Both RDW and NLR are predictors of chronic disease risk and mortality. Omega-3 index (O3I) values have previously been shown to be inversely associated with RDW and NLR levels. Our objective was to determine if total plasma long chain omega-3 fatty acids (Omega3%) measured in the UK Biobank cohort were associated with RDW and NLR values. RDW- and NLR- relationships with Omega3% were characterized in 109,191 adults (58.4% female). RDW- and NLR-Omega3% relationships were inversely associated with Omega3% (both p<0.0001). These cross-sectional associations confirm previous findings that increasing RDW and NLR values are associated with low O3I. Mounting evidence suggests that the omega-3 polyunsaturated fatty acids (PUFAs) eicosapentaenoic (EPA), docosahexaenoic (DHA) and docosapentaenoic (DPAn-3) acids are promising therapeutic options. However, meta-analyses of randomised controlled trials (RCTs) have produced inconsistent findings. This review assesses for the first time the efficacy of omega-3 PUFAs against the severity of anxiety and depression symptoms, measured by validated scales, with specific consideration of methodological issues encountered in this area. PubMed, CINAHL, PsycINFO, Cochrane Library and Web of Science were searched for eligible RCTs administering omega-3 PUFAs against anxiety and/or depression. This study adopts the PRISMA guidelines. Ten RCTs comprising 1509 participants were included in the quantitative synthesis. EPA-enriched interventions at [≥]60% of total EPA+DHA were associated with significant reduction in depression severity, compared to placebo (SMD: -0.32; 95% CI: -0.59, -0.06; p=0.02); however, EPA doses of [≥]2000 mg/day were not (SMD: -0.11; 95% CI: -0.43, 0.20; p=0.48). Only 10 RCTs fulfilled the eligibility criteria, and there were some concerns regarding bias and population heterogeneity, highlighting the lack of high-quality RCTs in this area. Overall, these results support previous observations where EPA at proportions [≥]60% of total EPA+DHA, up to 2000 mg, reduces depression scores. Starting from a diol, protected with good leaving groups (mesyl and tosyl), we performed a sequence of reactions with good yields: the carbon chain lengthening by reaction with KCN, the hydrolysis of the nitrile groups to carboxyl, the esterification of carboxyl to ester and finally the phosphonate synthesis, which gave one bis-β-ketophosphonate and two mono β-ketophosphonates. The new β-ketophosphonates are key intermediates for obtaining new prostaglandin analogues with a bicyclo[3.3.0]octene fragment in the ω-side chain. However, there is limited understanding of how fatty acids and their bioactive derivatives (oxylipins) change over the course of pregnancy. Here we document the changes in concentration of fatty acids and oxylipins during pregnancy and how fatty acid status and oxylipin concentrations are affected by supplementation with omega-3 LCPUFA. We also investigate the degree to which fatty acid and oxylipin changes across pregnancy are influenced by baseline omega-3 status.\u0000\u0000\u0000MATERIALS AND METHODS\u0000We profiled the fatty acids in all lipids in dried blood spots (total blood fatty acids) by gas chromatography and free (unesterified) fatty acids and their associated oxylipins in separate dried blood spot samples by LC-MS-MS collected from a random sample of 1263 women with a singleton pregnancy who participated in the ORIP (Omega-3 fats to Reduce the Incidence of Prematurity) trial. ORIP is a double-blind, randomized controlled trial involving 5544 participants and designed to determine the effect of supplementing the diets of pregnant women with omega-3 LCPUFA on the incidence of early preterm birth. Maternal whole blood finger prick samples were collected at baseline (~14 weeks gestation) and at completion of the study intervention period (34 weeks gestation).\u0000\u0000\u0000RESULTS\u0000The concentration of most total and free polyunsaturated fatty acids and their associated oxylipins declined over the course of pregnancy. Omega-3 LCPUFA supplementation increased total DHA and 7-HDHA and mitigated the decline in free DHA, 4-HDHA and 14-HDHA. The intervention had minimal or no effect on free EPA, LA, AA and their associated oxylipins. Omega-3 LCPUFA supplementation in women with higher omega-3 status at baseline was associated with a significant increase in 7-HDHA and 4-HDHA between the treatment and control whereas there were no differences between groups in 7-HDHA and 4-HDHA in women with intermediate or lower baseline omega-3 status.\u0000\u0000\u0000CONCLUSION\u0000Our data suggest a differential response with or without omega-3 supplementation for DHA and DHA-derived oxylipins, which may have an important role to play in modulating pregnancy duration.
