Because the clinical signs of human brucellosis are so varied and vague, scientific confirmation via organism isolation or antibody detection is required to confirm the diagnosis. The aim of this work is measurement of the sensitivity and specificity of PCR for the detection of Brucella abortus BCSP31gene compared with the blood culture methods.Methods: A total of 97 patients from different ages with symptoms of brucellosis were included in the study. This research study was carried out at Mansoura University Hospitals. The sensitivity and specificity of polymerase chain reaction (PCR) for the detection of BCSP31gene of Brucella abortus compared with blood culture methods.Results: In positive PCR cases of brucella (total 14), a single band of a 223-bp corresponding to BCSP31gene was obtained. The true positive brucellosis cases (n = 6) are positive of blood culture and PCR. PCR was sensitive than blood culture for diagnosis of brucellosis since the sensitivity and specificity of PCR are 87.5% and 98.73% and the sensitivity and specificity of blood culture are 84.62% and 96.15%. The positive and negative predictive values of PCR are 86.56% and 94.80% and the positive and negative predictive values of blood culture are 78.98% and 92.54%. The accuracy (%) of PCR and blood culture is 94.72% and 90.67%. The PCR assay was shown to be more sensitive and specific. The objective of this study was to diagnose typhoid fever by the amplification of hto gene. Methods: This study comprised 115 patients from different ages experienced the clinical symptoms of typhoid fever. This study was carried out at Mansoura University Hospitals. The blood samples collected were subjected to PCR to target the hto gene of Salmonella Typhi. Results: The PCR technique standardized for hto gene resulted in specific amplicon of 495-bp. In positive PCR cases of typhoid, a single band of 495bp corresponding to hisJ (hto) gene was obtained. The PCR assay was found to be highly specific and sensitive. Conclusion: When exceptional sensitivity is obligatory, PCR technique utilized as a very sensitive and particular test for the presence of salmonella in blood samples. Comparing results obtained with the limit of the Environmental Protection Agency for water body health showed the following: The mean of temperature and pH are within the permissible limits. Manganese is almost within the permissible limits, but in 2005, it was slightly above the permissible limit. The mean of total solids, hardness, biological oxygen demand, chemical oxygen demand, sodium, chloride, total nitrogen, total phosphorus, Iron are above the permissible limit. In some years, the dissolved oxygen is below the permissible limit. Plankton samples were taken monthly from 12 stations of Hurghada coast of the Egyptian Red Sea during the period from August 2014 to July 2015. Plankton nets of 20 µm mesh size were used in collecting samples, then samples fixed on board in 4% buffered formalin. A total of 149 species of tintinnids belonging to 12 families and 36 genera of the order Choreotrichida were identified based on the morphological features. Families Codonellidae, Tintinnidae, and Codonellopsidae had the higher number of species (40, 28 and 20 species, respectively), constituting collectively around 60% of the total number of recorded species. A total of 182 larvae belonging to seven fish families were collected and identified based on the morphological characteristics.The diseased larvae were clinically examined and macerated tissues were injected onto Trypton Soy Agar (1.5% NaCl ) and incubated at 25.5 °C for 2-5 days. The obtained bacterial isolates were identified as V. harveyi, V. parahaemolyticus, V. alginolyticus, V. vulnifacus, non virulence V. cholerae and unidentified Vibrio species by the morphological and biochemical characterization. The prevalence ratio of bacterial infection of all infected larvae was 101 larvae (55.5%). The highest number of infected larvae was recorded in summer (45 larvae, 86.5%), while the lowest was recorded in winter (10 larvae, 25%). The most frequently encountered Vibrio sp. was V. harveyi which was isolated from 28 fish larvae followed by V. parahaemolyticus which was isolated from 25 fish larvae. For synthesis of WTS/DaMK based geopolymer, five mixes of WTS/DaMK (75/25, 80/20, 85/15, 90/10 and 95/5, respectively) are tested to obtain the optimum synthesis condition based on the compressive strength. Results showed that the geopolymer mortar with 85/15WTS/DaMK has the highest compressive strength (8 MPa). In addition, X-ray Diffraction (XRD) and X-ray Fluorescence (XRF) techniques are used to characterize the mineral and chemical compositions of WTS and DaMK. WTS/DaMK based geopolymer mortar have been tested for leaching to study its immobilization behavior under the optimum condition. The Cd2+, Pb2+ and Hg2+ ions were used and effectively immobilized in this study. The heavy metals with concentrations of 100, 200 and 300 ppm were used in the geopolymer matrix giving about 98% of immobilization efficiency.
