This simplification often does not provide sufficient or correct information on the relationship between the composition of the products and the process variables studied. The obtention of profiles by gradient automated multiple development (AMD) is an interesting alternative to obtain extended SARA separations with increasing level of complexity for heavy petroleum products that adequately represent the entirety of the sample. An optimized 20-step gradient based on tetrahydrofuran (THF)‒dichloromethane‒ n -heptane over a total migration distance of 83 mm is proposed here to characterize all types of heavy oil products. The combination of ultraviolet (UV)–densitometry (at 201, 228, and 273 nm), fluorescence detection by intensity changes using berberine cation, together with on-plate recording of UV spectra of separated peaks and the comparison with those of standards, allow for a deep characterization of chemical families. In the apolar zone of the chromatogram saturates, naphthenes, naphteno-aromatics, alkyl-aromatics, and aromatics with different degrees of condensation are detected. The use of THF, both in the plate precleaning stage and in the first stages of the gradient, is responsible for the satisfactory resolution of the resins in the polar zone. AMD chromatographic zones can be related to peaks from hydrocarbon groups in classical SARA. A thin-layer chromatographic method combined with simple and inexpensive digital imaging is proposed for the determination and quantification of ibuprofen. The recommendations of the standard pharmacopoeia were followed with the addition of a minimal spotting method as a local calibration curve ( n = 3) and comparison with a spotted sample. Images were acquired using a smartphone and then converted to greyscale using Fiji, an open-source image analysis software. The peak area and density of each spot was then calculated using a densitometric principle. To analyse the present image data, several images were taken with different filters. Using a calibration curve ranging in the range of 3 to 5 mg/mL, linearity could be determined across the subjected images ( R 2 ranging from 0.984 to 0.996), with a mean accuracy of 99.9 ± 2.20% and RSD of 2.20%. The method studied, using smartphone imaging and open-source software (Fiji), offered a simple and inexpensive application for the quantification of ibuprofen in pharmaceuticals, applicable to preliminary studies and curricula. The analyzed mixture is composed of three active ingredients: propyphenazone (PRO), caffeine (CAF), and ergotamine tartrate (ERG), along with two official impurities which are PRO impurity: phenazone (PHN) and CAF impurity: theophylline (THEO). The suggested method was used for the quantitation of the three coformulated active ingredients in their marketed tablet and in human plasma. The studied compounds were separated on TLC silica gel 60F 254 plates using a mobile phase consisting of methanol–ethyl acetate–glacial acetic acid (1:9:0.1, V/V ) with diprophylline (DPP) as internal standard. Densitometric scanning was carried out at 210.0 nm. Method validation was assessed according to the International Council for Harmonisation (ICH) guidelines. The greenness profile for the proposed method was evaluated using the National Environmental Method Index (NEMI), analytical eco-scale, and Green Analytical Procedure Index (GAPI) tools. The proposed method offers the advantages of being simple, rapid, economic, and ecofriendly.
