From 84 isolated lactobacilli of indigenous poultry origin, 15 isolates were pre-selected for in vitro characterization on the basis of their activity (6.33±0.57-20.33±1.15 mm) against S. Enteritidis by well diffusion assay. All pre-selected isolates had variable tolerance to acidic pH (2, 3 and 4). All isolates also showed growth in MRS broth supplemented with 0.3, 1 and 1.8% bile salts. Isolates had varying degree of auto-aggregation (27.05±0.72 65.87±3.12%) and co-aggregation with S. Enteritidis (6.33±0.11 55.70±1.32%) within 2 hours. Safety profile of lactobacilli indicated that IKP23, IKP 111 and IKP 333 had no acquired antibiotic resistance. IKP 23, IKP 111 and IKP 333 were selected as potential probiotics on the basis of probiotic prerequisites and identified as L. fermentum, L. fermentum and L. salivarius, respectively by sequencing their partial 16S rRNA gene or 16S-23S intergenic spacer region. IKP 23, IKP 111 and IKP 333 inhibited S. Enteritidis (81, 99.3 and 93%, respectively) in co-culture experiments. This study insinuate that IKP 23, IKP 111 and IKP 333 have favourable probiotic potential and may be used for in vivo studies for the development of probiotics against S. Enteritidis. ©2019 PVJ. Dogs entering the trial were randomly assigned to receive only regular supportive therapy (control group) or combined hyperbaric oxygen therapy with symptomatic and supportive therapy (HBOT group). The regular supportive measurements consisted of intravenous fluid, analgesia, antibiotics, antiemetics, gastric acid suppression, nutritional management, and so on. If the patient had diabetes mellitus, hyperglycemia was controlled by insulin. The dogs of the HBOT group were conducted in hyperbaric animal chamber (PAHI-A, Pan-America Hyperbarics Inc., R.O.C.) and HBO therapy involved 90 min of 100% oxygen treatment at a pressure of 2.5 ATA, once to twice daily for a total of 3 to 5 sessions. There were significant findings regarding to specific canine pancreatic lipase immunoreactivity (Spec cPL) and C-reactive protein (CRP) in the survivors, but no significant differences were found in the non-survivors. The down trend of sequential CRP concentration was noticed in HBOT group. The level of creatinine may be as an important criterion when the patients with AP were first visit clinic. The prognostic factors such as CRP, Spec cPL had a good response to routine combined with HBOT in survivors group. Likewise, the level of Spec cPL was decreased significantly after treatment no matter routine or routine combined with HBOT in survivors group. HBOT combined with symptomatic and routine treatment might improve systemic inflammation probably in canine acute pancreatitis. In survivors group the creatinine levels was significant lower than that in non-survivors group. Therefore, the levels of creatinine can be as prognostic factor in dogs with acute pancreatitis. ©2019 PVJ. Most of the biological effects produced by polyphenols are largely attributed to their anti-inflammatory and antioxidant potential. The current research work describes the anti-inflammatory and antioxidant potential of newly synthesized phenolic derivatives (T2, T5, T6, T7 & T8) through in silico, in vitro and in vivo approach. The docking studies were performed against NADPH oxidase (PDB ID 3A1F-) and cyclooxygenase (COX-2 with PDB ID 5KIR Homosapien) using in silico techniques. Amongst phenolic derivatives (T2, T5, T6, T7 &T8) T2 and T6 exhibited maximum binding affinity with target protein NADPH oxidase (6.2 and -6.3 Kcal/mol respectively). However, in case of COX-2 as target protein, all the five newly synthesized phenolic derivatives showed encouraging results. In particular, T2 and T8 with binding affinity values -7.9 Kcal/mol and -8.0 Kcal/mol respectively which are promising than Ibuprofen with binding affinity -6.6 Kcal/mol. Then in vitro antioxidant potential of phenolic derivatives was analyzed through total reducing power, total antioxidant capacity and DPPH (1, 1-diphenyl2picryl-hydrazyl) assay. T2 and T6 showed antioxidant potential with IC50 values of 25.03μg/ml and 37.21 μg/ml respectively