{"title":"Sleep Basic Research on Verifying the Effects of Natural Compounds on Wakefulness or Sleep","authors":"S. Nishimon","doi":"10.14789/jmj.jmj21-0022-r","DOIUrl":"https://doi.org/10.14789/jmj.jmj21-0022-r","url":null,"abstract":"I studied at the Sleep and Circadian Neurobiology Laboratory, Department of Psychiatry and Behavioral Sciences, Stanford University School of Medicine, from April 2018 to March 2020. At Stanford University, I mainly researched the following themes: (1) sleep basic research using mice (administering compounds with sleep or wakefulness effects to mice and examining their effects), and (2) research on circadian rhythm disorders. There are only a few institutions in the world that can conduct sleep basic research using mice, and Stanford University is a wonderful environment to immerse yourself in research, as it is home to not only psychiatrists but also neurologists and many basic researchers. In this article, I would like to review the experiments I conducted during my study abroad, using mice to verify the effects of natural compounds on wakefulness or sleep. In one study, we evaluated the effects of ginkgolides (A, B, and C) and bilobalide on arousal, locomotion, and core body temperature. The results showed that only ginkgolide B dose-dependently increased the amount of arousal and decreased the amount of NREM sleep in the physiological sleep-wake cycle of mice. In another study, we tested the sleep-inducing effects of sake yeast in mice under an acute insomnia model. East Berlin was governed by the Soviet Union, and West Berlin was governed by France, Great Britain, and the United States. West Berlin and West Germany were connected by autobahns, railroads and planes, and West Berlin developed with West Germany. On November 9, 1989, the Berlin Wall suddenly collapsed. Many people were destroying the Berlin Wall with a hammer, as if they vented their anger for years. The work to remove the Berlin Wall began the next day, and East and West Germany were unified on October 3, the following year. I tried two studies on liver transplantation in West Germany. One was an experimental study on the effects of the donor’s fasting and graft viability after liver transplantation, and the other was a study on the role of Kupffer cells in the induction of tolerance after liver transplantation. The adrenal cortex secretes the corticosteroids cortisol and dehydroepiandrosterone (DHEA) under stress condition, and serum levels of DHEA are gender-dependent. Although BDNF expression is associated with these corticosteroids, the effect of gender on this association is not clear. To examine gender differences in the relationship between BDNF and corticosteroids in patients with MDD, we measured serum levels of cortisol, DHEA-sulfate (DHEA-S) and BDNF in men and women with depression. We recruited 189 inpatients with MDD (76 men and 113 women) from Juntendo Koshigaya Hospital. Serum cortisol, DHEA-S and BDNF levels were measured on the morning after admission. Multiple regression analyses were conducted to assess the effects of cortisol and DHEA-S on BDNF. After controlling for age, we found that serum BDNF levels were significantly influenced by the DHEA-S/cortisol ratio (p = 0.010) in female participants. However, this association was absent in male participants. These results suggest gender differences exist in the effects of corticosteroids on BDNF.
