DeepLabCut (DLC), a toolbox for markerless pose estimation, offers the ability to track user-defined features in 3-dimensions with human level accuracy. We demonstrate a hybrid method utilizing DLC and optogenetic motor mapping to localize the movements of multiple modalities to the mouse cortex. We outline a pipeline to map and characterize multiple motor representations in anesthetized and awake mice. In this paper, laser pulse shaping was designed and demonstrated for TD-DCS at 1064 nm. A quantum superconducting nanowire single-photon detector (SNSPD) with high photon detection efficiency (PDE) and low jitter collects the back-scattered light from the brain. Here, we present OCT guided ultrafast laser based non-viral targeted delivery PEDF-encoding genes to retina for neuroprotection. The ultrafast laser gene delivery showed layer-specific reliable expression of PEDF gene in retina without any detectable damage. Monitoring of IOP and electroretinogram after ultrafast laser transfection showed no adverse changes. The ultrafast laser transfection of large PEDF genes in retina exhibited significant therapeutic benefit in an injury model. By using this framework, we estimated the distributions of the mean RBC velocities in arterioles, venules, and capillaries, and as a function of the branching order from over 1,000 microvascular segments. We developed a dual fluorophore imaging system for simultaneous, high-speed mapping of neural, metabolic, and hemodynamic activity. Proof-of-concept measurements of spontaneous and stimulus-evoked dynamics are presented in awake and anesthetized mice.