as compared to 15.14 μg/ml with respect to ascorbic acid. In vivo anti-inflammatory potential was determined through carrageenan induced paw edema which resulted in T2 with promising percentage edema inhibition (68.64±3.34) as compare to standard piroxicam (83.84±3.87). Amongst all phenolic derivatives T2 has promising results in in silico, in vitro and in vivo studies. ©2019 PVJ. However, hepatotoxicity and nephrotoxicity are the most features of acetaminophen overdose. Ginger is a medicinal plant that has anti-tumorigenic, anti-apoptotic, anti-oxidant activities. Recently, ginger nanoparticles have been investigated in liver protection against alcohol-induced liver damage and in treatment of bowel colitis. Our experiment was aimed to judge the protective activity of ginger nanoparticles relative to ginger extract at dose of 120 mg/kg against acetaminophen toxicity at dose of 375 mg/kg (1/10 LD50) daily for three months in male rats from the histological and biochemical aspects. Serum Alanine aminotransferase and aspartate aminotransferase were determined. In addition, urea and uric acid levels were evaluated in serum. Moreover, oxidative stress was evaluated by determining malondialdehyde content and catalase enzyme activity. Meanwhile, histopathological changes in liver and kidney tissues were observed. The present study indicates that liver and kidney biochemical markers, oxidative stress and histopathological structure are improved in rat pretreated with ginger extract. However, rats treated with GNPs were more protective relative to ginger extract pre-treated rat. ©2019 PVJ. This study was aimed to identify Salmonella serotypes and to detect the prominent virulence genes and antimicrobial resistance of the obtained serotypes from samples collected from dead chicken embryos, dead duck embryos and commercial broilers. The pathogenicity of some serotypes as (Salmonella Pullorum, Salmonella Enteritidis, and Salmonella Typhimurium) was investigated in one-day-old commercial layer chicks. A total of 180 cases were collected, after isolation 18 Salmonella isolates were reported to be as follows 7/100(7%), 5/40(12.5%) and 6/40(15%) in broilers, dead chickens, and dead duck embryos, respectively. The most prevalent serotypes were S. Enteritidis, S. Typhimurium, S. Pullorum, S. Vejle, S. Amsterdam, S. Infantis, S. Petersburg, and S. Atakpame. The distribution patterns of virulence genes iroN, cdtB, spaN, invA, and orgA, expressed as follows; 17/18(94.4%), 15/18(83.3%), 14/18(77.7%), 13/18(72.2%), and 12/18(66.7%) of serotypes, respectively. Added to that, the sipV, IpfC, sopB, prgH, and sitC virulence genes were amplified in 7/18(38.8%), 7/18(38.8%), 7/18(38.8%), 5/18(27.7%) and 3/18(16.6%) of isolates respectively. Most of isolates 16/18 (88.9%) expressed multiple antibiotic resistances (MAR) indices ranged from ≥0.3 ≤1. Based on the pathogenicity testing, S. Pullorum was the most pathogenic due to the clear signs and a mortality rate of 35%. Hence, the genotypic characterization and continuous monitoring of antimicrobial resistance of Salmonella from poultry are of public health concern to implement effective control strategies against this notorious pathogen. ©2018 PVJ. A total of 78 fresh semen ejaculate were collected from breeding buffalo bulls, diluted with required concentrations and relationship between sperm number and absorbance was determined at 350, 450, 546 and 650 nm wavelengths by using spectrophotometer. The spectrometric results showed that λmax was obtained at 350 nm. The derived equation between sperm concentration counted by haemocytometer and absorbance at 350 nm was Y = 0.1135x + 0.0002 (R = 0.9924), at 450 nm, Y = 0.1011x-0.0436 (R = 0.9756), at 546nm, Y = 0.0825x-0.0101 (R = 0.9603) and at 650 nm, Y = 0.0774x-0.0235 (R = 0.9602). These results were later compared with relative sperm numbers at different dilutions. The results showed that 350nm wavelength appeared suitable for estimation of sperm concentration compared to others. Additionally, it was found that both photometer(P<0.01) and haemocytometer (P<0.01) significantly counted more