Yan","doi":"10.3724/SP.J.1143.2012.11144","DOIUrl":"https://doi.org/10.3724/SP.J.1143.2012.11144","url":null,"abstract":"Two fungi PM-1 and PA-1 were isolated from P.micranthum and P.armeniacum,respectively,the former belonged to Epulorhiza while the latter belonged to Tulasnella calospora.In this study,seedlings of P.armeniacum and P.micranthum were inoculated with PM-1 on DE medium,the biomass and the content of Ca,Mn of both P.armeniacum and P.micranthum seedlings were higher than those in the control seedlings,which indicated that PM-1 could improve the growth of those species.Seedlings of P.armeniacum and P.micranthum were inoculated with PA-1 on DE medium,the biomass in both species was no difference with those in the control ones.But PA-1 enhanced the dry weight of P.armeniacum according to the rate of dry weight increase.The impact of PM-1 on P.micranthum was higher than on P.armeniacum,while PA-1 impacted conversely,which demonstrated that fungi had stronger improving effect only on its own host plants instead of any other orchids.P.armeniacum seedlings were inoculated with PM-1 and PA-1 on Harvais medium.The growth of seedlings was inhibited by fungi.The results indicated that nutrient level of medium played an important role in affecting the symbiotic relationship between P.armeniacum seedings and endophytic fungi.Poor-nutrient medium was beneficial to the symbiotic relationship and nutrient-rich medium was not conducive to the symbiosis.","PeriodicalId":20246,"journal":{"name":"Plant Diversity and Resources","volume":"os-16 1","pages":"171"},"PeriodicalIF":0.0,"publicationDate":"2012-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"87195271","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Cai, Liping Tang, Zhu-Liang Yang","doi":"10.3724/SP.J.1143.2012.12140","DOIUrl":"https://doi.org/10.3724/SP.J.1143.2012.12140","url":null,"abstract":"Some species of the genus Amanita are economically important gourmet mushrooms,while others cause dramatic poisonings or even deaths every year in China and in many other countries.A DNA barcode is a short segment or a combination of short segments of DNA sequences that can distinguish species rapidly and accurately.To establish a standard DNA barcode for poisonous species of Amanita in China,three candidate markers,the large subunit nuclear ribosomal RNA (nLSU),the internal transcribed spacer (ITS),and the translation elongation factor 1-alpha (tef1-α) were tested using the eukaryotic general primers for their feasibility as barcodes to identify seven species of lethal fungi and two species of edible ones which can easily be confused with the lethal ones known from China.In addition,A.phalloides—a European and North American species closely related to one of the seven taxa,A.subjunquillea was also included.PCR amplification and sequencing success rate,intra-and inter-specific variation and rate of species identification were considered as main criteria for evaluation of the candidate DNA barcodes.Although the nLSU had high PCR and sequencing success rates (100% and 100% respectively),occasional overlapping occurred between the intra-and inter-specific variations.The PCR amplification and sequencing success rates of ITS were 100% and 85.7% respectively.ITS showed high sequence variation among species group and low variation within a given species.There was a relatively high PCR amplification and sequencing success rate for tef1-α (85.7% and 100% respectively),and its intra-and inter-specific variation was higher than that of ITS or nLSU.All three candidate markers showed hight species resolution.ITS and tef1-α had a more clearly defined barcode gap than nLSU.Our study showed that the tef1-α and nLSU can be proposed as supplementary barcodes for the genus Amanita,while ITS can be used as a primary barcode marker considering that the ITS region may become a universal barcode marker for the fungal kingdom.","PeriodicalId":20246,"journal":{"name":"Plant Diversity and Resources","volume":"174 1","pages":"614"},"PeriodicalIF":0.0,"publicationDate":"2012-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"79650329","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Preservation and utilization of plant diversity is the cornerstone of human welfare and civiliza鄄 tion, and an essential strategy to address global climate change. Although the plant conservation activities have been achieved significantly in the past decades world鄄wide, there are gaps existent that may minimize the effort to fulfill the Global Strategy for Plant Conservation (2011-2020), e. g., the complexity of species origin and evolution, the lack of sufficient species information, and the shrunken taxonomy expertise. We account for the plant conservation achievement in China and point part of constrains for the success of China爷s strategy for plant conservation, which could be removed with the conceiving and implementation of the next鄄generation Flora, or iFlora project. DNA bar鄄 coding is an important element of iFlora. However, little is known about phylogenetic distribution and evolutionary patterns of miR156-targeted SPLs. In this study, 183 SPLs from nine genome-sequenced species representing algae, bryophytes, lycophyte, monocots, and eudicots were computationally analyzed. Our results showed that miR156 responsive elements (MREs) on SPLs were present in land plants but absent from unicellular green algae. Phylogenetic analysis revealed that miR156-targeted SPLs only distributed in group II not group I of land plants, suggesting they originated from a common ancestor. In addition, group II were further divided into seven subgroups (IIa-IIg) and miR156-targeted SPLs distributed in some specific members of SPLs from six subgroups except subgroup IId. Such distribution pattern was well elucidated by gene structure evolution of miR156-targeted SPLs based on the correlation of phylogenetic classification and gene structure. They could suffer from the exon loss events combined with MREs loss during evolution. Moreover, gene duplication contributed to the abundance of miR156-targeted SPLs, which had significantly increased after angiosperms and lower plants split. With Arabidopsis as the model species, we found segmental and tandem gene duplications predominated during miR156-targeted SPLs expansion.
