The theory concerns the attainment of approximate sedimentation-diffusion equilibrium in minimum time of centrifugation. This insert acts as a container for holding infected plant tissues for the purpose of separating virus by centrifugal force from intact and fresh plants, from frozen and thawed plants and from plant tissue disintegrated by mechanical means. The extracted fluid represents 80 to 90% of that present in the untreated plant tissue. In addition to these components a “soluble antigen” was found which had a sedimentation coefficient of about 22 S. From calibration curves with sodium chloride solutions of different molarities and specific gravities, the densities of the fluid in the droplets were obtained by interpolation. The density of the solutions of proteins may be determined and used in the calculation of partial specific volumes. {"title":"Purification of viruses by centrifugation in gradients of inert substances.","authors":"A Polson","doi":"10.1080/10826069308544544","DOIUrl":"https://doi.org/10.1080/10826069308544544","url":null,"abstract":"<p><p>By using a reorienting gradient centrifuge rotor cut from a block of Nylon and fitted with eight septae, it was possible to separate the components of the haemolymph of the mollusc Turbo sarmaticus into three fractions in a sucrose gradient held in the bowl of the rotor. The fractions were (108 and 98)S, 44S and 16-22S. The success of the experiment was due to the large differences in the sedimentation coefficients of the components. When the rotor was applied to the natural mixture of the five viruses of the caterpillars of Nudaurelia cytheria only the main component could be isolated in a pure state.