sperm cells than visual assessment as used regularly on different SPUs. Moreover, haemocytometer counted significantly (834×10Vs 678×10, P=0.005) more sperm cell than Photometer. Finally, regression analysis between haemocytometer and Photometer showed significant slope of regression and regression coefficient was 0.771 which warns that photometer must be calibrated before proper employment for routine measurement of sperm concentration. ©2019 PVJ. Host-restricted Streptococcus equi subspecies equi is the etiology. In April 2019, strangles was diagnosed in a donkey in the quarantine area of an intensive donkey farm. The infection was confirmed by clinical signs, bacterial isolation, histochemistry, PCR and sequencing. Based on the M-protein typing method, analysis of SeM sequencing data indicated that the isolated S. equi strain CN190402 belongs to a novel genotype SeM-138. The present study increases knowledge of the molecular epidemiology of S. equi and will lead to a greater understanding of the diversity of this pathogen. Meanwhile, it emphasized the necessity of quarantine areas and the importance of biosecurity for intensive donkey farms to prevent future outbreaks. ©2019 PVJ. E) and Selenium (Se) supplementation on physiological and reproductive performance of bulls during humid hot summer in Pakistan. Eighteen Holstein Friesian bulls balanced by age and weight were divided into three treatment groups: 1) CTL, basal diet without Se and Vit. E supplementation; 2) SeS, basal diet supplemented with 3 g Selemax® (Biorigin, USA); 3) ESeS, basal diet with 3 g Selemax® and Vit. E @ 6000 IU/day). Basal diet consisted of 50% green fodder, 25% wheat straw and 25% concentrate on dry matter basis. Diets were offered for 13 weeks from July 2016 to October 2016 having temperature-humidity index ranging from 83 to 87. Data obtained were subjected to repeated measures ANOVA. Supplementation of Vit. E and Se did not affect dry matter intake, water intake, respiration rate, rectal temperature, and pulse rate. The ESeS treatment had higher sperm motility% compared to CTL and SeS (P<0.05). The amplitude of lateral head displacement (ALH) was higher in SeS and ESeS compared to CTL (P<0.05). Straightness was higher in SeS group compared to CTL and ESeS. However, Vit. E and Se had no effect on other semen traits including semen volume, concentration, progressive motility, DNA integrity, plasma membrane integrity, and live to dead ratio. Blood testosterone concentration was significantly higher in SeS and ESeS groups compared to CTL. Blood glucose and blood urea nitrogen were similar among the treatments. Therefore; the present study aimed to characterize the circulating FMD viruses during this period. Clinical data and vaccination histories were collected from nine affected farms and 27 individual cases owned by small farmers in 10 Egyptian governorates. A total of 59 vesicular epithelia were gathered and tested by real-time (rt) RT-PCR to detect and quantify the virus. Furthermore, the positive samples with sufficient genomic loads were analyzed by the less sensitive conventional RT-PCR to amplify the full length of the VP1 region; amplicons were sequenced and phylogenetic trees were constructed. Of 59 samples analyzed, 49 (83.1%) were positive by rt RT-PCR, but only 22/39 produced amplicons by conventional RT-PCR. About 19 amplicons were suitable for sequencing and showed serotypes O, A, and SAT2 in 15, 1 and 3 samples, respectively. Phylogenetic analyses clustered the characterized strains in serotypes O, topotypes EA-3; serotype A, African topotype of genotype IV; and serotype SAT2, topotype VII, respectively. Interestingly, two lineages of serotype O topotype EA-3 strains were distinguished and referred to as Ism-16 and Alx-17. Ism-16 lineage clustered with the previously characterized viruses in Egypt in 2016, but the Alx-17 lineage clustered in a separate clade and is hereby reported for the first time. Further studies are required to evaluate the cross-protection between the vaccine and heterologous circulating strains in Egypt, especially for serotype O. ©2019 PVJ.
