Loreto","doi":"10.15761/ohc.1000187","DOIUrl":"https://doi.org/10.15761/ohc.1000187","url":null,"abstract":"Aim : The present study aimed to verify the histone MacroH2A.1 immunohistochemical expression and localization after orthodontic force application on rat maxillary molars to deep insight the molecular mechanisms involved in tooth movement. Materials and Methods : Sixteen Sprague-Dawley rats were used as previously described. Tooth movement was induced, and rats were then killed. Molar-bearing segments of alveolar bone were cut from each side and further fixed in paraformaldehyde, decalcified, and then dehydrated. Specimens embedded in paraffin, including the crestal areas mesial and distal to the maxillary molars, were cut by microtome and processed for immunohistochemistry to detect macroH2A.1 according to literature protocols. Results : On the compression side, MacroH2A.1 was strongly immunostained in the spindle round cells of PDL, in cementoblast cells of root teeth and osteoclasts of alveolar bone. Sections also showed vacuolization of odontoblastic layer near to the pulp tissue. Alveolar bone evidenced the presence of marrow spaces. On the tension side, odontoblastic cells near to pulp tissue take spindle shape and showed a Journal of Oral Pathology and Medicine - Proof for Peer Review for Review Only scattered cell immunolabelling. Negative controls showed no immunolabelling. However, a polarization of caries in children of high-risk groups (frequently coming from families presenting a low educational level or a migratory background) has occurred [14]. A lack of oral health literacy and language barriers may be causative. Caries and/or pain constituted the reasons for seeking dental attendance in the preschoolers admitted to dental general anesthesia (DGA) who were investigated in this study [15,16]. DGA presents an accepted treatment method in individuals who are (due to infantile incomprehension, dental anxiety, intellectual and/or physical disablement, or systemic diseases) unamenable to routine dental treatment, even though it requires high expenditure and implicates the risk of anesthetic complications [17-19]. DGA has been shown to enhance children’s oral health-related quality of life and dental behavior [20-24]. In acute settings, DGA provides a rapid onset of action and an optimal titration of anesthetics [25]. However, emergency DGA presents an organizational challenge with respect to the 24/7 provision of pre-, periand postoperative care requiring the respective medical and nursing staff. For the children ́s benefit, scheduled DGA in non-pain patients displaying carious lesions seems preferable to a stressful emergency procedure, also with respect to the possible conservation/restoration of carious teeth instead of tooth extractions. The question of interest in this study was, if the presence of toothache is associated with higher numbers of deeply carious primary teeth. The literature was reviewed in order to investigate the failure and the reasons for failure of pediatric crowns. Electronic databases were screened, and eligible studies were hand-searched to find clinical studies evaluating the survival of different crowns placed in primary teeth with follow-up. Among those, an elevated number of failures were reported with every other type of crown. The high variation on failure rate among the crowns can be due to several contributing factors out of which children's behaviour during the clinical procedure can be the most vital. In contrast to the familial MEN2, little is known about the etiology of sporadic MTC. New approaches are required to elucidate the mechanisms underlying the pathogenesis of sMTC. Long noncoding RNAs (lncRNAs), are wellrecognized post-transcriptional regulators of genetic expression and recent studies have described multiple aberrantly expressed non-coding RNAs in thyroid cancers. In the current study we have aimed to perform the first screening of multiple lncRNAs in tumoral tissues from MTC patients by qRT-PCR. Our analysis showed the association of 15 lncRNAs from which 6 where new in association with this disease (RMST, SNHG16, FTX, GAS5, IPW, MEG3). MTC can occur either sporadically (75%) or as the dominant component of the type 2 multiple endocrine neoplasia syndromes (MEN2, 25%). It is considered a rare disease, with an estimated prevalence in the general population of 1/14,300 [http:// www.orpha.net; ORPHA No: 1332]. The broad term long non-coding RNA (lncRNA) refers to a class of non-coding RNA transcript of minimum 200 nucleotides in length. They have gained widespread attention in recent years as new players in transcriptional, epigenetic, or post-transcriptional regulation of gene expression [3]. To date, only one study has examined the expression of lncRNAs in patients with MTC [4]. Consequently, lncRNAs are attractive and promising targets in cancer prognosis and treatment. The purpose of this study is to bring insight and deeper understanding into the etiology of sMTC, to a deeper understanding of disease mechanisms, pathogenesis, and searching of new therapeutic targets. To afford this aim, we have analyzed the expression of lncRNAs in this type of tumors. Materials and methods Experimental subjects In this study, we have performed lncRNA expression analysis on four sMTC cases (Table 1). All MTC tissues and their corresponding adjacent non-tumor thyroid tissues were obtained from these patients after undergoing surgical resection.
