Blood Objectives: The purpose of this study is to evaluate cobas parathyroid hormone (PTH) measurement effect of plasma samples obtained from underfilled tubes with ethylenediaminotetraacetic acid (EDTA). Methods: Two blood collection tubes with K3-EDTA from 67 patients at the same time were taken. One of them was for routine PTH measurement, which was filled to its capacity, while another tube was underfilled. All EDTA tubes were immediately centrifuged at 4°C. Plasma PTH concentrations were measured by electrochemiluminescence immunoassay method on the cobas e 601 (Roche Diagnostics GmbH, Mannheim, Germany) analyzer. Results: The underfilled sample tubes were grouped according to their being up to 25% (n=9), 25–50% (n=35), and 50–100% (n=23) of the appropriate amount. In all groups of underfilled tubes, the PTH values were found to decrease concerning those fulfilled (p<0.001). The agreement among underfilled and fulfilled for measuring PTH demonstrated a bias of −3.2 pg/mL (−5.9%) for the underfilled tubes. Conclusion: Insufficiently filled blood in tubes with EDTA has a significant effect on PTH levels. However, further studies are needed to show whether this effect is clinically significant.","PeriodicalId":176166,"journal":{"name":"International Journal of Medical Biochemistry","volume":"1 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"1900-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"121227528","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

However, studies examining the relationship between plasma angiostatin and the characteristics and laboratory findings of systolic HF patients with CKD are limited. The hypothesis of this study was that an increase in angiostatin in the plasma might contribute to the impairment of angiogenesis in HF patients with CKD, and the relationship with the laboratory findings and medications of the patients were investigated. Materials and Methods Study sample and study protocol This comparative, cross-sectional study was conducted with a total of 69 individuals: HF patients with CKD (n=29) and participants without disease (n=40) who presented at the nephrology or internal medicine outpatient facilities of Gulhane Training and Research Hospital, Ankara, Turkey, between 2011 and 2013. All of the procedures applied were performed in accordance with the ethical standards of the committees on human trials (institutional and national) and the Declaration of Helsinki. This study was approved by Gulhane Education and Research Hospital local ethics council with protocol number 1491-181-12/1539-491 on February 28, 2012. The EF was less than 40% and between 40% and 50%, respectively, for the patients with HFrEF and HFmrEF. All of the patients and the control group participants were older than 18 years of age. In all, 17 of the participants in the control group were male and 23 were female. In the patient group, 17 were male, and 12 were female. The diagnosis of systolic HF was based on the symptoms, a physical examination (European Society of Cardiology, 2016), echocardiographic results, and N-terminal pro-brain natriuretic peptide (NT-proBNP) level. Patients with rheumatic disease or clinical signs of infection, atrial fibrillation, chronic obstructive pulmonary disease, high C-reactive protein (CRP) value (>5 mg/L) and/or known malignancy were excluded from the study. The Chronic Kidney Disease Epidemiology Collaboration (CKD-EPI) equation was used to determine the glomerular filtration rate (GFR) [11]. There were 15 patients with diabetes mellitus in the patient group and all were on insulin treatment. The serum angiostatin level of the HF patients with CKD and the control group were compared. The angiostatin level and the clinical findings of demographic features, laboratory results, comorbidities, and medications in the HF patient were compared with the CKD group to assess possible relationships. After an overnight fast, baseline blood samples were drawn from an antecubital vein in each participant into a BD Vacutainer venous blood collection tube containing clot activator and gel for serum isolation (Becton, Dickinson and Company, Franklin Lakes, NJ, USA). Centrifugation at 2000 g for 10 minutes was performed to separate the serum. A portion was used for analysis of the biochemical para","PeriodicalId":176166,"journal":{"name":"International Journal of Medical Biochemistry","volume":"8 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"1900-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"123370641","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The use of blood collection tubes with small-volume may reduce the need for blood transfusion and the associated risks due to frequent phlebotomy. Therefore, we evaluated the reliability and accuracy of the complete blood count (CBC) using new generation small-volume blood collection tubes (SV-BCT) instead of large-volume blood collection tubes (LV-BCT). Methods: Venous blood samples were taken from 40 adult in-patients and collected to SV-BCT/LV-BCT pairs of three different brand (Microtainer®MAP-0.5 ml/Vacutainer®-2.0 ml; Becton, Dickinson and Company, USA) ([Microvette®-0.5 mL/S-Monovette®-2.6 ml; Sarstedt Ag and Co. KG, Germany]) ([MiniCollect®Complete-0.5 ml/Vacuette®-2.0 ml; Greiner Bio-One GmbH, Austria]). All tubes contained K2EDTA except Microvette®. Sixteen parameters of CBC were analyzed using a DxH 800 (Beckman Coulter Inc., USA). CBC results in tube pairs were compared in terms of statistical and clinical (bias%) significance. Results: There were statistically significant differences between the results of SV-BCT and LV-BCT pairs of the same brands for some parameters. However, bias% between tubes for 16 parameters was within the desirable limits, the differences were not clinically significant. Conclusion: Health personnel and phlebotomists can safely prefer SV-BCT which is a new generation and technically useful for CBC, especially in patients requiring frequent phlebotomy. Thus, the volume of blood sampling may be reduced to prevent iatrogenic blood loss.","PeriodicalId":176166,"journal":{"name":"International Journal of Medical Biochemistry","volume":"24 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"1900-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"126339557","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
