivation practices, and varieties, biomass production was highly positively correlated with straw dry weight but not with panicle dry weight. When the ratio of water-broadcast seeding to machine-transplanting was analyzed, total dry weight and straw dry weight were positively associated with each other, whereas no relationship was found between total dry weight and panicle dry weight. Despite that water-broadcast seeded silage rice produced more panicles per area than machine-transplanted silage rice, the two cultivation practices had a similar dry weight per culm. Therefore, we conclude that the silage rice from the water-broadcast seeding, compared to the machine-transplanting, produced more biomass because of the combination of the increased panicle number per area and a similar dry weight per culm. Flax seedlings were cultivated in a greenhouse with average ambient temperature ranging from 35°C to 38°C during the growing period, and then PB and PC were applied on the leaves. The height of flax plants was reduced significantly by the foliar application of PB and PC. The highest ripening seed rate recorded was 94.5% with 2000 ppm PC; a 10.3% increase compared with that of the control. Both PB and PC markedly increased the fructification of flax seed. The application of two plant growth retardants resulted in higher seed production in flax seedlings grown in open field than in those grown in a greenhouse. The highest oil content was observed in flax treated with PC, whereas, the lowest oil content was observed in flax treated with PB. The oil content and ratio of unsaturated fatty acids were higher in flax seedlings grown in greenhouse than in those grown in open field. The level of two endogenous bioactive gibberellins (GA1 and GA4) in flax seeds decreased with the application of PB and PC. The flax plants treated with PB and PC presented higher secoisolariciresinol diglucoside content than that of the control. The optimal concentration of PC was found to be 2000 ppm.
